—方、 凝集阻害法は、 予めリガンド又はリガンド様物質を感作した担体粒 子とレセプタ一による凝集が測定対象であるリガンドによって阻止される割 合を測定する方法であり、 凝集法では特殊なリガンドを除き 2種類以上の特 異的レセプターが必須であるのに対して、 凝集阻害法は 1種類の特異的レセ プタ一があれば測定系を構築できることが大きな特徴である。
さらに、 免疫反応に関していえば、 凝集阻害法には地帯現象 (抗原過剰に よる見かけ上の反応低下現象) が起こらないという利点があるものの、 現在 までの適用は、 ハプテンのような低分子リガンドすなわち 2種類以上のレセ プターが利用できないリガンドを測定対象としているケースがほとんどであ る。 [0003] 凝集阻害法の適用が限定されている理由として、 凝集法と比較して測定濃 度範囲の調整が難しいという点が挙げられる。 凝集法においては、 リガンド 量に比例して凝集が進行するため、 例えばラテックス免疫凝集法に関する特 許文献 1に示されるように、 高濃度域の感度上昇を増強するだけで容易に測 定範囲を拡大できる。
一方、 リガンド量に比例して凝集阻止反応が進行する凝集阻害法の測定範 囲に関しては、 リガンド高濃度域の測定上限値は、 基本的には初期凝集 (リ ガンド量ゼロ時の凝集) の大きさに依存し、 リガンド低濃度域の測定下限値 は、 凝集阻止反応の感受性に依存するため、 二つの要因を複合的に考慮する 必要がある。
[0004] 凝集阻害法の初期凝集を増強する方法として、 例えば、 特許文献 2に記載 の方法では、 ハプテンの測定において非特異的な凝集促進物質を利用する方 法が提示されている。 本方法により高濃度域までリガンドの測定が可能とな り測定上限値を拡張できるものの、 リガンドとレセプター間の特異反応に関 係のない凝集増強であるため凝集阻止反応に対する感受性が低下し、 結果と して低濃度域の検出能は著しく悪化する。
[0005] また、 特許文献 3には単一のモノクローナル抗体感作担体と抗原感作担体 を利用する凝集阻害法が示されている。 本方法ではリガンドとレセプタ一を それぞれ担持した担体粒子を利用して、 光学的に感度の変化量を増強するこ とで、 凝集阻止反応に対する感受性を損なわずに低濃度域の検出能を向上し ている。 しかしながら、 該方法は抗原と抗体間の反応自体が増強されるわけ ではないため、 測定範囲を拡張するような効果は認められない。 同じく特許 文献 4に提案される抗原感作ラテックス、 抗体及び抗体に特異的なレセプタ 一を利用する凝集阻害法は、 抗原と抗体、 抗体とレセプターの特異反応のみ で凝集を増強しているため、 従来法と比較すると測定範囲の拡張が達成され る。 しかしながら実施例でレセプタ一として示されているプロテイン Aは抗 体 ( I g G ) 全般に反応性を有するため、 I g Gを含有する血清や血漿とい つた試料の測定には不向きであり、 抗マウス I g Gラットモノクロ一ナル抗 体に関しても、 一般的に利用される素材ではないため入手が困難なケースも 考えられる。 また測定原理自体が抗原-抗体、 抗体-レセプターという二相性 の反応を介しているため、 測定機器、 方法によっては性能が不良となる可能 性も考えられる。
[0006] 上記に示したように、 初期凝集は強く、 同時に凝集阻止反応への感受性も 高いという相反する特性を兼ね備えた凝集阻害測定法及び凝集阻害測定用試 薬は未だ確立されておらず、 これまでの測定法では通常のリガンド、 例えば 2箇所以上のレセプタ一結合部位を有するようなリガンドを高感度かつ広範 囲に再現性良く測定することは困難な状況にあった。

