背景技术
沙丁胺醇是一种支气管扩张药,被认为是β-肾上腺素拮抗剂,其能刺激β-肾上腺素受体,导致支气管平滑肌舒张,并抑制肥大细胞过敏。沙丁胺醇显示出可用于缓解会引起哮喘的支气管痉挛和可逆的阻塞性气道病。
沙丁胺醇又称为(±)∝-[(四-丁基氨基)甲基]-4-羟基-m-二甲苯-∝,∝’-二醇硫酸(2∶1)盐,化学式为(C13H21NO3)2.H2SO4,分子量576.7,结构式如下:
目前市售的沙丁胺醇如Volmax片,其有两种浓度,4和8mg,由Glaxo Wellcome Ltd.UK England,Muro Pharmaceutical Inc.,Tewksbury,Mass achusetts制备。口服给药Volmax片起效达12小时的持续作用,6小时内达到最大血浆药物浓度,血浆半衰期为大约9小时。Volmax片由药片一面上的激光钻孔洞(OROSTM技术)从聚合包衣片中释放药物。这种片包括控制速率的半透膜,和沙丁胺醇片芯及渗透剂。片芯组分周围的渗透性成分吸水进入片中,沙丁胺醇和渗透剂溶解,沙丁胺醇从激光钻孔洞中释放。

发明内容
本发明涉及缓释药物制剂。
本发明也涉及制备缓释药物制剂的方法。
本发明提供一种稳定的以硫酸沙丁胺醇缓释片形式存在的沙丁胺醇缓释片制剂,该缓释片制剂与Volmax片具有生物等效性,Volmax片包括4mg和8mg片,由Glaxo Wellcome Muro Pharmaceutical Inc.制备。
本发明提供一种能达到与使用渗透性装置制剂等效的扩散型控释制剂。
本发明也涉及控释药物制剂,其具有能在扩散型(diffusion)控释片中获得的想要的溶出速率。
本发明提供一种缓释片,含有沙丁胺醇和缓释剂的片芯;和包裹片芯,提供沙丁胺醇持续释放的缓释包衣。
缓释剂可以包括疏水性聚合物,疏水性聚合物可包括乙基纤维素。
缓释包衣可含有疏水性聚合物和亲水性聚合物,疏水性聚合物可包括乙基纤维素,且亲水性聚合物可包括甲基纤维素。
缓释包衣可含有乙基纤维素和甲基纤维素,缓释剂可包括乙基纤维素。
缓释包衣中的疏水性聚合物和亲水性聚合物以所述疏水性聚合物与所述亲水性聚合物的重量比为55-65∶45-35(重量百分比)的量存在;优选以疏水性聚合物与亲水性聚合物的重量比为53∶47的量存在。
缓释包衣的疏水性聚合物和亲水性聚合物可以分别包括乙基纤维素和甲基纤维素。
缓释包衣可具有大约5到25mg的重量,更优选8到13mg。
乙醇是制备片芯和缓释包衣的优选使用溶剂。

其生物等效性是采取对空腹的健康成年男性以随机、单剂量、双交叉生物等效性研究来测定,以36个样本和血浆沙丁胺醇水平为准,当其为含有4mg沙丁胺醇的制剂时,AUC 0-t为53074pg.h/ml的80-125%,AUC inf为55606pg.h/ml的80-125%,Cmax为4383pg.h/ml的80-125%。
生物等效性可以采取对进食和空腹的健康成年男性以随机、单剂量、三组交叉生物等效性研究来测定,以16个样本为基准,当其为含有8mg沙丁胺醇的制剂时,其中进食的成年男性血浆沙丁胺醇水平为,AUC 0-t为106139pg.h/ml的80-125%,AUC inf为109692pg.h/ml的80-125%,Cmax为7149pg.h/ml的80-125%。
生物等效性可以采取对进食和空腹的健康成年男性以随机、单剂量、双交叉生物等效性研究来测定,以37个样本和,血浆沙丁胺醇水平为基准,当其为含有8mg沙丁胺醇的制剂时,血浆沙丁胺醇水平为,其中AUC 0-T为1125573pg.h/ml的80-125%,Cmax为14522pg.h/ml的80-125%。

