Beschreibung
Gegenstand der Erfindung sind substituierte Diphenylazomethine,
ein Verfahren zu ihrer Herstellung und ihre Anwendung
in der Therapie, namentlich in Form von diese Verbindungen als Wirkstoffe enthaltenden Arzneimitteln.
Die erfindungsgemäßen Verbindungen entsprechen der nachfolgenden
allgemeinen Formel (I)
in der R eine Gruppe der folgenden Formeln -(CH-). OH,
-CH2-CHOH-C2H5, -CH2-CHOH-Ch2-COOH,
-CH2-CHOH-CH2CONh2, -CH2CH2-CHOh-CH2OH oder -CH3-CHOH-COOH
bedeutet.
Diejenigen erfindungsgemäßen Verbindungen, die ein asymmetrisches
Kohlenstoffatom enthalten, können in Form des
Racemats oder in Form ihrer Enantiomere!! vorliegen, die
ebenfalls Gegenstand der Erfindung sind.
Nach dem erfindungsgemäßen Verfahren stellt man die Verbindungen
dadurch her, daß man das Benzophenon der Formel

mit einer gegebenenfalls in Form eines optisch aktiven
Enantiomeren vorliegenden Verbindung der allgemeinen Formel
in der R die oben angegebenen Bedeutungen besitzt, umsetzt, und zwar vorzugsweise bei einer Temperatur im Bereich von
30 bis 1200C und in einem Lösungsmittel, wie Ether, Methanol,
Ethanol oder Toluol.
Die folgenden Beispiele dienen der weiteren Erläuterung der Erfindung. Die Analysenwerte, die IR-Spektren und die
NMR-Spektren bestätigen die angegebene Struktur der Verbindungen. Das als Ausgangsmaterial eingesetzte Benzophenon
ist in der FR-PS 75.24065 beschrieben. Die als Ausgangsmaterial eingesetzten Verbindungen der Formel EkN-R
sind aus der Literatur bekannt.
4-{/T2-Chlor-phenyl)-(5-chlor-2-hydroxy-phenyl)-methylen/-amino}-l-butanol
Man beschickt einen 500 ml-Kolben mit 8 g (0,03 Mol) (2-Chlor-phenyl)-(5-chlor-2-hydroxy-phenyl)-methanon,
150 ml Ether und 4 g (0,045 Mol) 4-Amino-1-butanol.

Man erhitzt die Reaktionsmischung zum Sieden, verdampft
das Lösungsmittel und reinigt den Rückstand durch Chromatographie über einer mit Siliciumdioxid beschickten Säule,
wobei man mit einer ChloroCorm/Ethylacetat-Mischung eluiert.
Die das gewünschte Produkt enthaltenden Fraktionen werden vereinigt und zur Trockne eingedampft. Man erhält
ein Öl, welches man aus Petrolether auskristallisiert. F (Mettler) = 91,80C.
4-{/T2-Chlor-phenyl)-(5-chlor-2-hydroxy-phenyl)-methylen/-amino}-2-butanol
Man erhitzt eine Mischung aus 3,4 g (2-Chlor-phenyl)-(5-chlor-2-hydroxy-phenyl)
-inethanon , 1,07 g 4-Amino-2-butanol und 200 ml Methanol zum Sieden am Rückfluß. Man verfolgt
den Ablauf der Reaktion dünnschichtchromatographisch und gibt regelmäßig 4-Amino-2-butanol zu, bis das üünnschichtchromatogramm
das Verschwinden des Ketons anzeigt.
Dann dampft man zur Trockne ein, nimmt den Rückstand mit Wasser und Methylenchlorid auf, dekantiert die organische
Phase ab, trocknet über Magnesiumsulfat, filtriert und dampft zur Trockne ein. Man reinigt den Rückstand über einer
mit Siliciumdioxid beschickten Säule, wobei man mit Ethylacetat eluiert, und kristallisiert das Material
schließlich nach der Behandlung mit Pflanzenkohle aus Pentan um.
F = 71,5 bis 73,50C.
Die Enantiomeren dieser Verbindung erhält man in gleicher
Weise durch Umsetzen von (2-Chl or-phenyl)-(5-chlor-2-hydroxy-pheny1)-methanon
und den entsprechenden Enantiomeren des 4-Amino-2-butanols.

Die erfindungsgemäßen Verbindungen wurden pharmakologisehen
Untersuchungen unterworfen, die ihre Wirkung auf dos Zentralnervensystem erkennen lassen.

