- BEREICH DER ERFINDUNG
- Die vorliegende Erfindung betrifft die Verwendung konjugierter Linolsäure (CLA) für die topische Behandlung von Fettdepots und Zellulite und neue topische Zusammensetzungen.
- Insbesondere betrifft die Erfindung kosmetische und dermatologische topische Zusammensetzungen für die kosmetische Behandlung von Fettdepots und Zellulite enthaltend CLA sowie Sets enthaltend CLA für die besagte Behandlung.
- HINTERGRUND DER ERFINDUNG
- Konjugierte Linolsäure (CLA) ist eine Mischung von Positions- und Konfigurationsisomeren von Octadecadiensäure, welche natürlich auftretende Substanzen sind, die in Milch und Milchprodukten sowie in Fleisch von Wiederkäuern gefunden werden.
- Der Ausdruck CLA umfasst die Familie von Positions- und Konfigurationsisomeren der C18:2 Fettsäuren, genauer die Cis- und Trans- Form der 9, 11 – 10, 12 – und 11, 13-Octadecadiensäuren.
- In viele Studien wird berichtet, dass synthetische CLA in Versuchsmodellen ein wirksames Mittel ist zum Hemmen von Brust-, Darm-, Vormagen- und Hautkrebsbildung, infolge ihrer Regulierung der Lymphozyt- und Makrophage-Aktivität. Neueste klinische und in vivo Werte haben neuartige biologische Wirkungen von CLA beschrieben, z.B. die antiatherogenische und anti-hyperinsulinemische Aktivitäten.
- Seitdem CLA in der internationalen wissenschaftlichen Gemeinschaft für ihre obigen therapeutische Eigenschaften aufgefallen ist, gewinnt sie weiter an Akzeptanz seitens der Verbraucher als Nährstoffergänzung, da gezeigt wurde, dass eine CLA-angereicherte Diät eine bedeutsame Besserung des gesamtheitlichen Gesundheitszustandes bewirkt.
- CLA ist auch als Abmagerungsmittel bekannt, deren orale Einnahme eine markante Abnahme des Körperfetts bewirkt mit einer Zunahme der mageren Körpermasse. Die Wirkungen von CLA auf das Verhältnis Körperfett/fettlose Körpersubstanz scheinen an der Hemmung sowohl der Vermehrung als auch der Differenzierung der Preadipozyten zu liegen, wie beobachtet von Brodie A.E. et al. in J. Nutr. 129:602-6 (1999).
- Die Verwendung von CLA in einer topischen Zusammensetzung und in einer kosmetischen Methode für die Behandlung von Hautzuständen, ausgewählt von der Gruppe umfassend runzelnde, hängende, photogeschädigte Haut, empfindliche Haut, trockene Haut, flockige Haut, gerötete Haut, irritierte Haut, juckende Haut und Altersflecken, ist in WO 0037040 beschrieben.
- Topische Zusammensetzungen mit Zinksalzen von CLA für die Behandlung von Hautbeschwerden, wie Ekzeme, Psoriasis und Dermatitis und so weiter sind in WO 98/17269 beschrieben. Die Verwendung von Zinksalzen von CLA ist jedoch auf pharmazeutische und dermatologische Zwecke begrenzt. Ausserdem vermindert die schwache Löslichkeit der Zinksalze von CLA, sowohl in lipophilen als auch in hydrophilen Medien, bedeutsam die biologische Verfügbarkeit des aktiven Wirkstoffs in der besagten topischen Behandlung.
- DETAILLIERTE BESCHREIBUNG DER ERFINDUNG
- In der vorliegenden Beschreibung umfasst der Ausdruck CLA entweder CLA in Form von freier Fettsäure oder ihren Derivaten, wie beispielsweise ihr Phospholipid, ihre Mono-, Di- und Triglyceride, Ether, Ester oder ihre Salze. Alle Derivate müssen physiologisch annehmbar sein, d.h. ungiftige Derivate von CLA. Bevorzugte Salze von CLA umfassen die Metallseifen von CLA mit alkalischen und/oder erdalkalischen Ionen, wie beispielsweise Natrium-, Kalium- oder Magnesiumionen, und die Stickstoff enthaltenden Salze, wie beispielsweise Ammoniak, Mono-, Di- oder Triethanolamine.