発明が解決しょうとする課題
[0007] 本発明は、 上記従来技術の問題点を解決するためのものであって、 試料中の リガンドを低濃度域から高濃度域まで高感度かつ広範囲に測定できると同時 に、 良好な測定再現性能を有する凝集阻害測定法及び凝集阻害測定用試薬の 提供を課題とする。
課題を解決するための手段
[0008] 本発明者らは、 凝集阻害測定法について鋭意検討を行ったところ、 リガン ドを担持した不溶性担体粒子と遊離状の特異的レセプターに加え、 リガンド 上の結合部位が前記遊離状レセプタ一とは異なる特異的レセプターを担持さ せた不溶性担体粒子を用いることで、 凝集阻止反応への感受性を保持したま ま初期凝集を増強でき、 リガンド低濃度域から高濃度域まで予想外に高感度 かつ広範囲に測定でき、 良好な測定再現性能を有することを見出し、 本発明 を完成するに至った。
すなわち、 本発明は以下の構成を有する。 ( 1 ) 二箇所以上のレセプター結合部位を有する測定対象リガンドを含む試 料と、 下記 (A) 〜 (C) 、
(A) 予めリガンド又はリガンド様物質を担持した不溶性担体粒子
(B) 前記リガンドに特異的に反応する遊離状レセプター
(C) 前記リガンドに特異的に反応し、 リガンド上の結合部位が前記遊離状 レセプターとは異なるレセプターを担持した不溶性担体粒子
とを混合し、 試料中の測定対象リガンド量に応じた粒子の凝集阻止反応を測 定することを特徴とする凝集阻害測定法。
(2) 不溶性担体粒子がラテックス粒子である上記 (1 ) に記載の凝集阻害 測定法。
(3) レセプターが抗体又はそれらの機能性部位を含む断片であり、 リガン ドが抗原であり、 レセプターとリガンドとの免疫反応に基づき、 粒子の凝集 阻止反応を測定する上記 (1 ) 又は (2) に記載の凝集阻害測定法。
(4) 遊離状レセプター及び不溶性担体粒子に担持されたレセプターが、 い ずれもモノクローナル抗体である上記 (1 ) 〜 (3) のいずれかに記載の凝 集阻害測定法。
(5) 測定対象リガンドが、 ヒトアルブミン、 レセプターが、 抗ヒトアルブ ミンモノクローナル抗体である上記 (1 ) 〜 (4) のいずれかに記載の凝集 阻害測定法。
( 6 ) レセプターを担持する不溶性担体粒子の平均粒子径が、 リガンド又は リガンド様物質を担持する不溶性担体粒子の平均粒子径の 1 / 2〜 1 / 1 0 である上記 (1 ) 〜 (5) のいずれかに記載の凝集阻害測定法。
(7) 下記 (A) 〜 (C) を含み、 試料中の測定対象リガンド量に応じた粒 子の凝集阻止反応を測定することを特徴とする凝集阻害測定用試薬。

[0009] 本発明の凝集阻害測定法及び凝集阻害測定用試薬は、 測定対象リガンドを 低濃度域から高濃度域まで高感度かつ広範囲に測定できると同時に、 良好な 測定再現性能を有する。 特に、 免疫反応を利用する場合、 地帯現象を発生し ないという特徴から異常高値を呈する可能性のあるリガンド測定時には粒子 凝集法よりも測定の信頼性が高い。 また、 本発明のレセプターにモノクロ一 ナル抗体を利用した場合には、 リガンドが全長でなくてもェピトープが残存 していれば凝集阻止反応が起こるため、 断片化したリガンドも全長のリガン ドと同様に測定できる可能性がある。
発明を実施するための最良の形態
[0010] (測定対象試料)
本発明の測定対象となるリガンドを含む試料としては、 例えばヒト又は動 物の血液、 血清、 血漿、 培養上清、 尿、 髄液、 唾液、 汗、 腹水、 又は細胞あ るいは組織の抽出液等が挙げられる。
