Abstract

Die Erfindung betriff die Messung der Konzentration von Analyten mit Hilfe einer Oxidase in einem Tauchsensor, der sich in einer Flüssigkeit oder flüssigkeitshaltigen Matrix befindet. Sauerstoff diffundiert in die Enzymschicht von innen aus einem gasgefüllten Raum, der mit der Atmosphäre bzw. einem Sauerstoffreservoir in Verbindung steht. Dies ermöglicht Sauerstoffsättigung der Oxidase in einem sauerstoffarmen oder sauerstofffreien Medium bzw. bei hoher Analytkonzentration. Die Diffusion der Analyten in die Enzymschicht erfolgt in einem diffusionslimitierenden wasserhaltigen Kanal oder mehreren solcher Kanäle, wobei der Kanal / die Kanäle an der Sensoroberfläche mit einer für Proteine undurchlässigen hydrophilen Matrix gefüllt ist / sind. Durch Vergrösserung des Kanalquerschnittes an der Sensoroberfläche und / oder durch Verbindung des Kanals / der Kanäle mit einer für Proteine undurchlässigen porösen hydrophilen Schicht an der Sensoroberfläche wird die Wirkung äusserer Ablagerungen auf den Diffusionswiderstand für den Analyten reduziert.
