Das im Nachfolgenden näher beschriebene Verfahren der Indigosynthese beruht auf der Beobachtung, dafs Phenylglykokoll (Phenylamidoessigsäure C6 H~ N H~ C H~ C O O H) beim Schmelzen mit Kali oder Natron in eine Leukoverbindung übergeht, deren alkalische Lösung bei Berührung mit dem Sauerstoff der Luft alsbald Indigo abscheidet.The process of indigo synthesis described in more detail below is based on the observation that phenylglycoll (phenylamidoacetic acid C 6 H ~ NH ~ CH ~ COOH), when melted with potash or soda, turns into a leuco compound, the alkaline solution of which is immediately indigo on contact with the oxygen in the air separates.

Unter Aufschäumen und Dampfentwickelung färbt sich die Schmelze tief orangegelb. Rascher wird dieser Punkt erreicht, wenn man die Temperatur weiter auf 300 oder 3500 steigert. Die angegebene Menge Kali braucht nicht streng festgehalten zu werden, man kann etwas weniger, aber auch, 'mehr Kali verwenden und wird zu demselben Resultat kommen, nur die Ausbeute an Indigo wird dadurch beeinflufst. Während der Schmelzoperation werden Proben genommen und in Wasser gebracht.