[0011] (リガンド)
本発明の測定対象となるリガンドとしては、 二箇所以上のレセプター結合 部位を有する物質であれば、 いずれも対象とすることができる。 具体的には
、 C反応性タンパク (CRP) 、 FDP、 D—ダイマ一、 前立腺特異抗原 ( PSA) 、 ヘモグロビン A 1 C、 アルブミン、 ぺプシノ一ゲン I (PG I ) 、 ぺプシノ一ゲン I I (PG I I ) 、 マトリックスメタ口プロティナ一ゼ (M MP) 、 トリプシン、 キモトリブシン、 ェラスタ一ゼ、 カテブシン等のタン パク質、 その他、 ペプチド、 糖類、 核酸、 脂質、 高分子薬剤などが挙げられ る。 理論的には二箇所以上のレセプター結合部位を有するリガンド(リガンド が抗原であれば二箇所以上のェピトープ (抗原決定基) を有するリガンド、 例えば二種類以上のモノク口一ナル抗体を利用したサンドィツチ測定で検出 可能なリガンド)は全て測定対象物質として適用可能である。
リガンド様物質とは、 レセプターとの関係において前記リガンドと同等、 同質の反応性 (リガンドが抗原であれば抗原性) を有するものをいう。 例え ば、 リガンド (抗原など) を分解して得られる断片、 遺伝子操作で得られた 組み替えリガンド、 リガンド構造が類似した物質 (リガンド類縁物質) 等が あげられる。
[0012] (レセプター)
本発明で用いるレセプターとしては、 測定対象であるリガンドに特異的に 結合する物質を用いる。 一般的にはゥサギ、 ヒッジ、 ャギ等に、 測定対象の リガンドを免疫して得られる抗リガンドボリク口ーナル抗体ゃ抗リガンドモ ノクロ一ナル抗体を用いることができる。 その中でも特異性の点からはモノ クローナル抗体の使用が特に望ましい。 用いる抗体は、 常法に従い調製した 抗体のフラグメントであってもよい。 その他、 リガンドの種類によってはレ クチン、 核酸塩基等も抗体と併用してあるいは単独でレセプターとして用い ることができる。 [0013] 本発明で用いるレセプターの種類は、 2種類以上であれば特に制限はなく 、 遊離状レセプターと担体に固定化されるレセプターとは異なるものを用い ることを要する。 すなわち、 遊離状レセプターと担体固定化レセプターとは 、 リガンド上の結合部位が異なるレセプターであることを要する。 すなわち 、 各レセプターがリガンドとの結合を相互に干渉しないような関係にあるレ セプターであればよい。 また、 遊離状レセプター及び担体固定化レセプター はともにモノクローナル抗体であることが望ましい。 本発明では、 遊離状レ セプターと固定化担体レセプタ一の両者を組み合わせて用いることにより、 初期凝集を促進し、 かつ、 高濃度域での感度変化をも増大させることができ 、 測定可能な範囲を拡張することができる。 レセプターの種類や含量、 存在 様式 (遊離状態あるいは担体担持状態) はリガンドのレセプター結合部位の 数や分析に必要な粒子の凝集度合い、 目的とする測定範囲などを考慮して適 宜選択することができる。
レセプターとしてモノクローナル抗体を用いる場合、 それらの選定は、 例 えば次のような手順で行われる。