- Wir haben zuerst die Möglichkeit in Betracht gezogen, CLA in der Behandlung von Zellulite, Vertiefungen der Haut der Oberschenkel und des Gesässes, welche durch Dermohyperdermose und oedematöse-sklerotische Panniculopathie verursacht sind, zu verwenden, in welcher die fibroblastische Reaktion über die kapillaro-venolären Veränderungen vorherrscht.
- Wie beobachtet von Rosenbaum M; Prieto V; Hellmer J; Boschmann M; Krueger J; Leibel RL; Ship AG (Plast Reconstr Surg, 101 (7):1934-9 1998), in der in vitro pathologischen Untersuchung von Keilbiopsien und in der in vivo sonographischen Untersuchung des Schenkels, zeigten beide ein diffuses Extrusionsmuster von unterliegendem Fettgewebe in der retikulären Dermis.
- Adipocyten von aufgebauschter Grösse dringen in Mikroknötchen und später in Makroknötchen ein, die durch mehr oder weniger strukturierte konjunktive Fibrille abgegrenzt sind, wie von Merlen JF; Curri SB; Sarteel AM (Phlebologie, 32(3):279-82, 1979) erwähnt.
- Infolge der Ungewissheit in der Zellulite Ätiologie, wurde der besagte unästhetische Zustand bisher mit einer Vielzahl von aktiven Wirkstoffen behandelt, die jeweils durch verschiedene Mechanismen wirkten.
- Wir haben nun herausgefunden, dass die topische Verabreichung von CLA die Abnahme von Fettdepots beschleunigt und verursacht.
- Wir haben somit herausgefunden, dass CLA wirksam ist in der topischen Behandlung von geschädigten ästhetischen Zuständen, welche durch subkutane Fettablagerungen verursacht sind, insbesondere bei pannicularer Adiposis, wo die besagte topische Behandlung zu einer bedeutsamen Verbesserung des Zellulitezustands führt.
- Daher ist eines der Aufgaben der vorliegenden Erfindung die Verwendung von CLA für die topische Behandlung und/oder für die Vorbeugung der Bildung von Fettdepots und Zellulite.
- CLA ist insbesondere geeignet für die topische Anti-Zellulite-Behandlung; es ist ein natürliches Produkt, welches nach einer topischen Behandlung keine beobachtete Nebenwirkungen oder Toxizität an Menschen gezeigt hat. Nach mehreren Anwendungen auf Menschen beider Geschlechter entweder mit normaler Haut oder mit Haut in einem pathologischen Zustand, z.B. mit Hautrötung und juckender Haut, ist keine Unverträglichkeit des Produkts in Erscheinung getreten.
- Die physikalischen und chemischen Eigenschaften von CLA sind insbesondere geeignet für eine topische Hautbehandlung, da sie eine gute Lipidlöslichkeit aufweist und leicht in die Hornschicht absorbiert wird.
- Um die kosmetische Behandlung gemäss der Erfindung zu nutzen, wird CLA vorzugsweise in Form einer topischen Zusammensetzung verabreicht, wobei die besagte Zusammensetzung einen CLA-Gehalt von 2 bis 70 Gew.% aufweist, gegebenenfalls mit Beimischung von geeigneten üblichen Hilfsmitteln.
- In einer bevorzugten Ausführungsform der vorliegenden Erfindung wird CLA mit einem oder mehreren wohlbekannten Anti-Zellulite-Wirkstoffen kombiniert.
- Insbesondere bevorzugte bekannte Anti-Zellulite-Wirkstoffe sind Substanzen, die eine Beta-Stimulation (Adrenergische Beta-Agonisten) aufweisen, um die Lipolyse in den dermischen Adipocyten weiter zu fördern. Beispiele von solchen Substanzen sind Xanthine wie beispielsweise Koffein, Theophyllin, Theobromin und Aminophyllin, welche gekennzeichnet sind durch eine hohe Hautverfügbarkeit und eine hohe Wirksamkeit. Xanthine sind vorzugsweise verwendet in einem Anteil von mindestens 0.05 Gew.%, im Allgemeinen in einem Anteil von 0.05 Gew.% bis 20 Gew.%, vorzugsweise von 0.10 Gew.% bis 10 Gew.%, optimal von 0.5 Gew.% bis 3.0 Gew.%, berechnet auf das Gewicht der Zusammensetzung, um die Wirksamkeit bei optimalen Kosten zu maximieren.
