Description
Die Erfindung betrifft weiche und harte Kontaktlinsen mit
einer ionischen Ladung an der Oberfläche, die mit einer Lin
senlösung behandelt worden sind, welche ein entgegengeladenes
ionisches Polymer enthält, und wobei ein hydrophiler Poly
elektrolyt-Komplex an der Linsenoberfläche gebildet wird,
der als Hydrogel wirkt und die Eigenschaften der Linse ver
bessert.
Kontaktlinsen mit einer verbesserten Sauerstoffdurchlässig
keit sind unter anderem aus der De-OS 26 00 177 und der
US-PS 38 08 178 bekannt. Weiterhin ist es auch bekannt,
daß die Oberflächen von Kontaktlinsen stark hydrophil sind,
damit sie von den Tränen benetzt werden und unverschleierte
Sicht gewähren. Zu diesem Zweck wird gemäß der DE-AS
11 64 120 auf den Kontaktlinsen ein optisch reiner Über
zug aus Proteinen und/oder Polysacchariden aufgebracht.
In der US-PS 39 87 163 wird eine ophthalmische Lösung auf
Basis von Polystyrolsulfonat beschrieben, die zur Behand
lung von durch das Tragen von Kontaktlinsen verursachten
Augenschäden geeignet ist.
Zur Verbesserung der Hydrophilität hat man bereits Kon
taktlinsen mit polymerisierbaren, hydrophilen Monomeren be
handelt und dadurch eine Aufpfropfung der hydrophilen Grup
pen bewirkt. Diese Verfahrensweise ist jedoch umständlich.
Aufgabe der Erfindung ist es, die Hydrophilität von Kontakt
linsenoberflächen zu verbessern, sowie ein Verfahren für
diese Verbesserung zur Verfügung zu stellen. Diese Aufgabe
wird durch eine Kontaktlinse gemäß dem Patentanspruch 1
und ein Verfahren gemäß dem Patentanspruch 7 gelöst. Die
Unteransprüche 2 bis 6 und 8 bis 10 beinhalten bevorzugte
Ausführungsformen.
Gemäß einer bevorzugten Ausführungsform ist die Linse eine
sauerstoffdurchlässige harte Linse mit einer ionischen Ladung
oder der Möglichkeit, eine solche ionische Ladung zu haben.
Vorzugsweise wird die Linse beschichtet, indem man lediglich
die Linse in eine Lösung taucht, die im wesentlichen aus
einem ionischen Polymeren besteht, das in einer Wasserlösung
gelöst ist oder aus einer Wasserlösung, die lösliche organische
Komponenten enthält, welche 0,001 bis 10 Gew.-% der Lösung aus
machen. Das ionische Polymer kann irgendein ionisches Polymer
sein, das mit dem Auge verträglich ist und keine Reizung des
Auges bewirkt und das einen Hydrogel bildet, wenn es elektro
statisch an die Oberfläche der Kontaktlinse gebunden wird.
Weiche und harte Kontaktlinsen aus synthetischen Polymeren
werden im allgemeinen aus neutralen Monomeren und/oder Polyme
ren hergestellt. Bei der vorliegenden Erfindung werden sowohl
weiche als auch harte Kontaktlinsen so hergestellt, daß diese
ionische Stellen an der Linsenoberfläche haben und diese Stel
len können mit einer "Linsenlösung" umgesetzt werden, die
ein entgegengesetzt geladenes, hydrophiles Polymer enthält.
Falls die Oberfläche der Linse polyanionisch ist, kann die Ober
fläche mit einem hydrophilen Polykation umgesetzt werden, wo
durch sich ein hydrophiler Polyelektrolyt-Komplex bildet.
Polyelektrolyt-Komplexe haben gleiche Mengen an Kationen und
Anionen, die jeweils von einer verschiedenen Quelle erhalten
wurden. Diese insgesamt elektrisch neutralen Komplexe liegen
außerdem als ionisch vernetzte Hydrogele vor, die wirksam
Hydrationswasser zurückhalten. Bei der vorliegenden Erfindung
wird eine Oberflächenbeschichtung mit einem Polyelektrolyt-
Komplex auf der Linsenoberfläche geschaffen. Weiche Kontakt
linsen, die vollständig aus einem Polyelektrolyt-Komplex beste
hen, sind bekannt aber sie haben nicht die gewünschten Eigen
schaften, welche die erfindungsgemäß erhaltenen Linsen aufweisen.