まず、 測定対象リガンドを免疫源として調製したモノクローナル抗体とリ ガンドとの反応性をウェスタンブロット法ゃ E L I S A法等で確認し、 反応 性が強い抗体を 2種類以上選択する。 さらに、 選択されたこれらの抗体を組 み合わせたときの反応性をサンドイッチ E L I S A法で確認する。 このとき 、 反応性が強ければそれだけ、 感度が高く測定範囲を拡張できる可能性が高 いため、 モノクローナル抗体の組み合わせは、 測定対象、 要求性能に応じて 適宜選択することが望ましい。 このようにして得られたモノクローナル抗体 の組み合わせを、 一方を担体に固定化させた状態で、 もう一方を担体に固定 化しない遊離の状態で、 実際の測定系に供し、 初期凝集、 測定感度、 測定範 囲、 リガンドと類似した構造を有する測定対象外の物質との交差反応性等を 考慮していずれの組み合わせが適当か最終的な判断を行う。
[0014] (担体粒子)
本発明で使用するリガンドあるいはレセプターを担持させる不溶性担体粒 子は、 特に限定されないが平均粒子径 5 0〜 1 0 0 0 n mのラテックスが好 ましい。 ラテックスの材質としては、 リガンドあるいはレセプターの担持に 適したものなら良く、 一般的に用いられるポリスチレンを主成分とするラテ ックスの他に、 スチレン一ブタジエン共重合体、 (メタ) アクリル酸エステ ル類ポリマーなどが挙げられる。 また、 金属コロイ ド、 ゼラチン、 リポソ一 ム、 マイクロカプセル、 シリカ、 アルミナ、 力一ポンプラック、 金属化合物 、 金属、 セラミックス又は磁性体等の材質よりなる粒子を使用することもで きる。 担体粒子に前記リガンド又はリガンド様物質及びレセプターを担持す る方法としては、 一般的に使用されている物理吸着法のほか化学結合法も採 用できる。
[0015] また、 本発明で用いるリガンド及びレセプターをそれぞれ担持する担体粒 子は、 その材質が同一でも異なっていてもよい。 また、 それぞれの担体粒子 の粒子径は分析方法や目的に適した粒子径を選択することができるが、 本発 明の効果を充分発揮するには、 レセプターを担持する担体粒子の平均粒子径 は、 リガンドを担持する担体粒子の平均粒子径の 1 / 2から 1 / 1 0である ことが好ましい。
[001 6] (緩衝液)
本発明における凝集反応は、 緩衝液中で行われ、 その緩衝液は、 凝集反応 が最適に行われる種類、 濃度、 p Hが選択され、 リン酸緩衝液、 トリス塩酸 緩衝液、 炭酸緩衝液、 グリシン緩衝液、 グッドの緩衝液等を用いる事ができ る。 その緩衝液の緩衝剤の濃度は、 5 m M〜5 0 O m M程度で用いられ、 p Hは中性域から塩基性域で用いられることが多く、 通常 7 . 0〜9 . 5の範 囲で用いられる。
[001 7] (凝集シグナルの測定方法)
凝集シグナルの測定は、 通常凝集阻止反応測定に用いられる方法であれば いずれでもよく、 吸光度比による評価、 粒子数測定、 粒子サイズ測定 (凝集 すればサイズは大きくなる) 、 散乱光測定や吸収スペク トルの測定 (凝集す れば増大もしくはシフトする) など当業者が用いうる手段があげられる。 さ らに、 光学的な検出を可能な範囲で電気化学的な検出によって置き換えるこ とも可能である。
[0018] 凝集シグナルの測定は、 前記の如く種々の方法があるが、 ラテックス粒子 と汎用の生化学分析装置を用いる方法が便利である。 例えば、 測定対象であ るリガンドを含む試料に、 遊離状のレセプターと別種のレセプターを担持し たラテックス、 リガンドを担持したラテックス等の不溶性担体粒子を含む試 薬を加え、 一定温度で一定時間加温し、 この間の吸光度を測定し、 吸光度の 変化量を検出し、 予め濃度の判っている標準液を試料とした場合の検量線か ら被検試料中のリガンドの濃度を算出することができる。 ラテックス凝集阻 害法では、 通常 5 0 0〜9 0 0 n mの波長の吸光度が用いられ、 反応時の吸 光度の変化量を定量に用いるのが一般的である。 本発明を利用した測定範囲 は、 測定対象とするリガンドの種類、 レセプターのアビディティ一、 各レセ プターの量比などを考慮して適宜所望の測定範囲に設定できる。 例えば、 測 定対象が尿中アルブミンの場合、 臨床検査での使用を考慮すると 1 g /m Lから 1 m g /m Lの範囲が好適であり、 後述する実施例のように、 本発明 により正確に測定することができる。