不希望受到理论约束,扩散包括原子由于它们的无规则运动的动力能量所导致的混入过程。相反地,渗透作用是被动的选择性扩散过程,比如,由溶剂中原子内部能量被迫产生。
当在本发明说明书和权利要求书中提到溶出度或溶出特性时,溶出是依照实施例中所陈述的溶出步骤,或与其等同的溶出技术进行的。
总体上说,本发明涉及缓释药物制剂。这些制剂特别适用于硫酸沙丁胺醇的缓释。具体的,本发明涉及一种包括含有硫酸沙丁胺醇和缓释剂的片芯;和包括亲水性剂和疏水性剂的缓释包衣,且可有一个或多个包衣。片芯和缓释包衣分别互相制剂,以获得药学可接受的体外溶出特性,尤其是与渗透装置所获得的沙丁胺醇生物等效的溶出特性或特征。
根据本发明的片剂获得想要的体外溶出特性,和FDA证实的沙丁胺醇片剂即Volmax片的生物等效,在片剂内,与被渗透控制如Volmax片相比,本发明的片是通过扩散控制的。
根据本发明,标注量的沙丁胺醇在每个特定的时间间隔内释放的优选百分比是通过实施例中阐述的溶出度测定试验测定,对8mg沙丁胺醇,是:
第2小时 不超过30%
第6小时 50-75%
第10小时 不少于75%
而且,根据本发明,标注量的沙丁胺醇在每个特定的时间间隔内释放的优选百分比是通过实施例中阐述的溶出度测试得出的,对4mg沙丁胺醇,是:
第2小时 不超过20%
第6小时 45-70%
第10小时 不少于75%
片剂的含水量优选不超过0.5重量%。而且,在制剂中使用的乙醇的残留含量优选少于0.5重量%。
根据本发明,片芯包括活性药物成分和缓释剂。活性成分优选包括硫酸沙丁胺醇。
根据本发明的片芯,优选用包含活性成分和缓释剂的微粒通过制粒过程制备的。这些颗粒优选具有平均颗粒直径大约125μ到1700μ,更优选大约180μ到1000μ,最优选大约250μ到600μ。
片芯中的活性成分的含量优选大约4或8mg活性成分。例如,对硫酸沙丁胺醇而言,片芯的成分可优选大约4.8到9.6mg。
片芯优选构成可提供全部剂量4或8mg的沙丁胺醇。片芯优选用4.8mg沙丁胺醇制备,以提供全部剂量4mg的沙丁胺醇,片芯优选用9.6mg沙丁胺醇制备,以提供全部剂量8mg的沙丁胺醇。因此,本发明的片剂既可采用包含的一定量活性成分如沙丁胺醇制备,也可采用其药学上可接受的盐优选硫酸沙丁胺醇制备。
制剂中使用的硫酸沙丁胺醇优选具有粒径分布,采用在空气法中的颗粒根据Mavern Mastersizer(Mavern 2600c with PS 64 dry powderfeeder,镜头100mm,obscuration 0.1到0.4)测定。优选的,被添加到片芯中的硫酸沙丁胺醇颗粒至少50%不超过(NMT)25μm,至少90%不超过55μm。不希望受到理论约束,认为这些参数内的颗粒能同时提供良好的溶出度和片芯硬度。具体的沙丁胺醇分批测定具有下述表1中表示的粒径分布:

缓释剂可以包括一种或多种成分,其与缓释包衣联合控制活性药物成分尤其是沙丁胺醇从片芯中的释放,以获得想要的溶出特性,特别的,与FDA证实的硫酸沙丁胺醇制剂具有生物等效性。该缓释剂可以包括多种水不溶性材料,这样的水不溶性材料包括如疏水性聚合物,包括,但不限于,一种或多种乙基纤维素、丁基纤维素、醋酸纤维素、丙酸纤维素、聚乙烯乙酸酯、聚乙烯丁酸酯、乙基丙烯酸酯-甲基异丁烯酸酯-乙基铵三甲基氯异丙烯酸酯共聚物和N-乙烯基-2-吡咯烷基纤维素酯、纤维素酯、聚乙烯酯、具有四元铵-烷基的丙烯酸类聚合物,以及PlasdoneK-90、聚维酮、N-乙烯基-2-吡咯烷酮的均聚物。优选的,该缓释剂包括一个或多个乙基纤维素、丁基纤维素、醋酸纤维素、丙酸纤维素、聚乙烯乙酸酯、聚乙烯丁酸酯、乙基丙烯酸酯-甲基异丁烯酸酯-乙基铵三甲基氯异丙烯酸酯共聚物和N-乙烯基-2-吡咯烷酮。更优选的,该缓释制剂包括乙基纤维素,比如Ethocel N-10,其购自Dow Chemical,Midland,Michigan公司。