Erfindungsgemäß ist es möglich, daß durch die Umsetzung von
ionischen Stellen auf einer Polymeroberfläche oder von poten
tiellen ionischen Stellen die gleichzeitig einen niedrigmole
kulargewichtigen Elektrolyt, wie Natriumchlorid, Chlorwasser
stoff, Natriumsulfat, Natriummethylsulfat oder irgendeinen
anderen ähnlichen Elektrolyt abgeben, eine einschichtige Be
schichtung mit einem Polyelektrolyt-Komplex bewirkt wird.
Polyelektrolyt-Komplexe sind zwar sehr hydrophil aber wasser
unlöslich und können im allgemeinen nur mit gewissen Schwie
rigkeiten in ternären Lösungsmittelsystemen aus Wasser, einer
wasserlöslichen organischen Verbindung und einem niedrigmole
kulargewichtigen Elektrolyten gelöst werden. Dieses Löslich
keitsverhalten bedingt daß die erfindungsgemäß mit einem
Polyelektrolyt-Komplex behandelte Oberfläche sich nur durch
die wäßrige Augenflüssigkeit sehr schwer löst und von der
Linsenoberfläche abtrennt, obwohl diese Oberflächenbeschich
tung durch mechanische Wirkung im Auge aufgrund von Abrieb
erudiert werden kann. Sollte ein Verbrauch des Polyelektrolyt-
Komplexes auf der Linsenoberfläche eintreten, so kann dieser
leicht ersetzt werden, indem man die Linse mit einer entspre
chend geladenen polyionischen Lösung behandelt.
Der Polyelektrolyt-Komplex auf der Linsenoberfläche kann auf
verschiedene Weise hergestellt werden. Ist eine anionische
Oberfläche erwünscht, so kann man diese schaffen, indem man in
eine Linsenformulierung ein Monomer oder mehrere Monomere
einbringt aus der Gruppe der Acrylat- oder Methacrylatsalze,
Vinylsulfonatsalzen, Allyl- oder Methallylsulfonat- oder
-sulfatsalz, Styrolsulfonatsalz, Acryloyloxyethyl- oder
Methacryloyloxyäthylsulfatsalz, einen substituierten Acrylamido-
oder Methacrylamidosulfonatsalz oder von entsprechenden Phos
phonat-, Phosphat- und Phosphitsalzen von polymerisierbaren
Monomeren. Alternativ kann man eine potentielle anionische
Oberfläche für eine anschließende Behandlung mit einem Polykation
nach der Eliminierung einer niedrigmolekulargewichtigen Säure
(wie Chlorwasserstoffsäure) schaffen oder durch anschließende
Behandlung mit einem neutralen basischen Polymer, wodurch
man eine Säure-Base-Neutralisationsreaktion erhält. Solche
anionische Monomere schließen Verbindungen ein, wie Acryl-
und Methacrylsäure, Vinylsulfonsäure, Allyl- und Methallyl
sulfonsäure oder Schwefelsäure, Styrolsulfonsäure, Acrylamido-
oder Methacrylamidosulfonsäure oder eine polymerisierbare
Phosphon- oder Phosphorsäure.
Wird eine kationische Oberfläche gewünscht, so wird diese
dadurch hergestellt, daß man in die Linsenzusammensetzung irgend
ein quaternäres oder protoniertes Monomer oder Monomere ein
bringt aus der Gruppe der Acrylat- oder Methacrylatsalze,
Vinylpyridiniumsalze, Vinylimidazoliumsalze, Vinylimidazolinium
salze, Vinylthiazoliumsalze, Vinylbenzylammoniumsalze, Diallyl
dialkylammoniumsalze oder ähnlichen alkylierten oder proto
nierten polymerisierbaren Sulfonium- oder Phosphoniumsalzen.