- Andere bevorzugte an sich bekannte Anti-Zellulite-Wirkstoffe sind Substanzen, die als Stimulatoren der Kollagen-Synthese wirken, wie beispielsweise Askorbate und Triterpenoide von Centella Asiatica, d.h. Asiatische Säure, Madekassische Säure, Asiaticosid, Madecassosid, Inositolphosphat und Phytinsäure.
- Andere bevorzugte an sich bekannte Anti-Zellulite-Wirkstoffe sind Substanzen, die den schwachen Vaskularitätszustand in Verbindung mit dem zellulitischen Bereich verbessern, durch eine vasokinetische Aktivität, wie beispielsweise Minoxidil, Nicotinate, Escin, Ivy und Methylsalizylat.
- Andere bevorzugte an sich bekannte Anti-Zellulite-Wirkstoffe sind natürliche Substanzen, die Adenylat-Cyclase Agonisten und/oder Anti-Phosphodiesterase-Aktivitäten ausüben, welche die Verminderung von Fettdepots beschleunigen, die sich in dem von Zellulite betroffenen Bereich befinden. Die erste Gruppe kann Extrakte von Ipomea spp., von Salvia spp. und von Rosmarinus officinalis enthalten, die letztere Gruppe die Yohimbinartige Alkaloide und jene Pflanzenextrakte (z.B. Ginkgo Biloba), welche dimere Flanone enthalten, wie beispielsweise Amentoflavon, Bilobetin, Sciadopitisin, Ginkgonetin oder Extrakte von gewissen Malvaceae (z.B. Malva, Althea, Hibiscus, Hoheria, Sidalcea, Abutilon und Gossypium).
- Für die Behandlung von Zellulite und Fettdepots kann topische CLA auch in Kombination mit Vanadium-Verbindungen verwendet werden.
- Vanadium-Verbindungen sind bekannt für ihre Wirkung als insulinmimetische Substanzen, und sind somit fähig, die Glycolise und die metabolische Umsetzung in Zellen, umfassend Adipocyte, zu verbessern.
- Gemäss einer besonderen Ausführungsform betrifft die vorliegende Erfindung somit eine kosmetische Zusammensetzung, welche CLA und mindestens eine Vanadium-Verbindung enthält.
- Vanadium (IV) oder (V) – Verbindungen sind in den kosmetischen Zusammensetzungen der vorliegenden Erfindung geeigneterweise in einer Konzentration im Bereich von 10–10 bis 10–3 mol/kg, vorzugsweise von 10–10 bis 10–5 mol/kg enthalten.
- Erläuternde Beispiele von geeigneten Vanadium (V) – Verbindungen, die nützlich sind für die Anwendung gemäss der vorliegenden Erfindung umfassen Natrium Metavanadat (NaVO3), Orthovanadat (Na3VO4) und Pyrovanadat (Na4V2O7), die entsprechenden Salze mit Kalium (KVO4), Ammonium (NH4VO3), Kalzium [Ca3(VO4)2], Eisen (Fe(VO3)3] und die entsprechenden Salze von Vanadaten mit Magnesium, Zink, Aluminium und Ähnlichen; die Vanadium (V) – Oxyde wie beispielsweise Pentoxid (V2O5), Oxytrichlorid (VOCl3), Oxytribromid (VOBr3) und Ähnliche, sowie Polymere wie beispielsweise ein Dimer (H2V2O7), ein Trimer (V3O9), ein Decamer (HV10O28) und Ähnliche.
- Erläuternde Beispiele von geeigneten Vanadium (V) – Verbindungen, die nützlich sind für die Anwendung gemäss der vorliegenden Erfindung, umfassen Vanadylsulfat (VOSO4) und entsprechende Verbindungen mit Acetat, etc.; Vanadium (IV) – Oxyhalogenide wie beispielsweise Oxychlorid (VOCl2), Oxydibromid (VOBr2) und Oxydifluorid (VOF2); Vanadium (IV) – Halogenide wie beispielsweise Tetrachlorid (VCl4), Tetrabromid (VBr4) und Tetrafluorid (VF4) und Ähnliche; Vanadiumdioxid (VO2) und Vanadium-Tetraoxid (V2O4).
- Im Weiteren können Vanadium (IV) oder (V) – Verbindungen auch enthalten sein in Form von Chelaten, Clathraten oder anderen Komplexen, umfassend jene mit Aminosäuren, Proteinen, peptidischen Wachstumsfaktoren, Nukleinsäuren, Phosphaten, Phospholipiden, Fettsäuren, Prostaglandine, AHAs, Retinoide, Tris-Edatat, Glykole, Catechole, Glutathion und Ähnliche.