片芯可以采用任意方法制备,比如采用任何常规的制备技术。因此,片芯可通过混合活性成分和至少一种可限制活性成分自身及其它成分释放的赋形剂。例如,片芯可包括常规的多种赋形剂,比如那些常规包含在片剂片芯中的组分,包括稀释剂、粘合剂、制粒溶剂、制粒剂、抗聚集剂(antiaggregating agent)、缓冲剂、润滑剂。
例如,稀释剂包括,但不限于,一种或多种糖类如蔗糖、乳糖、甘露糖;淀粉;微晶纤维素;山梨醇,麦芽糖糊精,乳清,磷酸钙;硫酸钙;硫酸钠或类似的无水硫酸钠;乳酸钙;无水乳糖;及乳糖一水化物。优选的,稀释剂包括硫酸钙和/或乳糖一水化物。优选的,无水乳糖不包括在片芯中,因为它有从溶剂如乙醇中吸水的能力,从而促进不想要的增重。
粘合剂的例子包括,但不限于,一种或多种聚乙烯醇、聚丙烯酸、聚异丙烯酸、聚乙烯基吡咯烷酮、蔗糖、山梨醇、羟乙基纤维素、羟丙基甲基纤维素、羟丙基纤维素、聚乙二醇、阿拉伯胶、明胶、琼脂、淀粉等。
润滑剂和抗聚合剂包括,但不限于,一种或多种滑石、硬脂酸镁、硬脂酸钙、胶态二氧化硅、硬脂酸、石蜡、氢化植物油、聚乙二醇、苯甲酸钠、月桂酰基硫酸钠、月桂酰基硫酸镁和dl-亮氨酸。优选的,润滑剂包括硬脂酸和/或硬脂酸镁。
溶剂包括,但不限于,一种或多种丙酮、乙醇、异丙醇(IPA)和/或亚甲基氯。优选的,用于片芯的溶剂包括乙醇。优选避免应用氯化溶剂如亚甲基氯。因此,片剂最好不包括氯化溶剂,比如亚甲基氯。可调整溶剂的量到能实现颗粒制剂的足够量而并不需要会使混合物转化成大团块而不是自然颗粒的高量。而且,优选控制溶剂加入时间以实现充分的制粒。制粒溶液的温度在填料时优选是不超过26℃,以避免结块。
组分可以在一步或多个步骤中混合和/或采取任意合适的技术制粒,,如低剪切制粒机、流化床、高剪切制粒机。
随着制粒技术的发展,采用或不采用硫酸钙分别制剂以研究它对产品溶出特性的冲击作用是很重要的。基于硫酸沙丁胺醇可从不含硫酸钙的片剂中快速释放药物,优选将硫酸钙添加到片芯中以控制药物从片剂中释放。
接着是片芯颗粒的生成,颗粒在充分条件下干燥,以获得优选含有不超过0.5重量%乙醇和优选不超过0.5重量%水的颗粒。因此,对商业批量而言,优选的批量干燥在大约50℃温度下优选干燥至少16小时,以确保溶剂如乙醇和/或水挥发。
片芯压缩成片以达到硬度优选在大约2到6kP之间,更优选值为大约4kP。
缓释包衣被投料到片剂的片芯的表面,优选直接分层扑到片芯上。缓释包衣与片芯缓释剂制剂以获得想要的溶出特性,与市售硫酸沙丁胺醇片剂如Volmax片具有生物等效性。
缓释包衣包括优选由疏水性和亲水性聚合物结合的膜。优选的,活性药物成分,优选硫酸沙丁胺醇在缓释包衣中不存在。在这点上,优选的,活性成分优选沙丁胺醇仅存在于片芯中。
构成薄膜的疏水性聚合物并不受具体限制,只要其具有成膜能力,且在水中不溶,而溶于与水易混合的有机溶剂中。疏水性聚合物的例子包括,但不限于,一种或多种乙基纤维素、丁基纤维素、醋酸纤维素、丙酸纤维素、聚乙烯乙酸酯、聚乙烯丁酸酯、乙基丙烯酸酯-甲基异丁烯酸酯-乙基铵三甲基氯异丁烯酸共聚物和N-乙烯-2-吡咯烷酮纤维素酯、纤维素酯、聚乙烯酯、具有四元铵-烷基的丙烯酸类聚合物、和PlasdoneK-90、吡咯烷酮、N-乙烯-2-吡咯烷酮的均聚物。优选的,可以包括,例如乙基纤维素、丁基纤维素、醋酸纤维素、异丁烯酸纤维素、聚乙烯乙酸酯、聚乙烯丁酸酯、乙基丙烯酸酯-甲基异丁烯酸酯-乙基铵三甲基氯异丁烯酸共聚物和N-乙烯-2-吡咯烷酮。最优选的,疏水性聚合物包括乙基纤维素、比如购自Dow Chemical MidlandMichigan公司的Ethocel N-10。