[001 9] 本発明の凝集阻害測定用試薬は、 (A ) 予めリガンド又はリガンド様物質 を担持した不溶性担体粒子、 ( B ) 前記リガンドに特異的に反応する遊離状 レセプター、 (C ) 前記リガンドに特異的に反応し、 リガンド上の結合部位 が前記遊離状レセプターとは異なるレセプターを担持した不溶性担体粒子を 含み、 (B ) 、 ( C ) を含む第一試薬と (A ) を含む第二試薬とに分けるこ とが望ましい。
以下、 実施例を挙げて本発明を更に詳細に説明するが、 本発明はこれらに限 定されるものではない。
実施例
[0020] 凝集阻害測定法による微量アルブミンの測定
( 1 ) レセプタ一 (抗アルブミンモノクローナル抗体) の調製
精製ヒトアルブミン (シグマ社製) 1 0 0 gを 1回の免疫に使用した。 初 回免疫は、 アルブミンとフロインドの完全アジュバンドを等量混合して調製 したェマルジヨン 200 U Lを用い、 これを BA L B/cマウスの腹腔に注 射して行った。 追加免疫は、 フロインドの不完全アジュバンドを使用して前 記と同様に調製したェマルジヨン 200 U Lを用い、 2週間の間隔で 3回、 腹腔注射を繰り返した。
[0021] マウス眼底静脈より採血した血液中の抗体価を、 精製ヒトアルブミンを固 相化した E L I S A法にて測定し、 抗体価の高いマウスを選んで細胞融合に 供した (なお抗体価の確認方法は後述する培養上清中の抗アルブミン抗体の 存在確認方法と同様である) 。 4回目の免疫から 2週間後に、 アルブミン 1 00 /J gを生理食塩液 200 /J Lに溶解したものをマウス腹腔に注射し、 3 日後に脾臓を摘出した。 該摘出した脾臓を R PM I 1 640培地中でほぐし た後、 1 500 r p mで遠心分離して脾細胞を回収した。 これを牛胎児血清 フリーの R PM I 1 640培地で 3回以上遠心■沈殿による洗浄後、 沈殿部 に対して 1 5%牛胎児血清を含む R PM I 1 640培地 2 m Lを加えて懸濁 し、 脾細胞懸濁液とした。 脾細胞とミエローマ細胞 S P 2ZO— AG 1 4を 細胞数で 6対 1の割合で混合した後、 50 %ポリエチレングリコール存在下 で細胞融合させた。 1 500 r p mの遠心分離で沈殿部を集め、 G K N液 ( グルコース 2 g、 塩化カリウム 0. 4 g、 塩化ナトリウム 8 g、 リン酸水素 ニナトリウム 1 . 4 1 g及びリン酸二水素ナトリウム二水和物 0. 7 8 gを 精製水に溶かして 1 リットルとしたもの) に懸濁■遠心分離により洗浄後、 沈殿部を回収した。 該沈殿部を 1 5%牛胎児血清を含む R PM I 1 640培 地 3 Om Lに懸濁したものを 1ゥエルあたり 1 00 L分注し、 同じゥエル に、 フィーダ一細胞として B A L B/cマウスの胸腺細胞を 2. 5 X 1 06個 /m L含む H A T培地を 1ゥエルあたり 200 L分注した。 9 6穴マイク 口プレート 3枚に分注し、 3 7 °Cにて 5 %炭酸ガス培養器中で培養した。