- Vanadium (IV) oder (V) – Verbindungen können auch als Salze von organischen Säuren enthalten sein und als Vanadium, das in Tunikaten (Seescheiden), gewissen Pilzarten und Pflanzen und andere organische Quellen enthalten ist. Spezifische Beispiele von Vanadium Organometallverbindungen umfassen Vanadylsalze von organischen Säuren wie beispielsweise: Vanadyllinoleat, -oleat, -palmitat, -phenolat, -resinat und -stearat.
- Alle erfindungsgemässen Zusammensetzungen können auch jegliche kosmetisch annehmbare Bestandteile umfassen. Der Ausdruck „kosmetisch annehmbare Bestandteile" bezeichnet in der vorliegenden Beschreibung Produkte, die zur Verwendung in kosmetischen Behandlungen geeignet sind, zum Beispiel diejenigen, die in der von der European Cosmetic Toiletry and Perfumery Association (COLIPA) erstellten und im 96/335/EC „Annex to Commission Decision of 8 May 1996" herausgegebenen INCI Liste enthalten sind.
- Eine Vielfalt von aktiven Wirkstoffen kann im Weiteren den erfindungsgemässen Zusammensetzungen zugefügt werden. Obwohl die Wirkstoffe nicht auf diese Kategorie begrenzt sind, umfassen die allgemeinen Beispiele entzündungshemmende Mittel und hautbleichende Mittel, Antioxidationsmittel und die Faltenbildung hemmende Mittel.
- Geeignete entzündungshemmende Verbindungen umfassen, ohne sich jedoch auf diese zu beschränken, Rosmarinsäure, Giycyrrizinat Derivate, alpha-Bisabolol, Azulen und deren Derivate, Asiaticosid, Sericosid, Ruscogenin, Escin, Escolin, Quercetin, Rutin, Betulinsäure und deren Derivate, Catechin und deren Derivate.
- Geeignete hautbleichende Mittel umfassen, ohne sich jedoch auf diese zu beschränken, Ferulsäure, Hydroquinon, Arbutin und Kojinsäure.
- Geeignete Antioxidationsmittel und die Faltenbildung hemmende Verbindungen umfassen, ohne sich jedoch auf diese zu beschränken, Retinol und Derivate, Tocopherol und Derivate, Salicylate und deren Derivate.
- Ein anderes wichtiges Mittel, das der kosmetischen Zusammensetzung gemäss der Erfindung zugefügt werden kann, ist eine alpha-Hydroxysäure. Bevorzugte alpha-Hydroxysäuren sind Monocarboxylsäuren, die das Hauteindringen und die Wirksamkeit von CLA verbessern und weitere an sich bekannte Zellulite hemmende Mittel, wie beispielsweise Milchsäure, Glycolsäure, Mandelsäure und deren Gemische. Vorzugsweise ist der Anteil des alpha-Hydroxysäure – Bestandteiles in der kosmetischen Zusammensetzung der Erfindung von 1.5 Gew.% bis 15 Gew.%, insbesondere von 3.0 Gew.% bis 12.0 Gew.%, berechnet auf das Gewicht der Zusammensetzung.
- Ein anderes wichtiges optionales Bestandteil ist ausgewählt von essentiellen Fettsäuren (EFAs), die eine wichtige Rolle in der Hautabwehr gegen oxidativen Stress ausüben, durch ihr Eingreifen in die Lipidbiosynthese der Epidermis und das Vorlegen von Lipiden für die Barrierebildung der Epidermis. Bevorzugte essentielle Fettsäuren sind ausgewählt aus der Gruppe umfassend Linolsäure, Gamma-Linolensäure, Homo-Gamma-Linolensäure, Columbinsäure, Eicosan-(6,9,13)-triensäure, Arachidonsäure, Gamma-Linolensäure, Timnodonsäure, Hexaensäure und deren Gemischen.
- Die kosmetischen Zusammensetzungen der Erfindung können im weiteren Substanzen umfassen, die als Verdünnungsmittel, Dispergiermittel oder Träger für CLA wirken, welche den Zusammensetzungen gemäss gut bekannten Techniken zugefügt werden, in jedem geeigneten, dem Fachmann gut bekannten Anteil, zum Beispiel in einem Bereich von etwa 30 Gew.% bis etwa 98 Gew.%, vorzugsweise von etwa 50 Gew.% bis 98 Gew.%, berechnet auf das Gesamtgewicht der Zusammensetzung.