亲水性聚合物可以是水不溶性聚合物,包括但不限于,一种或多种甲基纤维素、可选择的具有硫酸基的多糖如pullulan、环糊精、烷甲基藻酸盐等;具有羟基或羧烷基的多糖,如羟丙基纤维素、羟丙基甲基纤维素、羧甲基纤维素钠等;甲基纤维素、聚乙烯吡咯烷酮、聚乙烯醇或聚乙二醇等也都可以被包括。优选的,亲水性聚合物包括甲基纤维素,如购自Dow Chemical Midland Michigan公司的Methocel E-15。
改变缓释包衣中的疏水性聚合物与亲水性聚合物的重量百分比可影响片剂中沙丁胺醇的溶出。因此,对每个具体的制剂而言,可改变疏水性聚合物与亲水性聚合物的重量百分比以获得制剂想要的溶出特性。优选的,疏水性聚合物与亲水性聚合物的重量百分比包括55-65∶45-35,优选的疏水性聚合物与亲水性聚合物的重量百分比为57∶43。
缓释包衣的溶剂可包括多种溶剂,包括,但不限于,丙酮、乙醇、异丙醇(IPA)、和/或亚甲基氯。优选的,缓释包衣的溶剂包括乙醇。优选尽量避免使用氯化溶剂如亚甲基氯。也可以使用多种溶剂的组合,比如,但不限于乙醇和异丙醇、及异丙醇与水,变性醇(denaturedalcohol)∶水和乙醇∶水的联合。
发现使用丙醇作为包衣溶剂比使用乙醇为包衣溶剂制备的片剂释药更慢。而且,当异丙醇被同时用于制粒和包衣时,发现比同时使用乙醇制粒和包衣制备的产品的释药更慢。因而,乙醇是制粒和包衣的优选溶剂。
为获得具有Volmax片生物等效性的产品,疏水性聚合物和亲水性聚合物互相混合制备的缓释包衣与包在片芯中的缓释剂共同作用可获得具有想要的溶出特性和/或生物等效性的缓释包衣。例如,采用这里公开的优选制剂,重量百分比为57份的Ethocel N-10与43份的Methocel E-15,优选的包衣增重为大约每片5-25mg。优选的,对采用R&D装置制备的4mg和8mg片,包衣增重为大约每片20mg,其具有与Volmax类似的释药特性。对4mg和8mg硫酸沙丁胺醇片而言,市售片分别增重8.7和12.4mg。缓释包衣的优选增重为大约5到25mg,更优选大约8到13mg。
缓释包衣的包衣可以采用多种技术制备,比如常规的包衣技术如穿孔盘(perforated pan),或使用流化床技术。这些方法容易被操作片剂制备领域的人员理解。在喷雾包衣中使用的喷枪的角度可以调整,比如,但不限于30°和75°之间,优选角度为65°。
不希望受理论的约束,亲水性聚合物提供了活性成分从片芯中扩散的多孔层。在这一点上,认为扩散是通过亲水性聚合物溶出而出现的孔进行的。
在本发明的制剂中,通过充分控制缓释包衣的重量、疏水性聚合物与亲水性聚合物的重量百分比和片芯中缓释剂的量,获得具有想要的体外溶出速率的片剂是可能的。例如,也不希望受理论的约束,当片芯中的药物活性成分是一种想要在给药后的短期内起效的药物时,最好制成薄膜,亲水性聚合物溶出后会出现许多大孔,然而当药物想要持续释放一段较长时间时,最好制成厚膜,亲水性聚合物溶出后会出现许多小孔。
对一种固定的缓释包衣的组合物而言,活性药物成分溶出速率的不同可以通过改变包衣的重量而实现。因此,较高量的缓释包衣同较低量的缓释包衣相比,可使活性成分溶出的平衡浓度保持较长时间。所以,缓释包衣的量在片剂制备过程中应当准确的监测,以确保获得想要的缓释包衣组合物的溶出速率。例如,缓释包衣的重量可通过测定在应用缓释包衣之前的片芯重量,然后确定片芯和缓释包衣的总重量而监测。这两个值之间的差将是缓释包衣引起的增重。例如,增重10mg将获得比增重9mg更慢的溶出速率。