Alternativ kann man eine potentiell kationische Oberfläche
schaffen, die dann anschließend mit einer Polysäure behan
delt wird unter Ausbildung einer Säure-Base-Neutralisations
reaktion. Solche potentiellen kationischen Monomeren schließ
en Verbindungen ein, wie Dialkylaminoethylacrylat oder
-methacrylat, Vinylpyridin, Vinylimidazol, Vinylbenzylamin,
einen Vinylalkylether oder ein -sulfid oder ein polymerisier
bares Vinylphosphin.
Es ist auch möglich, ionische Ladungen auf der Linsenober
fläche durch chemische oder elektrische Modifizierung einer
neutralen wiederkehrenden Monomereinheit auszubilden. Bei
spielsweise erhält man eine anionische Oberfläche indem man
ein Polymeres, wie Polymethylmethacrylat, mit einer
wäßrigen Base, wie Natriumhydroxid, behandelt, wodurch man
Natriummethacrylat-Einheiten auf der Linsenoberfläche bildet.
Alternativ kann ein Polyestermaterial mit einer Säure hydroly
siert werden unter Ausbildung von Methacrylsäureeinheiten
auf der Linsenoberfläche, die als potentielle anionische Stel
len wirken. In ähnlicher Weise kann man eine kationische
Oberfläche erhalten, indem man die Linsenoberfläche alkyliert
oder nukleophile Amine, Sulfide oder Phosphin-Einheiten protoniert.
Praktisch alle Materialien für harte oder weiche Kontaktlin
sen sind elektrisch neutrale Polymere oder Copolymere. Solche
Materialien kann man modifizieren, so daß sie ionische
Oberflächengruppen tragen. Eine Behandlungsweise für alle
Linsen besteht darin, daß man die Oberfläche mit Strahlen
hoher Energie in Gegenwart von Luft behandelt, wodurch ionische
Oberflächen gebildet werden (siehe Chapiro, Radiation Chemistry
of Polymeric Systems, Bd. VX, Interscience, New York, 1962
und F. A. Makhlis, Radiation Physics and Chemistry of Polymers,
Wiley and Sons, New York, 1975).
Ein anderes Verfahren zur Modifizierung von Formulierungen
für polymere Linsen besteht darin, daß man ionische (oder
potentiell ionische) Monomere einverleibt. Polymethylmeth
acrylat, welches zur Zeit für harte Linsenflächen das Material
der Wahl ist, ist solchen Modifizierungen zugänglich. Beispie
le für solche Modifizierungen sind die Copolymerisation von
Acrylsäure, Methacrylsäure oder Dimethylaminoethylmethacrylat,
um dadurch Polymethylmethacrylat-Linsen mit ionischen Gruppen
an der Oberfläche zu schaffen.
Ein weiteres Beispiel zum Modifizieren von sauerstoffdurch
lässigen Linsenformulierungen ist aus US-PS 38 08 178 er
sichtlich. Diese Formulierungen sind Copolymere von Methyl
methacrylat mit einem Siloxanylalkylester der Methacrylsäure
und diese können modifiziert werden durch die Zugabe von
entweder Acrylsäure, Methacrylsäure oder Dimethylaminoethylmeth
acrylat.
In ähnlicher Weise kann man monomere Acrylsäure, Methacryl
säure oder Dimethylaminoethylmethacrylat als Coreaktanten
mit Hydroxyethylmethacrylat verwenden unter Bildung eines Ma
terials, das für weiche Kontaktlinsen geeignet ist, die
außerdem ionische Oberflächen tragen.
Zellulosepolymere, wie Zelluloseacetat-butyrat (CAB), finden als
Material für Kontaktlinsen mit guter Sauerstoffdurchlässig
keit Verwendung. Polymere dieser Art enthalten restliche
Zellulosealkoholfunktionalitäten, die als Modifizierungsstel
len verwendet werden können. Die Umsetzung von Natriumchlor
acetat mit den Alkoholfunktionalitäten ergibt Carboxylat
gruppen längs der Polymerkette. Aus diesem modifizierten
CAB-Material hergestellte Kontaktlinsen können inhärent be
feuchtbar gemacht werden mit einer ionischen Oberfläche, die
zur Ausbildung einer Polyelektrolyt-Komplex-Bildung geeignet
ist.
Die Linsenoberfläche der Kontaktlinse hat vorzugsweise
eine Gesamtionenladung von 0,001% bis 10%, was bedeutet,
daß 0,001 bis 10% der Oberfläche geladen sind. Die La
dungsdichte liegt im allgemeinen bei etwa 5%.