- Ein Öl oder öliges Material kann mit Wasser zusammen mit einem Emulgator (bekannt als „oberflächenaktive Verbindung") enthalten sein, um entweder w/o oder o/w Emulsionen herzustellen, weithin abhängig von dem durchschnittlichen hydrophilenlipophilen Gleichgewicht (HLB) des Emulgators. Oberflächenaktive Stoffe können in jedem geeigneten Anteil, der dem Fachmann gut bekannt ist, eingearbeitet werden, zum Beispiel von etwa 0.5 Gew.% bis etwa 30 Gew.%, vorzugsweise von etwa 1 Gew.% bis etwa 15 Gew.%.
- Kationische, nicht-ionische, anionische oder amphotere oberflächenaktive Verbindungen und deren Kombinationen können ebenfalls verwendet werden. Nichtionische oberflächenaktive Verbindungen können alkoxylierte Verbindungen umfassen, basierend auf Fettalkoholen, Fettsäuren und Sorbitan, Copolymere von Polyoxypropylen-Polyoxyethylen und Alkylpolyglycosiden. Anionische oberflächenaktive Verbindungen können Fettsäureseifen Natriumlaurylsulphat, Natriumlaurylethersulphat, Alkylbenzensulphonat, Mono- und/oder Dialkylphosphate und Ähnliche umfassen. Amphotere oberflächenaktive Verbindungen umfassen Dialkylaminoxide, verschiedene Arten von Betainen und natürlichen Phospholipiden.
- In einer wasserbasierten kosmetischen Zusammensetzung kann auch ein Verdickungsmittel enthalten sein in jedem geeigneten, dem Fachmann gut bekannten Anteil, zum Beispiel von 0.1 Gew.% bis 10 Gew.%, vorzugsweise von etwa 0.5 Gew.% bis 5 Gew.%. Beispiele von Verdickungsmitteln sind quer vernetzte Polyacrylatverbindungen (Carbopol®) und Gummis wie Xanthan, Carrageenan, Gelatine, Karaya, Pektin und Johannisbrotgummi. Die besagte wasserbasierte kosmetische Zusammensetzung kann mit Konservierungsmitteln gegen das Wachstum von Mikroorganismen geschützt werden. Geeignete Konservierungsmittel umfassen Alkylester von p-Hydroxybenzoesäure, Hydantoinderivate, Propionatsalze, Methylparaben, Propylparaben, Imidazolidinyl Harnstoff, Natrium-Dehydroxyacetat-Benzylalkohol und eine Vielzahl von quaternären Ammoniumverbindungen. Konservierungsmittel können in jedem geeigneten, dem Fachmann gut bekannten Anteil zugefügt werden, zum Beispiel in einem Anteil im Bereich von etwa 0.2 Gew.% bis 1 Gew.%.
- In einer flüssigen nicht-wässrigen kosmetischen Zusammensetzung können ebenfalls Siliconpolymere in jedem geeigneten, dem Fachmann gut bekannten Anteil enthalten sein, zum Beispiel in Mengen im Bereich von 5 Gew.% bis 95 Gew.%.