片剂可包括选择性的包衣,比如可以提供cosmetic elegance和/或便于产品包装的包衣。例如,可选择的包衣可包括彩色包衣,但不限于Opadry Green。另一种可选择的包衣也包括,例如,清澈的溶液,比如,但不限于Opadry Clear。这些可选择的包衣可以联合应用,比如将CLEAR COAT应用到颜色包衣的外面。
根据本发明的药物活性成分优选包括硫酸沙丁胺醇,但也包括其它活性成分。然而,本发明特别适合采用硫酸沙丁胺醇作为活性药物成分。在这一点上,如上述提及的,本发明提供具有与那些符合渗透原理的药物片相似溶出速率和生物等效的硫酸沙丁胺醇缓释制剂。
本发明将通过下述实施例进一步阐述。这些实施例是非限定性的,不限制本发明的范围。
除非另有说明,实施例中的所有的百分比、部分等都是按重量计。
实施例
采用与本发明有关的下述步骤:
A.溶出步骤—确定本发明的溶出速度和溶出特性
根据本发明采用的溶出步骤包括下述步骤:
使用符合USP24、Method<724>的Van Kel Model VK-7000溶出装置。六个实施例中溶出部件都具有符合USP24 Method<711>标准的浆柄(paddle shafts)或类似部件,并采用装有UV监测器的HPLC系统。溶出步骤采用下述试剂和标准:

溶出介质的制备:
将9.0g氯化钠溶解在1000ml水中,加热到37℃±0.5℃,过滤、脱气、形成0.9%氯化钠溶液。
流动相的制备:
将600mg 1-辛烷硫酸钠盐和20.0g醋酸铵溶入920mg水中。再加入30ml 2-丙醇、10ml甲醇和40ml冰醋酸,适当混合。
真空抽滤,超声脱气5分钟。也可采用氦喷雾。
标准溶液的制备:
A.常备标准溶液:
精确称重大约24mg硫酸沙丁胺醇,并定量转入100ml的含有50ml溶解介质的容量瓶中。超声溶解标准液,再用溶解介质稀释到定量,混合。
B.制备标准溶液:
吸取4.0ml常备标准溶液移入容量瓶,对4mg片用200ml容量瓶,对8mg片用100ml容量瓶。
用溶解介质稀释到容量体积并混合。
将一部分溶液通过0.45μm孔径的SRI(Scientific Resources,Inc.Catalog:#4425-NC)或等值的nylon disk fiter,弃除最初的几ml液。用过滤的物质填入1ml HPLC玻璃瓶。
溶出度测定的色谱系统:

溶出度测试是采用900ml溶出介质(0.9%氯化钠溶液)和在划槽搅拌速度为50rpm下进行的,样品按照USP24 Method<711>处理。由于每次抽出10ml样品用于测定,将预加热到37℃±0.5℃的10ml溶出介质加入到溶出试管中以代替抽出用于测定的溶解液。
采用符合下述参数平均值的标准HPLC技术进行样品分析:
—平行测定沙丁胺醇进样液的峰面积相应值的相对标准偏差(RSD)应当不超过3.0%。
—沙丁胺醇的拖尾因子不超过2.0。
用于沙丁胺醇蜂值的每体积理论板数应当不少于2000。
微粒粒径和粉末纯度可采用符合USP-23NF18的筛分析机测定,例如采用ATM Sonic Sifter Model L3P。

用#18孔筛过筛包括乳糖一水合物、硫酸沙丁胺醇和硫酸钙的组分,并用#12孔筛过筛缓释剂(Ethocel N-10)。在不含I-BAR的双壳混合机(twin shell blender)中混合成分形成均一的混合物。将所有组分倒入AMF搅拌机中,设置为45±5RPM。
