Description
Die vorliegende Erfindung betrifft ein Verfahren zur Her
stellung einer bioverträglichen Stoffzusammensetzung, die
Poly-2-hydroxyethylmethacrylat und Hyaluronsäure oder ein Salz
davon enthält, sowie geformte Gegenstände, die solche Zusam
mensetzungen enthalten oder daraus gebildet sind.
Es ist bekannt, prothetische Vorrichtungen, wie künstliche
Herzklappen, vaskuläre Transplantate etc. für ihre in-vivo-
Anwendung durch Modifizieren des Artikels bzw. der prothe
tischen Vorrichtung mit einer körperverträglichen Substanz
bioverträglicher zu machen. Hyaluronsäure ist eine solche
bekannte, natürlich vorkommende Substanz, die viele Anwen
dungen in der Medizin und der Biologie aufweist. Hierzu wird
beispielsweise auf die US 42 72 522 und die darin angege
benen Veröffentlichungen hingewiesen.
Aus der DE-OS 26 31 908 ist ein vernetztes Polymeres aus
Kollagen und einem Mucopolysaccharid bekannt. Als ein solches
vernetztes Polysaccharid wird dort beispielsweise auch
Hyaluronsäure beschrieben. Dabei werden jedoch
Mucopolysaccharidlösungen eingesetzt.
Die Erfindung hat nun zum Ziel, polymere Gegenstände wie
Gußartikel, Filme, Folien, Fasern, Gewebe und Gewirke be
reitzustellen, die Hyaluronsäure oder Salze davon enthalten,
wobei die Hyaluronsäure durch den Artikel hindurch dispergiert
und/oder als Überzug bzw. Auflage an der Oberfläche vorliegen
soll.
Dieses Ziel wird durch das in Anspruch 1 definierte Verfahren
erreicht. Dabei kann in einer bevorzugten Ausführungsform die
Hyaluronsäure auch vernetzt werden, und zwar nach Herstellung
des Artikels. Die auf diese Weise modifizierten polymeren
Artikel sind daher gut geeignet zur Verwendung bei verschie
denen prothetischen Vorrichtungen, wie künstliche Herzklappen,
intraokulare Linsen, vaskuläre Transplantaten, Schrittma
cherleitungen und dergleichen, sowie bei verschiedenen Arten
von Arzneimittelzuführungssystemen.
Bei dem erfindungsgemäßen Verfahren wird die Hyaluronsäure in
ein Polymer ein- und/oder auf die Oberfläche eines polymeren
Körpers aufgebracht, indem man die Hyaluronsäure mit einer
Polymerlösung oder einer Emulsion mischt und daraus einen
Artikel oder Gegenstand, beispielsweise einen Film, bildet
oder indem man diese Mischung als Überzug bzw. Auftrag auf
einen solchen Gegenstand oder Artikel aufbringt. Dabei kann
sowohl die Oberfläche des polymeren Substrates als auch die
Hyaluronsäure aktiviert werden.
Gemäß in dem erfindungsgemäßen Verfahren wird die
Hyaluronsäure in fester Form und zwar vorzugsweise als Pulver
einer Polymerlösung aus Poly-2-hydroxyethylmethacrylat beige
mischt und danach ein Artikel, beispielsweise ein Film aus der
erhaltenen Mischung geformt und/oder ein Gegenstand wird
mittels einer solchen Mischung überzogen und beschichtet.
Schließlich ist es erfindungsgemäß auch möglich, in dem zuvor
beschriebenen Verfahren die Hyaluronsäure vor oder nach dem
Mischen mit dem Polymer unter Verwendung verschiedener
Vernetzungsmittel zu vernetzen.
Die Erfindung wird durch die folgenden Beispiele näher erläu
tert.
Bei diesem Beispiel wurde ein nichtkovalent vernetztes
Hydrogel, das Natriumhyaluronat enthält, in einer Hydro
gelmatrix von Poly-2-ydroxyethylmethacrylat zuberei
tet. Das Natriumhyaluronat ist durch die gesamte Matrix
des Hydrogels und an der Oberfläche des Hydrogels ver
teilt. Diese Substanz besitzt verbesserte elastische
Eigenschaften und Bioverträglichkeitseigenschaften ge
genüber dem gleichen Material, das Natriumhyaluronat
nicht enthält.
1g einer linearen Poly-2-hydroxyethylmethacry
lat-p-HEMA Polymerzubereitung wurde verwendet. Das
Polymer wurde 10-15 Minuten lang mit dem Minimum von
destilliertem Wasser vorgequollen. Das vorgequollene
p-HEMA wurde dann langsam mit Ethanol bedeckt, wobei
gerührt wurde, bis ein Gesamtbetrag von 20 ml zugege
ben worden war, was eine viskose Lösung zur Folge hatte.