- Weitere Bestandteile, die in der kosmetischen Zusammensetzung gemäss der vorliegenden Erfindung enthalten sein können, sind Weichmacher. Unter gewissen Umständen können Weichmacher eine doppelte Funktion haben, das Wirken sowohl als Träger, um die Dispersion der CLA als aktiven Bestandteil zu erleichtern und als Hautweichmacher. Weichmacher können in die kosmetische Zusammensetzung der Erfindung eingearbeitet werden in jedem geeigneten, dem Fachmann gut bekannten Anteil, zum Beispiel in einem Bereich von etwa 0.5% bis etwa 50%. Geeignete Weichmacher können unter solchen allgemeinen chemischen Kategorien als Ester, Fettsäuren und Alkohole, Polyole und Kohlenwasserstoffe klassiert werden. Geeignete Fettsäure-Di-Ester umfassen Dibutyladipat, Diethylsebacat, Diisopropyldimerat, Propylenglycol-Myristylether-Acetat, Diisopropyladipat und Dioctylsuccinat. Geeignete verzweigtkettige Fettsäureester umfassen 2-Ethylhexylmyristat, Isopropylstearat und Isostearylpalmitat. Geeignete tribasische Säureester umfassen Triisopropyl Trilinoleat, Frilouryl-Citrat, Tributirrin und gesättigte oder ungesättigte pflanzliche Öle. Geeignete geradkettige Fettsäureester umfassen Laurylpalmitat, Myristyllactat, Oleylerucat, Stearyloleat Cococaprylat/Caprat und Cetyloctanoat. Geeignete Fettalkohole und -säuren sind C10-C20 – Verbindungen wie beispielsweise Cetyl-, Myristyl-, Palmitin- und Stearylalkohole und -säuren. Geeignete Polyole sind lineare und verzweigtkettige Alkylpolyhydroxyl – Verbindungen, wie beispielsweise Propylen- und Butylenglycol, Sorbitol, Glycerin, sowie auch polymere Polyole wie beispielsweise Polypropylenglycol und Polyethylenglycol. Geeignete Kohlenwasserstoffe sind lineare C12-C30 Kohlenwasserstoffketten, wie beispielsweise Mineralöl, Petroleumgel, Squalen und Isoparaffine.
- Sonnenschutzmittel können ebenfalls in die erfindungsgemässen kosmetischen Zusammensetzung eingearbeitet werden. Erläuternde Verbindungen sind die Derivate von PABA, Cinnamat und Benzophenon, wie beispielsweise Octylmethoxycinnamat, 2-Hydroxy-4-Methoxy-Benzophenon. Der Anteil des verwendeten Sonnenschutzmittels hängt von dem erwünschten Schutzgrad gegenüber UV Strahlung ab.
- Andere geringfügige Bestandteile können ebenfalls der erfindungsgemässen kosmetischen Zusammensetzung zugefügt werden, umfassend Färbemittel, Trübungsmittel und Parfüme, jedes jeweils gegebenenfalls enthalten in geeigneten Anteilen, zum Beispiel von 0.001 Gew.% bis zu 20 Gew.%, berechnet auf das Gewicht der Zusammensetzung.
- Die topische Zusammensetzung für die Hautbehandlung gemäss der Erfindung kann als Lotio, flüssige Creme, Creme oder Gel formuliert sein. Die Zusammensetzung kann in einem geeigneten Behälter verpackt sein, entsprechend ihrer Viskosität und der beabsichtigten Verwendung durch den Verwender. Zum Beispiel, eine Lotio oder flüssige Creme kann verpackt werden in einer Flasche, in einem Rollenapplikator, in einer Kapsel, in einer mit einem Sprühmittel angetriebener Aerosolvorrichtung oder einem mit einer Pumpe versehenen für die manuelle Handhabung geeigneten Behälter. Wenn die Zusammensetzung eine Creme ist, kann sie einfach in einer unverformbaren Flasche oder in einem Drückbehälter, wie beispielsweise eine Tube oder ein mit einem Deckel versehenes Gefäss, aufbewahrt werden.
- Gemäss einem anderen Aspekt betrifft die vorliegende Erfindung ein Set für die topische Verabreichung von CLA.
- Das besagte Set umfasst (a) Zusammensetzungen enthaltend CLA in Form von Einzeldosierungen, gegebenenfalls mit Beimischung von geeigneten üblichen Excipientien und Antioxidationsmittel, vorzugsweise in Form einer öligen Flüssigkeit und (b) eine Einzeldosierungsform enthaltend mindestens einen hydrophilen Anti-Zellulite-Wirkstoff, gegebenenfalls mit Beimischung von geeigneten üblichen Excipientien und alpha-Hydroxysäure, in einer wässrigen oder wässerig-alkoholischen Lösung.
- Der Verpackungsbehälter des Sets enthält im Weiteren ein Merkblatt, welches die Anweisung gibt, erstens die wässrige Lösung (b) für die wirksame Absorbierung des hydrophilen Bestandteils und zweitens die ölige Flüssigkeit (a) anzuwenden.
- Eine der Vorteile des Sets ist, dass das Eindringen des hydrophilen Anti-Zellulite-Wirkstoffs einfacher ist unter Abwesenheit der öligen Phase, welche anschliessend auf die Haut aufgetragen wird.
- Die folgenden Beispiele zeigen im Detail, wie die vorliegende Erfindung ausgeführt werden kann, ohne diese zu beschränken.