[0022] 培養上清中の抗アルブミン抗体の存在は、 精製ヒトアルブミンを固相化し た E L I S A法で確認した。 培養 1 0日後に全てのゥエルで融合細胞の増殖 を確認した。 抗アルブミン抗体の存在確認方法の詳細は以下である。 先ず、 1 0 μ g/m Lのアルブミン及び 1 50mM塩化ナトリウムを含む 1 0 mMリン酸緩衝液 ( p H 7. 2 ; 以下、 P B Sと略す) 1 00 Lを 9 6 穴マイクロプレートに分注し、 4°Cで 1晚放置した。 次にこれを 0. 05% T w e e n 20及び 1 %牛血清アルブミンを含む P B S 300 U Lで 3回洗 浄した後、 培養を行った 3枚の 9 6穴マイクロプレート各ゥヱルの培養上清 を 50 L/ゥエル加え室温で 1時間放置した。 その後、 0. 05%Tw e e n 20を含む P B Sで 3回洗浄の後、 ペルォキシダーゼ標識抗マウス抗体 (第一化学薬品社製) を 50 !_/ゥエル加え、 室温で 1時間放置した。 こ れを 0. 5%T w e e n 20を含む P B Sで 3回洗浄後、 0. 2 %オルトフ ェニレンジァミン及び 0. 02 %過酸化水素を含むクェン酸緩衝液 ( p H 5 ) 50 L/ゥヱルを加え、 室温で 1 5分間放置後、 4. 5 N硫酸 50 Ι_ /ゥエルを加えて反応を停止させ、 波長 49 2 n mにおける吸光度を測定し た。 測定の結果、 吸光度の高いゥエルを抗アルブミン抗体の存在する (即ち 、 抗アルブミン抗体産生融合細胞の存在する) ゥエル (陽性ゥエル) として 選択した。
単クローン化は限界希釈法で行った。 すなわちフィーダ一細胞として BA L B/cマウスの胸腺細胞を 1ゥヱルあたり 1 06個ずつ分注した 9 6穴マイ クロプレートに、 陽性ゥエル中の融合細胞を 1 0個 Zm Lとなるように希釈 したものを 0. 1 m Lずつ分注した。 培地は、 初回は H T培地を、 2回目以 降は 1 5%牛胎児血清を含む R PM I 1 640培地を用い、 3 7°Cにて 5% 炭酸ガス培養器中で 1 0日間培養した。 前記した、 精製ヒトアルブミンを固 相化した E L I S A法による陽性ゥエルの選択及び限界希釈法による単クロ ーン化操作を各 3回繰り返して 30種の抗アルブミンモノクローナル抗体産 生細胞を得た。 各細胞の約 1 05個をプリスタン前処理したマウス腹腔に投与 し、 生成した腹水をそれぞれ採取した。 採取した各腹水から遠心分離により 不溶物を除去し、 等量の飽和硫安液を加え、 撹拌しながら 1晚放置後、 遠心 分離で沈殿を回収した。 回収した沈殿を 20 mMトリス緩衝液 ( p H 8 ) に 溶解し、 同緩衝液で透析した。 透析内容物それぞれを同緩衝液で平衡化した D E A E—セファロ一スカラムに別個に吸着させた後、 それぞれ同緩衝液中 の塩化ナトリウム 0〜30 OmMの濃度勾配で溶出させて得た I g G画分を 50 m Mグリシン緩衝液で透析し得られた 30種の精製抗体のうち、 サンド イッチ法で最も高感度が得られる 2種類の抗体の組み合わせ (抗体 1 一 8と 抗体 3_8) を選択した。
[0024] (2) アルブミン測定用ラテックス試薬の調製