Diese wurde dann zentrifugiert, um irgendwelches ungelö
stes p-HEMA oder einen sonstigen unlöslichen Stoff zu
entfernen. 2-4 mg Na-HA wurde vorgequollen, dann in
10 ml destilliertem Wasser gelöst. Ein gleiches Volumen
Ethanol wurde dann beigemischt. Die p-HEMA-Lösung und
die Na-HA-Lösungen wurden dann einander beigemischt,
und das Gemisch wurde entweder in Glasschalen gegossen
und in einem Gebläseofen bei 60°C getrocknet, um Filme
zu erhalten, oder auf einen rotierenden Glaskern (mit
Silikon bedeckt) aufgebracht und durch Verdampfung in
einem warmen Luftstrom getrocknet, um eine Röhre zu er
halten. Das getrocknete Polymergemisch ergab einen sprö
den Film, der in Wasser quellen mußte, um ein elastisches,
nichtsprödes Material mit opakem Aussehen zu erhalten.
Die Bioverträglichkeit der Zubereitungen gemäß der
Erfindung wurde durch den nachstehend beschriebenen Ver
such bewiesen.
Die Freisetzung von 3H-Serotonin durch menschliche
Blutplättchen wurde in vorausgehenden Studien angewendet,
um die Blutreaktivität des Produktes des Beispiels 1 ab
zuschätzen. Normales menschliches venöses Blut wurde in
Kunststoffspritzen abgezogen und unmittelbar in Kunst
stöffröhrchen übermittelt, die 3,8% Natriumcitrat (ein
Teil Citrat und neun Teile Vollblut) enthielten. Blut
plättchenreiches Plasma wurde durch Zentrifugierung bei
4°C 15 Minuten lang, 125 x g, zubereitet und durch eine
serologische Pipette in eine Kunststoff- oder silizierte
Teströhre entfernt. 3H-Serotonin (3H-5-hydroxytryptamine,
3H-5HT: New England Nuclear, 26,3 Ci/mmol, lmCi/ml
ethanol-water) wurde dem plättchenreichen Plasma (plate
let rich plasma = PRP), 0,2-0,5 ul/ml PRP, beigemischt
und 15 Minuten lang bei 37°C inkubiert. Bei der Prüfung
wurden silizierte oder Polypropylen-Teströhren verwendet;
Thrombin wurde als Positivkontrolle benutzt, belegte und
unbelegte Proben wurden geprüft. 1,0-2,0 ml 3H-5HT-PRP
wurde jeder von Duplikatröhren, die zu untersuchende
Proben enthielten zugegeben; eine 50 ul Aliquote wur
de zur Ermittlung der Gesamtradioaktivität aus dem Kon
trollgemisch entfernt. Im Anschluß an die entsprechende
Inkubationszeit (10-120 Minuten) wurden 0,2-0,5 ml Ali
quoten der Suspension entfernt und 2 Minuten bei
12 000 x g über Silikonöl in einer Eppendorf-Microfuge
zentrifugiert. 50 ul des Überstandes wurden aus jeder
Röhre entfernt, mit 5 ml eines flüssigen Szintillations
fluids vermischt, und die Radioaktivität wurde durch
Betaspektrometrie gemessen. Der Betrag von 3H-5HT, durch
Thrombin freigesetzt oder die Testproben war die Zu
nahme an Radioaktivität des Überstandes (Radioaktivität
der experimentellen Proben minus Radioaktivitätskontrol
le). Die überzogenen Proben des Produktes des Beispiels
1 waren konsistent weniger reagierend, vom Betrag des
freigesetzten 3H-5HT aus betrachtet; die nichtüberzo
genen Proben bewirkten eine 52% größere Freisetzung als
die überzogenen Proben.
Claims (6)
1. Verfahren zur Herstellung einer bioverträglichen
Poly-2-hydroxymethacrylat und Hyaluronsäure oder ein Salz
davon enthaltende Stoffzusammensetzung, indem man zu einer
Lösung des Acrylats Hyaluronsäure oder ihr Salz zusetzt und
die Zusammensetzung abscheidet,
dadurch gekennzeichnet,
daß man die Hyaluronsäure oder ihr Salz
- a) in fester pulverförmiger Form oder,
- b) in Form von mit Hyaluronsäure oder dem Salz beschichteten Partikeln einsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man
aus der Stoffzusammensetzung einen Körper bildet und die an
der Oberfläche des Körpers vorliegende Hyaluronsäure
vernetzt.
3. Ein geformter Gegenstand, hergestellt aus einer
Zusammensetzung nach einem der Ansprüche 1 oder 2.
4. Gegenstand nach Anspruch 3, dadurch gekennzeichnet, daß er
ein Gußartikel, ein Film, eine Folie, eine Faser, ein
Gewebe oder ein Gewirke ist.
5. Geformter Gegenstand, beschichtet mit einer Zusammensetzung
nach einem der Ansprüche 1 oder 2.
6. Gegenstand nach Anspruch 5, dadurch gekennzeichnet, daß er
ein Gußartikel, ein Film, eine Folie, eine Faser, ein
Gewebe oder ein Gewirke ist.
