Die japanischen Offenlegungsschriften
Nr. 102 827/74 und 42 025/75 beschreiben Verfahren zur Herstellung eines Mittels mit langsamer Freigabe,
bei dem eine physiologisch aktive Substanz auf ein Protein unterschiedlicher Herkunft wie Kollagen und Gelatine,
aufgelagert ist Die Wirksamkeit der nach dem herkömmlichen Verfahren hergestellten Mittel mit langsamer
Freisetzung erstreckt sich aber nur auf einige Stunden bis zu gut etwa 10 Tagen. Keines der bisher entwikkelten
Mittel mic langsamer Wirkstoff-Freigabe macht die meisten Proteine als Substa-.z mit dem lebenden
Gewebe verträglich. Die Erfinder haben Untersuchungen an einem Mittel mit Langzei Freigabe durchgeführt,
bei dem auf Protein eine physiologisch aktive Substanz aufgelagert ist die ihre Aktivität bei langsamer
Freigabe über einen verlängerten Zeitraum zeigt Ein Ergebnis dieser Untersuchungen ist die Erfindung eines
Verfahrens zur Herstellung eines Verbundstoffes mit Langzeit-Freigabe, bei dem eine physiologisch aktive
Substanz in ein thermisch denaturiertes Protein eingeschlossen ist Über diese Erfindung ist bereits berichte»
worden. Dieses Verfahren hat jedoch einen Mangel: Zur thermischen Proteindenaturierung ist im allgemeinen
die Anwesenheit von Wasser nötig, und in Anwesenheit von Wasser ist der größere Teil der gebildeten Protein-Matrix
porös, so daß sich die eingeschlossene physiologisch aktive Sbustanz bald aus der Matrix löst. Bei der
Suche nach Wegen zur Lösung dieses Problems haben die Erfinder nun gefunden, daß die langsame Freisetzung
bei dem herkömmlichen Produkt durch Einschluß der physiologisch aktiven Substanz in Polypeptid verbessert
werden kann. Hierfür wurde eine Patentanmeldung eingereicht.
Die vorliegende Erfindung ist eine weitere Verbesserung
dieser Maßnahme. Die Erfindung betrifft ein Verfahren zur Herstellung eines Verbundstoffes mit langsamer
Freigabe einer darin eingeschlossenen physiologisch aktiven Substanz, bei dem man ein System, das
N-Carboxyaminosäureanhydrid und die physiologisch
aktive Substanz umfaßt, unter Druck und bei tiefen Temperaturen einer Formgebung unterzieht.
Das Verfahren zur Herstellung eines Verbundstoffes mit Langzeitfreigabe wurde erfindungsgemäß dadurch
verbessert, daß als Matrixbasis das Monomer eines N-Carboxyaminosäureanhydrids (nachfolgend auch als
NCA bezeichnet) eingesetzt wurde. Bekanntlich polymerisiert der NCA-Kristall an der Atmosphäre, wie von
H. Mark, J. Appl. Phys, 20 (1949), S. 531, und N. H.
Grant, J. Am. Chem. Soa, 85 (1966), S. 4071, berichtet
wurde. Nach diesen Berichten wird NCA in fester Phase unter Benutzung von ionisierender Strahlung als PoIymerisationsinitiator
polymerisiert In beiden Veröffentlichungen wird jedoch nur von einem Polymerisat berichtet
dessen Molekulargewicht so niedrig wie das eines Oligopeptids ist Die Untersuchungen der vorliegenden
Erfindung haben nun jedoch gezeigt, daß NCA dadurch zu einem Polypeptid umgesetzt werden kann, daß
man es bei einer Temperatur zwischen — 200C und 400C und einem Druck zwischen 10 und 20 000 kg/cm2
in eine geeignete Form bringt und daß das Polypeptid zu einem Produkt mit höherem Molekulargewicht dadurch
umgesetzt werden kann, daß man es bei einer Temperatur zwischen — 100°C und 400C mit Licht oder
ionisierender Strahlung bestrahlt Als en Beispiel wurde L-Valin-NCA-Monomer bei Zimmertemperatur unter
2000 kg/cm2 in eine geeignete Form gebracht und das geformte Produkt bei — 500C mit /-Strahlen aus 60Co
bestrahlt, bis sich eine absorbierte Dosis von 1 Mrad ergab. Das Molekulargewicht des Polypeptids wurde in
Trichloressigsäure bei einer Konzentration von 0,25 g Hl gemessen; es wurde gefunden, daß es ein hohes Molekulargewicht
hat (spezifische Viskosität/Konz.=2£).