Die dünne Schicht des Polyelektrolyten-Komplex-Überzuges
auf der Kontaktlinse hat im allgemeinen eine Dicke im Be
reich von 20 bis 2500×10-10 m.
Ionische Polymere, die in einer Benetzungslösung vorliegen
und zur Befeuchtung der eine ionische Gruppe enthaltenden
Linsenoberfläche geeignet sind, werden nachfolgend beschrieben.

Man verwendet vorzugsweise eine Benetzungslösung, welche
das ionische Polymer in einer Menge von 0,001 bis 10 Gew.-%
enthält. Verwendet man als ionisches Polymer ein kationi
sches Polymer, dann ist das kationische Polymer bevorzugt
kationische Zellulose, insbesondere kationische Hydroxy
ethylzellulose.
Bei einer bevorzugten Kontaktlinse ist das Polymermaterial
aus Monomeren der Formel
worin bedeuten:
R₁ = H, CH₃, CH₂COOH, CH₂COOCH₃ oder
CH₂COOC₆H₅,
und
R₂ =
R₂ =
worin a eine ganze Zahl von 1 bis 3 ist, b und c ganze Zah
len von 0 bis 2 sind, d eine ganze Zahl von 0 bis 1 ist,
A ausgewählt ist aus Methyl- und Phenylgruppen, B ausge
wählt ist aus Methyl- oder Phenylgruppen, C und D entweder
keine Gruppe (zyklischer Ring von c zu d) oder Methylgruppen
oder Phenylgruppen bedeuten, hergestellt worden, wobei die
ses Polymermaterial ionische Gruppen enthält.
Bei einem weiteren bevorzugten Polymermaterial besteht
dieses im wesentlichen aus wiederkehrenden Einheiten der
Formel
worin R₁ und R₂ H, CH₃, C₂H₅, C₃H₇, C₆H₅, COOH, CH₂=CH-
oder -O-Gruppen sind.
Gemäß einer bevorzugten Ausführungsform enthält die Lin
senoberfläche Ionenstellen, die komplex mit eine Gegenla
dung aufweisenden Polymerstellen aus den Gruppen COOX,
SO₃X und PO₃X, worin X=H, oder ein einwertiges anorga
nisches Ion bedeuten, N⁺(R)₃-Gruppen, worin R=H, CH₃ oder
C₂H₅-Gruppen bedeuten, S⁺(R′)₂, worin R′=H, CH₃ oder
C₂H₅-Gruppen bedeutet, P⁺(R′′)₃, worin R′′=H, CH₃, C₂H₅
oder Phenylgruppen bedeuten, Pyridiniumgruppen oder Imi
dazoliumgruppen, verbunden sind.
Proben von harten Polymeren wurden aus Methylmethacrylat (MMA)
und aus einer Comonomer-Mischung aus Methylmethacrylat (MMA)
und Methacrylsäure (MA) hergestellt. Eine geringe Menge an
Tetraethylenglykoldimethacrylat (TEGDM) wurde zu beiden Formu
lierungen als Vernetzungsmittel zugegeben. Als freiradikali
scher Katalysator wurde 2,2′-Azobisisobutyronitril (AIBN) zur
Auslösung der Polymerisation verwendet. Die Zusammensetzungen
für die Formulierung (in Tabelle I in Gewichtsprozent gezeigt)
wurden gründlich vermischt, in Reagenzgläser übergeführt, die
verschlossen und entgast wurden, worauf dann Stickstoff auf
gefüllt wurde. Die Reagenzgläser wurden in ein Wasserbad bei
40°C gestellt und während 2 Tagen wurde die Polymerisation
durchgeführt. Dann wurden die Gläser weitere 3 Tage in einen
Ofen von 60°C gestellt und dann wurden die polymerisierten
Stangen aus den Reagenzgläsern entnommen. Die Stangen wurden
ungefähr 15 Stunden bei 100°C im Vakuum konditioniert um den
Polymerisationsprozess zu beenden und um alle mechanischen
Spannungen auszugleichen. Probestücke in Form von 0,48-1,27 cm dicken
Scheiben wurden aus den Stangen herausgeschnitten. Die
flache bearbeitete Oberfläche der Scheiben wurde hochpoliert,
um eine geeignete Oberfläche für Kontaktwinkelmessungen zu schaf
fen.