在温的(50℃±5℃)溶剂中(乙醇)制备缓释剂(Ethocel N-10,2kg)溶液。搅拌溶液至澄清,比如最少搅拌1小时。然后,冷却溶液至不超过26℃。开启搅拌器,泵入(pump into)Ethocel N-10溶液粒化成分,比如进给溶液3分钟。混合获得充足的微粒,比如用时大约6到9分钟。铺开干燥,比如置于50℃±5℃的不锈钢盘上,用时不超过16小时,直到在75℃时的平均干燥损失小于0.5%。
在具有调节给料器(screwfeeder)、有孔无锈钢盘的#0065的fitzmill中碾磨干燥颗粒,设置为中等速度、knives forwardand feeder at 3.5±0.5mg。
用#18孔筛过筛其余成分硬脂酸和硬脂酸镁,在合适的容器中收集。转移碾磨过的组分,用不含I-BAR的双壳混合机过筛组分,并混合直至均匀,优选大约5分钟。
设置表压为”转,深凹、plan tooling,并挤压混合物到标准值100mg±7mg(compress the blend at a nominal value of at 100mg±7mg)。在4kp(范围2-6kp)硬度下压缩。这些实施例中的片芯具有厚度为0.130”-0.155”,具有的脆碎度小于1%。
缓释包衣溶液的制备:
将Methocel E-15和Ethocel N-10溶于乙醇和纯净水的混合物中,混合直至澄清,比如最少5小时,制备得缓释(ER)包衣溶液。加入甘油三乙酸酯,搅拌优选15多分钟,至分布均匀。应用前贮存这种溶液最少10小时,比如15到16小时。

将片芯片转入48”带孔盘中,设置喷雾枪为65°角度,并调节雾化空气压为25PSI雾化空气压。调整用于缓释包衣溶液的泵以提供溶液为130ml/min/gun(范围:100-130ml/min/gun)。设置空气压为75℃或更小以使片芯片变暖。调节包衣盘(coating pan)为6RPM盘速(4-8RPM范围)。在入口空气温度为70℃±5℃下干燥缓释包衣溶液包衣片30分钟。测定包衣增重。
用膜包衣溶液包衣
采用上述包衣,只是液体速率为90±15ml/min/gun.。在70℃±5℃下干燥片10分钟。确定包衣增重。

实施例10
该实施例涉及健康受试者在进食和空腹条件下进行的4和8mg产品的生物等效性研究。
临床研究的目的是以Sidmak产品作为参照,比较相对稳定态的生物利用度。该研究方案是比较成年健康男性在进食和空腹条件下其对Sidmak和Muro(Volmax)4和8mg硫酸沙丁胺醇缓释片的随机双交叉稳定态研究。
根据本发明用于试验产品的临床供给品是根据实施例1、和参照品、Glaxo Wellcome公司按照Lot No:10460727制备的8mg片和按照Lot No.D008194制备的4mg片的Volmax。
试验过程中,在最初给药直到给药12小时后的合适的时间采集血样。总之,表中显示的编号受试者的数据是根据FDA指导的“口服给药产品-通常考虑的-的生物利用度和生物等效性研究(Bioavailability and Bioequivalence Studies for Orally administeredDrug Products-General Considerations.)”进行分析。

Claims (28)
1.缓释片,包括:
含有沙丁胺醇和缓释剂的片芯;和
包裹片芯,提供沙丁胺醇持续释放的缓释包衣。
2.根据权利要求1的缓释片,其中缓释剂包括疏水性聚合物。
3.根据权利要求2的缓释片,其中疏水性聚合物包括乙基纤维素。
4.根据权利要求1的缓释片,其中缓释包衣包括疏水性聚合物和亲水性聚合物。
5.根据权利要求4的缓释片,其中疏水性聚合物包括乙基纤维素,并且亲水性聚合物包括甲基纤维素。
6.根据权利要求1的缓释片,其中缓释包衣包括乙基纤维素和甲基纤维素,并且缓释剂包括乙基纤维素。