選択した 2種類の抗アルブミンモノクロ一ナル抗体のうち、 抗体 3 _ 8を 2. 8 m g/m L含む 2 OmMトリス緩衝液 ( p H 8. 5) 3m Lに、 平均 粒子径 50 n mの 4%ラテックス (積水化学工業社製) 懸濁液 3m Lを加え 、 4 °Cにて 2時間撹拌した。 これに 0. 4%牛血清アルブミンを含む 2 Om Mトリス緩衝液 (p H 8. 5) 6m Lを加え、 4 °Cで 1時間撹拌した。 遠心 分離後、 沈殿を 5mM MO P S緩衝液 (p H 7. 0) で、 波長 600 n mに おける吸光度が 4 OmODとなるように再懸濁し、 抗アルブミン抗体 3— 8 感作ラテックスを調製した。 遊離状態の抗体 1 _ 8と抗体 3 _ 8感作ラテツ クスを混合し第一試薬を調製した。
[0025] 次に、 ヒト血清アルブミン 0. 5 m g/m Lを含む 1 OmM CH ES緩衝液
( p H 5. 5) 3 m Lを用い、 平均粒径 300 n mの 0. 5%ラテックス (積 水化学工業社製) 懸濁液 3m Lを加え、 4°Cにて 1時間撹拌した。 遠心分離 後、 上清を除去し、 沈殿を 5mM MO P S緩衝液 (p H 7. 0) で、 波長 6 00 n mにおける吸光度が 2. 1 5 O Dとなるように再懸濁し、 アルブミン感 作ラテックス溶液として第二試薬を調製した。
[0026] (3) アルブミンの測定
汎用型の日立 7 1 70型自動分析装置を用いて各濃度のアルブミン試料を 測定し、 測定感度を確認した。 具体的には、 第一試薬 1 00 Lに各濃度の アルブミンを含有する試料液 3 Lをそれぞれ加えて撹拌後、 37°Cで 5分 間加温し、 さらに第二試薬 1 00 Lを添加して 37 °C、 5分間の主波長 5 70 n 副波長 800 n mにおける吸光度変化量を測定した。 まず、 アル ブミン濃度 0 g/m Lの際の初期凝集を表 1に示した。 さらに 5、 1 2. 5 、 25、 50、 1 00、 200、 400、 800 g Lのアルブミンを 含有する各試料測定時の初期凝集からの吸光度変化量を測定し、 アルブミン 濃度対一 (マイナス) 吸光度 (以下同様) の反応曲線として図 1に実線で示 した。 この反応曲線の形状に基づき 0、 5、 25、 1 00、 400、 800 U g/m Lの 6ポイントから検量線を作成し、 各濃度のアルブミン試料の吸 光度変化量をアルブミン濃度に換算した測定値を表 2に示した。

比較例として実施例と最終の抗体濃度が同一となるように下記 (A) 〜 ( E) の 5種類の第一試薬を調製した。
(A) 遊離状態の抗体 1 一 8を単独で含有する第一試薬
(B) 抗体 1 _8感作ラテックスを単独で含有する第一試薬
(C) 遊離状態の抗体 1 _8と抗体 1 _ 8感作ラテックスの両方を含有す る第一試
(D) 遊離状態の抗体 1 一 8と遊離状態の抗体 3— 8の両方を含有する第 —試
(E) 抗体 1 _8感作ラテックスと抗体 3— 8感作ラテックスの両方を含 有する第一試薬
上記 (A) 〜 (E) の各第一試薬と、 (2) で調製した第二試薬を組み合 わせ、 (3) の方法に従って測定を行った。 実施例と同様に初期凝集の結果 を表 1に、 アルブミン濃度対一吸光度の反応曲線を図 1に、 各比較例の反応曲 線の形状に基づき 0〜 800 U g/m Lの範囲から選択した至適な濃度ボイ ン卜から検量線を作成し、 各濃度のアルブミン試料の吸光度変化量をアルブ ミン濃度に換算した測定値を表 2に示した。

表 1に示したように、 実施例の初期凝集 (吸光度) に対して、 比較例 A、 B、 Cは約半分、 比較例 Dは 2/3程度の初期凝集しか得られておらず、 凝 集阻害法における基本的な測定性能が劣っていることが容易に推察された。 一方、 比較例 Eでは実施例よりも大きい初期凝集が得られた。 [0029] [表 1]