Die Kontaktwinkel wurden an hydratisierten Proben nach dem
Eintauchen in H₂O während 2 Tagen bestimmt, indem man die Werte
für die Winkel maß, die Wassertropfen auf der polierten Oberflä
che bildeten. Niedrige Winkel sind ein Zeichen für benetzbare
Materialien und sind einer polareren Oberfläche zuzuschreiben,
entweder als Ergebnis von vorhandenen chemischen Gruppen oder
von vorhandenen gebundenen Wassermolekülen.
Der erheblich niedrigere Kontaktwinkel an der Oberfläche B
zeigt deutlich das Ergebnis der vorliegenden Erfindung. Die
kationische Hydroxyethylzellulose ist ionisch gebunden an die
Carboxylatgruppen (anionisch) an der Oberfläche unter Aus
bildung eines Polyelektrolyt-Komplexes, der Wasser gebunden
enthält.
Man arbeitet wie in Beispiel I und stellt harte polymere Pro
ben aus Methylmethacrylat (MMA), Methacryloyloxypropyl-tris-
(trimethylsilyl)-siloxan (TRIS) und Methacrylsäure (MA) her. Ein
kleiner Anteil an Tetraethylenglykoldimethacrylat (TEGDM) wur
de als Vernetzungsmittel zugegeben. Zur Auslösung der Polymeri
sation wurde als freiradikalischer Initiator 2,2′-Azobisiso
butyronitril (AIBN) verwendet. Die Konzentration der verwende
ten Reagenzien, die Oberflächenbehandlung und die Kontakt
winkelwerte sind in Tabelle II angegeben.
Die Proben mit den niedrigeren Kontaktwinkeln waren nach den
Methoden B oder C behandelt worden und dies zeigt, daß die
Erfindung auf polymere Materialien für harte, sauerstoffdurch
lässige Kontaktlinsen anwendbar ist.
Man arbeitet wie in Beispiel I und stelle harte polymere Pro
ben her aus Methylmethacrylat (MMA), Methacryloyloxypropyl
tris-(trimethylsilyl)-siloxan (TRIS) und Dimethylaminoethyl
methacrylat (DMAEM). Als Vernetzungsmittel wird eine geringe
Menge an Tetraethylenglykoldimethacrylat (TEGDM) verwendet.
Als freiradikalischer Katalysator zur Bewirkung der Polymeri
sation wird 2,2′-Azobisisobutyronitril (AIBN) verwendet. Die
Konzentration der verwendeten Reagenzien, die Oberflächenbe
handlung und die gemessenen Kontaktwinkel werden in Tabelle
III angegeben.
Dieses Beispiel zeigt den Einbau eines kationischen Monomeren
in eine polymere Formulierung, die einer Behandlung mit einem
anionischen Polymeren unter Bildung einer Oberflächenschicht
eines Polyelektrolyt-Komplexes zugängig ist. Dieses Verhalten
zeigt die Vielseitigkeit der vorliegenden Erfindung, bei der
man entweder ein anionisches (Beispiele I und II) oder ein
kationisches (Beispiel III) Monomer in eine polymere Formulie
rung einbauen kann, welches in der Lage ist, einen Polyelektrolyt-
Komplex mit einem Polyion entgegengesetzter Ladung zu bilden.
Claims (10)
1. Kontaktlinse aus einem optischen klaren transparenten
Körper mit linsenförmiger Oberfläche, wobei die linsen
förmige Oberfläche aus einem Polymermaterial mit ioni
schen Gruppen besteht und sich auf der Linsenoberfläche
eine mit ihr elektrostatisch verbundene dünne Schicht
befindet, dadurch gekennzeichnet, daß
die Beschichtung ein ionisches Polymer ist, das einen
Polyelektrolytkomplex mit den ionischen Gruppen an der
Linsenoberfläche bildet.