- Herstellungsbeispiel 1 – Synthese von CLA durch alkalische Isomerisation von Traubenkernöl in Glycerin (Die folgende Synthese ist Gegenstand einer parallelen Patentanmeldung).
- 1 kg Glycerin, 235 g Kaliumhydroxid (KOH) und 1000 g Traubenkernöl wurden in einen 4-Hals-Rundglaskolben (5000 ml) gegeben, der mit einem mechanischen Rührer, einem Thermometer, einem Rückflusskühler und einem Stickstoffeinlass ausgestattet ist, wobei der Stickstoff im ersten Durchgang durch zwei Sauerstoffklappen eingeführt wird.
- Stickstoff wurde während 20 Minuten in die Reaktionsmischung gesprudelt, und die Temperatur wurde dann auf 90–100°C erhöht und während etwa 20 Minuten unter mechanischem Rühren gehalten, um das Triglycerid in die entsprechenden Kaliumsalze umzuwandeln. Das Doppelphasen-System verschwindet, um eine Glycerin-Seifensuspension zu formen, die anschliessend auf 230°C in einer inerten Atmosphere erhitzt und während 4 Stunden gerührt wird.
- Die Reaktionsmischung wurde auf etwa 100°C abgekühlt und das Rühren beendet, im Moment wo die Reaktionsmischung dazu neigt, beim Abkühlen eine sehr hohe Viskosität zu erreichen. 2 Liter Wasser wurden dann langsam zugefügt und die Mischung während 2 Stunden bei 95°C gehalten. Dieser Arbeitsvorgang ist nötig, da die vernachlässigbare Anwesenheit von Wasser und der hohe Gehalt an Glycerin verursachen, dass Fettsäuren als Mono- und als Diglycerid von 5 Gew.% bis 10 Gew.%, berechnet auf das Gesamtgewicht des Lipidgehalts, anwesend sind. Da partielle Glyceridester dazu neigen W/O Emulsion zu formen, verschafft die Wasserzugabe und das Wiedererhitzen die vollständige Verseifung der verbleibenden veresterten Fettsäure.
- Die Mischung wurde in einen Becher übertragen, dann auf Raumtemperatur abgekühlt und 50% w/v Schwefelsäure wurde der Mischung zugefügt, dieselbe wurde während einer Stunde gerührt, bis der pH-Wert sich bei etwa 3 stabilisierte.
- Die angesäuerte Ölphase bildete eine niedrigere Hydroglycerinschicht und eine CLA enthaltende obere Fettsäure-Ölschicht, welche durch Dekantierung abgetrennt wurde. Es ist zu bemerken, dass die Abtrennung in einem industriellen Arbeitsvorgang durch Zentrifugieren ausgeführt werden kann.
- Die CLA wurde mit Wasser gewaschen und schliesslich mit Natriumsulphat getrocknet und gefiltert, dann wurde sie in einer dunklen Flasche bei 4°C bis zur Verwendungszeit gelagert. Die Gesamtausbeute ergab etwa 770 g eines gelben Öls, dessen GC-Analyse in Tabelle 1 gezeigt ist.
- Die Zusammensetzung von CLA scheint eine komplexe Mischung zu sein, d.h. enthaltend 9c, 11t- und 8c, 10t- Octadecadiensäuren in einem Anteil von 30,90 %, 11c, 13t-10t, 12c-Octadecadiensäuren in einem Anteil von 32,05 %, 11t, 13c-8c, 10c-9c, 11c-Octadecadiensäure in einem Anteil von 1,55 % und 10c, 12c-11c, 13c-11t, 13t, 9t, 11t-10t, 12t-8t, 10t-Octadecadiensäuren, die den übrigen Teil bilden.
- Vergleichsbeispiel 1 und Anwendungsbeispiele 1,2 – Körpercremen
- Drei verschiedene O/W Emulsionen wurden unter Rühren zubereitet, durch Turbomischen der öligen Phase und der wässrigen Phase, jeweils separat vorerhitzt bei 75°C; die Zusammensetzung ist im Folgenden gezeigt:
- Wie bemerkt werden kann, enthält die topische Formulierung kein CLA, kein CLA alleine, respektive kein CLA mit Koffein.