Hinsichtlich der Art des Lichtes und der ionisierenden Strahlung, die bei der Erfindung benutzt werden können,
besteht keine besondere Beschränkung; es werden jedoch Strahlungsquellen mit großer Durchdringungsenergie
bevorzugt, wie /-Strahlen aus Kobalt 60 und
^-Strahlen aus Strontium 89. Die absorbierte Dosis liegt im allgemeinen in dem Bereich von 5 · 104 bis
5 · 10°rad, wobei eine Dosis von etwa 1 · 10°rad
zweckdienlich ist Die Bestrahlungstemperatur ist im allgemeinen 00C oder weniger, vorzugsweise liegt sie in
dem Bereich zwischen -70 und -1000C Ein Vorteil dieser niedrigen Bestrahlungstemperaturen besteht darin,
daß die physiologisch aktive Substanz nicht inaktiviert wird und daß sie in dem hergestellten Verbundstoff
mit Langzeit-Freigabe gleichmäßig verteilt wird.
Nach der vorliegenden Erfindung dient Polypeptid als
Matrix für den Einschluß der physiologisch aktiven Substanz. Der Grund für die Verwendung des Polypeptids
als Matrix ist der, daß es in hohem Maß mit dem Iebenden Gewebe verträglich ist und durch Enzyme leicht
verdaut wird. Das als Matrix benutzte Polypeptid des erfindungsgemäßen Verbundstoffes mit Langzeit-Freigabe
wird aus NCA hergestellt, ohne daß das Verfahren zur Herstellung des NCA einer besonderen Beschränkung
unterliegt. Bei der praktischen Ausführung der Erfindung wird eine NCA enthaltende, polymerisierfähige
Zusammensetzung zweckmäßigerweise bei einer Tempratur zwischen -200C und 400C polymerisiert.
Diese Temperatur hängt natürlich von der Polymerisationszeitab.
Beispiele für physiologisch aktive Substanzen, die nach der vorliegenden Erfindung verwendet werden
können, sind Bleomycin-Hydrochlorid, Mitomycin C, Adriamycin, Carbazylchinon, Rhomstin, Diphosphamid,
Thioinosin, Citarabin, Fluorouracil, 1 -(2-Tetrahydrofuryl)-5-fluorouracil,
Citotein, Chlorambutyl, Dibrommannit, Thio-TEPA, Cyclophosphamid, Acetylurin, Noradrenalin.
Serotonin, Callicrein, Gastrin, Secretin, Adrenalin, Insulin, Glukagon, /7-Methazon, Indometasin,
ACTH, Wachstumshormon, Gonadotrophin. Oxytocin, Vasopressin, Thyroxin, Testicularhormon, Vesicularhormon,
Lutealhormon, Adrenal-Cortical-Hormon, Prostaglandin, Antithistamine, Blutdrucksenkungsmittel,

Gefäßverengungsmittel, Kapillarstabilisatoren, Magen/ Verdauungsregler, Darmregler, Empfängnisverhütungsmittel,
dermatologische Bakteriozide/Desinfektionsmittel. Mittel zur Behandlung parasitischer Hautkrankheiten,
entzündungshemmende Mittel, Vitamine, Enzympräparate, Impfstoffe, Antiprotozoen-Mittel,
Substanzen, die Interferon induzieren, Anthelmintika
(Wurmmittel), Mittel zur Behandlung von Fischkrankheiten, Agrochemikalien, Auxin-Gibberellin, Cidocainin,
Abietinsäure, Insektenhormon usw.
Beispiele für bei der vorliegenden Erfindung verwendbares NCA sind u. a. Glycin-NCA, Alanin-NCA,
Valin-NCA, Leucin-NCA, Cystein-NCA, Tyrosin-NCA,
Prolin-NCA, Methionin-NCA, Histidin-NCA, Thyroxin-NCA, Asparaginsäure-NCA, Glutaminsäure-NCA,
Oxyglutaminsäure-NCA, Lysin-NCA, Ornithin-NCA
und Arginin-NCA. Es können auch andere Aminosäuren eingesetzt werden, wenn aus ihnen NCA hergestellt
werden kann.