2. Kontaktlinse gemäß Anspruch 1, dadurch gekenn
zeichnet, daß das Polymermaterial aus Monomeren
der Formel
worin bedeuten:
R₁ = H, CH₃, CH₂COOH, CH₂COOCH₃ oder CH₂COOC₆H₅ und
R₂ = worin a eine ganze Zahl von 1 bis 3 ist, b und c ganze Zahlen von 0 bis 2 sind, d eine ganze Zahl von 0 bis 1 ist, A ausgewählt ist aus Methyl- und Phenylgruppen, B ausgewählt ist aus Methyl- oder Phenylgruppen, C und D entweder keine Gruppe (zyklischer Ring von c zu d) oder Methylgruppen oder Phenylgruppen bedeuten, hergestellt worden ist, und ionische Gruppen enthält.
R₁ = H, CH₃, CH₂COOH, CH₂COOCH₃ oder CH₂COOC₆H₅ und
R₂ = worin a eine ganze Zahl von 1 bis 3 ist, b und c ganze Zahlen von 0 bis 2 sind, d eine ganze Zahl von 0 bis 1 ist, A ausgewählt ist aus Methyl- und Phenylgruppen, B ausgewählt ist aus Methyl- oder Phenylgruppen, C und D entweder keine Gruppe (zyklischer Ring von c zu d) oder Methylgruppen oder Phenylgruppen bedeuten, hergestellt worden ist, und ionische Gruppen enthält.
3. Kontaktlinse gemäß Anspruch 1, dadurch gekenn
zeichnet, daß das Polymermaterial im wesentli
chen aus wiederkehrenden Einheiten der Formel
worin R₁ und R₂ H, CH₃, C₂H₅, C₃H₇, C₅H₅, COOH,
CH₂=CH- oder -O-Gruppen sind, besteht.
4. Kontaktlinse gemäß einem der Ansprüche 1 bis 3,
dadurch gekennzeichnet, daß die
Linsenoberfläche Ionenstellen enthält, die
komplex mit eine Gegenladung aufweisenden
Polymerstellen aus den Gruppen COOX, SO₃X und PO₃X, worin
X=H, oder ein einwertiges anorganisches Ion bedeuten,
N⁺(R)₃-Gruppen, worin R=H, CH₃ oder C₂H₅-Gruppen bedeuten,
S⁺(R′)₂, worin R′=H, CH₃ oder C₂H₅-Gruppen bedeutet, P⁺(R′′)₃
worin R′′=H, CH₃, C₂H₅ oder Phenyl-Gruppen bedeuten,
Pyridiniumgruppen oder Imidazoliumgruppen, verbunden sind.
5. Kontaktlinse gemäß Anspruch 1, dadurch gekenn
zeichnet, daß die Linsenoberfläche eine Ge
samtionenladung von 0,001% bis 10% hat.
6. Kontaktlinse gemäß Anspruch 5, dadurch gekenn
zeichnet, daß die dünne Schicht des Poly
elektrolytenkomplexüberzuges eine Dicke im Bereich
von 20 bis 2500 · 10-10 m hat.
7. Verfahren zur Herstellung einer Polyelektrolytkomplex-
Beschichtung auf einer polymeren Kontaktlinse, da
durch gekennzeichnet,
- a) daß man auf der Linsenoberfläche ionische Gruppen erzeugt und
- b) daß man die Linsenoberfläche mit einer Benetzungs lösung befeuchtet, die ein ionisches Polymer ent hält und dadurch eine Beschichtung aus einem ioni schen Polymer aufbringt, das elektrostatisch an die Oberfläche gebunden ist, indem es mit den ionischen Gruppen an der Linsenoberfläche eine dünne Schicht eines Polyelektrolytkomplexes bildet.
8. Verfahren gemäß Anspruch 7, dadurch gekenn
zeichnet, daß man eine Benetzungslösung verwen
det, welche das ionische Polymer in einer Menge von
0,001 bis 10 Gew.-% enthält.
9. Verfahren gemäß einem der Ansprüche 7 oder 8, dadurch
gekennzeichnet, daß man als ionisches
Polymer ein kationisches Polymer verwendet.
10. Verfahren gemäß Anspruch 9, dadurch gekenn
zeichnet, daß man als kationisches Polymer katio
nische Zellulose, insbesondere kationische Hydroxyethyl-
Zellulose verwendet.