7.根据权利要求4的缓释片,其中所述缓释包衣中的疏水性聚合物和亲水性聚合物以所述疏水性聚合物与所述亲水性聚合物的重量比为55-65∶45-35的量存在。
8.根据权利要求7的缓释片,其中所述缓释包衣中的疏水性聚合物和亲水性聚合物以疏水性聚合物与亲水性聚合物的重量比为53∶47的量存在。
9.根据权利要求7的缓释片,其中所述缓释包衣中的疏水性聚合物和亲水性聚合物分别包括乙基纤维素和甲基纤维素。
10.根据权利要求8的缓释片,其中所述缓释包衣中的疏水性聚合物和亲水性聚合物分别包括乙基纤维素和甲基纤维素。
11.根据权利要求7的缓释片,其中缓释包衣的量为大约5到25mg。
12.根据权利要求11的缓释片,其中缓释包衣的量为大约8到13mg。
13.根据权利要求4的缓释片,其中将乙醇作为制备片芯和缓释包衣的溶剂。
14.根据权利要求1的缓释片,其中片芯包括无水硫酸盐。
15.根据权利要求1的缓释片,其中无水硫酸盐包括硫酸钙。
16.根据权利要求1的缓释片,其中片芯包括乳糖一水合物。
17.根据权利要求1的缓释片,其包括当其为含有8mg沙丁胺醇的制剂时,具有如下沙丁胺醇溶出特性:
第2小时 不超过30%
第6小时 50-75%
第10小时 不少于75%。
18.根据权利要求1的缓释片,其包括当其为含有4mg沙丁胺醇的制剂时,具有如下沙丁胺醇溶出特性:
第2小时 不超过20%
第6小时 45-70%
第10小时 不少于75%。
19.根据权利要求1的缓释片,其与硫酸沙丁胺醇渗透性装置制剂具有生物等效性。
20.用于持续释放硫酸沙丁胺醇的扩散型控释片,其包括在其扩散型控释制剂结构中含有硫酸沙丁胺醇,且当其为含有8mg沙丁胺醇的缓释片制剂时,能提供具有如下的沙丁胺醇溶出特性:
第2小时 不超过30%
第6小时 50-75%
第10小时 不少于75%。
21.用于持续释放硫酸沙丁胺醇的扩散型控释片,其包括在扩散型控释制剂结构中含有硫酸沙丁胺醇,且当其为含有4mg沙丁胺醇的缓释片制剂时,能提供具有如下的沙丁胺醇溶出特性:
第2小时 不超过20%
第6小时 45-70%
第10小时 不少于75%。
22.用于持续释放沙丁胺醇的扩散型控释制剂,其包括在扩散型控释制剂结构中含有硫酸沙丁胺醇,且与硫酸沙丁胺醇渗透性装置制剂具有生物等效性。
23.根据权利要求22的扩散型控释制剂,当其为含有4mg沙丁胺醇的制剂时,其生物等效性是采取对空腹的健康成年男性以随机、单剂量、双交叉生物等效性研究来测定,以36个样本和血浆沙丁胺醇水平为准。
24.根据权利要求23的扩散型控释制剂,其中AUC 0-t为53074pg.h/ml的80-125%,AUC inf为55606pg.h/ml的80-125%,Cmax为4383pg.h/ml的80-125%。
25.根据权利要求22的扩散型控释制剂,当其为8mg沙丁胺醇的制剂时,其中生物等效性是采取对进食和空腹的健康成年男性以随机、单剂量、三组交叉生物等效性研究来测定,以16个样本和血浆沙丁胺醇水平为基准。
26.根据权利要求25的扩散型控释制剂,其中对进食健康男性,AUC 0-t为106139pg.h/ml的80-125%,AUC inf为109692pg.h/ml的80-125%,Cmax为7149pg.h/ml的80-125%。
27.根据权利要求22的扩散型控释制剂,当其为8mg沙丁胺醇的制剂时,其生物等效性是采取对空腹的健康成年男性以随机、单剂量、双交叉生物等效性研究来测定,以37个样本和血浆沙丁胺醇水平为基准。
28.根据权利要求27的扩散型控释制剂,其中AUC 0-t为1125573pg.h/ml的80-125%,Cmax为14522pg.h/ml的80-125%。