- Anwendunasbeispiel 3 – Klinische Prüfung der antizellulitischen Aktivität bei topischer Anwendung von CLA und CLA mit einem Xantin
- 9 weibliche Testpersonen wurden ausgewählt, basierend auf deren Zellulite-Intensität im Schenkelbereich, die eine bilaterale Symmetrie hat. Testpersonen mit Zellulite des ersten und zweiten Grades wurden ausgewählt, wobei eine 5-Punkte Gradierungsskala verwendet wurde, um die Stärke der Zellulite von jeder Testperson zu bewerten. Die Skala ging von 0 bis 4, wobei 0 = keine Zellulite; 1 = kleine Erhöhungen oder Vertiefungen; 2 = Dehnungssteifen und Erhöhungen; 3 = Betonte Zusammenballung der Haut und Dehnungsstreifen; 4 = alles obige und zusätzlich harte unterflächige Knötchen.
- Die Testpersonen wurden in 3 Gruppen von jeweils 3 Personen aufgeteilt und angewiesen, die einen auf dem rechten Schenkel die Zusammensetzungen des Vergleichsbeispiels 3, die einen die Zusammensetzungen des Anwendungsbeispiels 4, respektive die einen die Zusammensetzungen des Anwendungsbeispiels 5 aufzubringen.
- Die Testpersonen wurden angewiesen, die Anwendung zweimal täglich, morgens und abends, während 2 Monaten auszuführen. Nachher wurde der Zustand der Zellulite gemäss Smith WP (Cosmetics & Toiletries, 61–70, Juni 1995) gemessen, mit Vergleich des rechten Schenkels zum linken Schenkel. Die Resultate sind in der Tabelle 2 angegeben.
- Die obigen Resultate zeigen, dass die CLA enthaltende Zusammensetzung den Zustand von Zellulite wirksam verbessert, mit einer zusätzlichen Verbesserung durch die Kombination mit Koffein.
- Anwendungsbeispiel 4 – Set für die Behandlung von Zellulite
- Eine ölige Mischung (a) und eine wässerig-alkoholische Lösung (b) wurden separat vorbereitet durch Vermischen der folgenden Bestandteile:
- Die beiden Zusammensetzungen wurden separat in 25 ml Flaschen abgefüllt und in der gleichen Set-Verpackung kombiniert, zusammen mit der Anweisung, zuerst (b) und 10 Minuten später (a) anzuwenden.
- Zu bemerken ist, dass die spezifischen Formen der Erfindung, die hierin erklärt und beschrieben sind, nur beispielhaft sein sollen. Veränderungen, umfassend diejenige, die in der vorliegenden Beschreibung vorgeschlagen sind, ohne sich jedoch auf diese zu beschränken, können in den erklärten Ausführungsformen vorgenommen werden ohne sich von der klaren Lehre der Offenbarung zu entfernen. Entsprechend, um den gesamten Umfang der Erfindung festzulegen, sind die folgenden angefügten Ansprüche miteinzubeziehen.
Claims (5)
- Verwendung einer Zubereitung enthaltend 2 bis 70 Gew.% konjugierter Linolsäure (CLA, vom Englischen « Conjugated Linoleic Acid ») in Form freier Säure oder deren Salze, gewählt aus der Gruppe bestehend aus Metallseifen mit alkalischen und/oder erdalkalischen Metallionen, und Ammoniaksalzen sowie Mono-, Di- oder Triethanolamin zur Herstellung einer kosmetischen oder dermatologischen Zubereitung für die topische Anwendung zur Behandlung oder Verhütung von Fettdepots und Zellulite.
- Verwendung nach Patentanspruch 1 dadurch gekennzeichnet, dass die CLA zugleich 9cis-, 11trans- und 10trans-, 12cis-Isomere beinhaltet.
- Verwendung nach Patentanspruch 1 oder 2 dadurch gekennzeichnet, dass die Zubereitung einen oder mehrere Anti-Zellulite-Wirkstoffe, gewählt aus der Gruppe bestehend aus Xanthin, Koffein, Theophyllin, Theobromin, Aminophyllin und deren Gemischen, enthält.
- Verwendung nach einem der vorherigen Patentansprüche dadurch gekennzeichnet, dass die Zubereitung eine Vanadium-Verbindung enthält.
- Verwendung nach einem der vorherigen Patentansprüche dadurch gekennzeichnet, dass die Zubereitung in Form einer Creme, eines Ges, einer Lotion, eines Öls oder eines Sprays vorliegt.