Die akute Toxizität wurde an der Maus bei intraperitonealer
Verabreichung untersucht. Die DL,_0-Werte (Dosis letalis
50 %) variieren von 200 bis 600 mg/kg.
Die antidepressive Wirkung der erfindungsgemäßen Verbindungen
wurde über den Antagonismus gegen das durch die Verabreichung von L-5-Hydroxytryptophan an Mäuse verursachte
"Kopf zucken" nachgewiesen. Hierzu verabreicht man männlichen Mäusen (CDI, Charles River Frankreich, mit einem
Körpergewicht von 18 bis 22 g) die zu untersuchenden Verbindungen in wachsenden Dosierungen oder auch nur das Lösungsmittel
gemeinsam mit L-5-Hydroxytryptophan in einer Dosis von 250 mg/kg auf subkutanem Wege. 45 Minuten nach
der Injektion von L-5-Hydroxytryptophan zählt man für jede Maus das "Kopfzucken" während einer Minute aus. Für jede
Behandlung berechnet man den Mittelwert der "Kopfzukkungen" sowie den Prozentsatz der Variation im Vergleich
zu der Kontrollgruppe. Ausgehend von der Wirkungs/Dosis-Kurve bestimmt man den DA5~-Wert (d.h. die aktive Dosis
50 % oder die Dosis, die den Mittelwert der KopfZuckungen
um 50 % vermindert) mit Hilfe der graphischen Methode von Miller und Tainter (1944). Der DA^-Wert der erfindungsgemäßen
Verbindungen variiert bei intraperitonealer Verabreichung von 20 bis 40 mg/kg.
Die antikonvulsive Wirkung der erfindungsgemäßen Verbindungen
wurde über den Antagonismus gegenüber der durch die Verabreichung von Bicucullin an Mäuse verursachten
Mortalität nachgewiesen. Bicucullin ist ein relativ selektiver
Blocker der GABA-ergischen, postsynaptischen Rezeptoren, dessen konvulsive und letale Wirkungen durch
Verbindungen antagonisiert werden, die den cerebralen GABA-Spiegel steigern oder die eine GABA-mimetische Wirkung
entfalten (GABA = ^-Aminobuttersäure).
Man bestimmt die aktive Dosis 50 % (DA50-Wert) der unter-

suchten Verbindungen, d. h. die Dosis, die 50 % der Tiere gegen die Wirkung des Bicucullins schützt. Bei den erfindungsgemäßen
Verbindungen variiert der DA1-„-Wert bei intraperitonealer
Verabreichung von 20 bis 100 mg/kg. 5
Die erfindungsgemäßen Verbindungen sind als Antidepressiva
und Antikonvulsiva wirksam und entfalten weiterhin anxiolytische,
analgetische und antiinflammatorische Wirkungen. Sie können in der Human- und Veterinärtherapie zur Behandlung
von verschiedenartigen Erkrankungen des Zentralnervensystems eingesetzt werden, beispielsweise zur Behandlung
von Depressionen, Psychosen und bestimmten Nervenerkrankungen, wie der Epilepsie, spastischen Erkrankungen,
der Dyskinesie etc.
Gegenstand der Erfindung sind daher auch pharmazeutische Zubereitungen oder Arzneimittel, die die Verbindungen der
obigen allgemeinen Formel (I) als Wirkstoffe in Kombination mit für den Verabreichungsweg geeigneten, pharmazeutisch

können insbesondere auf oralem Wege (in Form von Tabletten, Dragees, Gelatinekapseln, Kapseln, Cachets
oder trinkbaren Lösungen oder Suspensionen) oder auf parenteralem Wege verarbreicht werden. Die tägliche Dosierung
kann sich von 250 bis 5000 mg erstrecken.

- 3. 4-{/T2-Chlor-phenyl)-(5-chlor-2-hydroxy-phenyl)-methyl en/-ainino} -2-butanol.
- 4. Verfahren zur Herstellung der Verbindungen nach den Ansprüchen 1 bis 3, dadurch gekennzeichnet, daß man das Bcnzophenon der Formelmit einer gegebenenfalls in Form des optisch aktiven Enantiomeren vorliegenden Verbindung der allgemeinen Formelin der R die in Anspruch 1 angegebenen Bedeutungen besitzt, umsetzt.
- 5. Arzneimittel, dadurch gekennzeichnet, daß es als Wirkstoff eine Verbindung nach den Ansprüchen 1 bis J enthält.
- 6. Pharmazeutische Zubereitung, dadurch gekennzeichnet , daß sie eine Verbindung nach den Ansprüchen 1 bis 3 in Kombination mit pharmakologisch annehmbaren, geeigneten Bindemitteln, Trägermaterialien und/oder Hilfsstoffen enthält.