[0030] 図 1のアルブミン濃度対— (マイナス) 吸光度の反応曲線より、 実施例 (― 書—) では凝集阻止反応の感受性が高く、 アルブミン低濃度域 (5— 50 g/mL) で大きな感度 (一吸光度) 変化を示すと同時にアルブミン高濃度域 (1 00— 800 g/mL) でも明瞭な感度変化を示すことから、 低濃度 域から高濃度域まで広範囲に精度良くアルブミンを測定できることが確認さ れた。 一方、 比較例 A (-□-) 、 同 C (-Δ-) ではアルブミン低濃度域 でほとんど感度変化が認められないため、 低濃度域を正確に測定することが できない。 また、 比較例 D (- X -) はアルブミン濃度対一吸光度の反応曲 線の形状が実施例と近似しているものの、 初期凝集や感度変化自体が実施例 に対して小さく、 測定精度に課題がある。 比較例 B (_〇_) 、 同 E ( __ -) ではアルブミン低濃度域で大きな感度変化を示すが、 高濃度域では感度 変化がないため、 低濃度域から高濃度域までを広範囲に測定することができ ない。
[0031] 表 2に示したように、 実施例では低濃度から高濃度までほぼ理論値どおり の測定値が得られた。 比較例 A、 Cではアルブミン 0 g/mLを含む低濃 度域を正確に測定することができなかった。 また、 比較例 B、 Eでは高濃度 域を正確に測定することができなかった。 比較例 Dではいずれの濃度でも理 論値との乖離が大きく、 測定値全般の正確性に問題が認められた。

[0033] 上記実施例及び比較例について、 同条件で同時再現性試験を行った (η = 5) 。 結果を表 3に示す。
表 3に示したように、 実施例では低濃度から高濃度まで再現性良く、 変動 係数 6%以下の値が得られ、 測定平均値もアルブミン濃度レベルと同等の測 定値が得られた。 比較例 A、 C、 Dでは再現性が悪く、 特に低濃度の変動係 数が 20%を上回る場合があった。 比較例 B、 Eでは、 再現性良く測定でき ているものの、 高濃度域における測定平均値はアルブミン濃度レベルと乖離 しており、 正確性が低かった。

産業上の利用可能性 [0035] 本発明によれば、 測定対象リガンドを低濃度域から高濃度域まで高感度か つ広範囲に測定できると同時に、 良好な測定再現性能を有する凝集阻害測定 法及び凝集阻害測定用試薬を提供することができる。 特に、 免疫反応を利用 する場合、 地帯現象を発生しないという特徴から測定値の信頼性が高く、 ま た、 異常高値を呈する可能性のあるリガンドを測定する場合には粒子凝集法 よりも正確な測定方法を提供することができる。

請求の範囲
[1 ] 二箇所以上のレセプター結合部位を有する測定対象リガンドを含む試料と、 下記 (A ) 〜 (C )
( A ) 予めリガンド又はリガンド様物質を担持した不溶性担体粒子
( B ) 前記リガンドに特異的に反応する遊離状レセプター
( C ) 前記リガンドに特異的に反応し、 リガンド上の結合部位が前記遊離 状レセプターとは異なるレセプターを担持した不溶性担体粒子
とを混合し、 試料中の測定対象リガンド量に応じた粒子の凝集阻止反応を測 定することを特徴とする凝集阻害測定法。

[6] レセプターを担持する不溶性担体粒子の平均粒子径が、 リガンド又はリガン ド様物質を担持する不溶性担体粒子の平均粒子径の 1 / 2〜 1 / 1 0である 請求項 1〜 5のいずれかに記載の凝集阻害測定法。
[7] 下記 (A ) 〜 (C ) を含み、 試料中の測定対象リガンド量に応じた粒子の凝 集阻止反応を測定することを特徴とする凝集阻害測定用試薬。