Die vorliegende Erfindung wird nun unter Bezugnahme auf die folgenden Beispiele und Vergleichsbeispiele
näher beschrieben, die hier nur zur Erläuterung angegeben sind und in keiner Weise den Umfang der"Erfindung
einschränken. In den Beispielen wurden Versuche durchgeführt, um zu zeigen, wie verschiedene physiologisch
aktive Substanzen aus dem Verbundstoff mit Langzeit-Freigabe in 100 ml Wasser in Lösung gehen.
Die Versuche wurden nach USP XIX bei 37" C mit einem
mit 100 Umdrehungen pro Minute rotierenden Korb durchgeführt.
Die Fig. 1 und 2 sind graphische Darstellungen, die
den Verlauf der Freigabe der physiologsich aktiven Substanzen aus den Proben des Verbundstoffes mit
Langzeit-Freigabe zeigen, die nach der vorliegenden Erfindung in den Beispielen 1 bis 5 hergestellt wurden.
1 Mrad ergab. Es wurde ein Test durchgeführt, um festzustellen,
wie das 5-FU aus dem Verbundstoff freigegeben wurde. Das Freigabeprofil ist durch das Symbol
— · — in F i g. 2 dargestellt Der Verbundstoff wurde Mäusen unter der Rückenhaut implantiert. Vier Monate
später betrug das Gewicht der den Mäusen entnommenen Probe 60% des Anfangsgewichtes.
Es wurde ein Verbundstoff mit Langzeit-Freigabe wie in Beispiel 4 hergestellt, wobei jedoch das 5-FU durch
Mitomycin C (MMC) ersetzt wurde. Es wurde ein Test durchgeführt, um festzustellen, wie das MMC aus dem
Verbundstoff freigesetzt wird. Das Freigabeprofil war ähnlich dem, das man mit der in Beispiel 4 hergestellten
Probe erhielt

Beispiele 1 und 2
Zwei innige Teilchenmischungen aus 300 mg ^-Benzyl-L-glutamat-NCA
und 300 mg 5-FluorouraciI (5-FU) wurden unter 1000 kg/cm2 (Beispiel 1) und unter
5000 kg/cm2 (Beispiel 2) bei 400C und r. h. 60% geformt
und so Verbundstoffe aus Poly-^Benzyl-L-glutamat mit
darin eingeschlossenem 5-FU hergestellt Es wurde ein Test durchgeführt, um festzustellen, wie das 5-FU aus
den Verbundstoffer, freigesetzt wird Das Freigabeprofil
ist in Fig. 1 dargestellt, wobei die Symbole — O —
und — Δ— die Daten für die in den Beispielen 1 bzw. 2 hergestellten Proben angeben.

Es wurde ein Verbundstoff mit Langzeit-Freigabe wie in Beispiel 1 hergestellt, wobei jedoch das ^-Benzyl-L-glutamat-NCA
durch ein Gemisch aus 150 mg L-Leucin-NCA und 150 mg L-Valin-NCA ersetzt wurde. Es
wurde ein Test durchgeführt, um festzustellen, wie das 5-FU aus dem Verbundstoff freigegeben wird. Das Freigabeprofil
ist in F i g. 2 durch das Symbol — ®— dargestellt.

Ein Verdauungsverbundstoff mit Langzeit-Freigabe aus Copoly-(L-Leucin, L-Valin) mit eingeschlossenem
5-FU wurde wie in Beispiel 3 hergestellt mit der Abweichung, daß es bei -300C mit ^-Strahlen aus 60Co bestrahlt
wurde, so daß sich eine absorbierte Dosis von Hierzu i Blatt Zeichnungen
Claims (2)
1. Verfahren zur Herstellung eines Verbundstoffes mit Langzeit-Freigabe einer darin eingeschlossenen,
physiologisch aktiven Substanz, dadurch gekennzeichnet, daß man eine innige Mischung
aus einem oder mehreren N-Carboxyaminosäureanhydriden und einer oder mehreren physiologisch
aktiven Substanzen bei Temperaturen zwischen - 20° C und 40° C unter einem Druck in dem Bereich
von 10 bis 20 000 kg/cm2 in eine geeignete Form bringt
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet,
daß man den geformten Körper bei einer Temperatur zwischen -1000C und 400C mit Licht
oder ionisierender Strahlung bestrahlt
