Abstract

A system and method for analyzing one or more particles in a flow cell comprises focusing the one or more particles in a fluid stream with an acoustic standing wave to acoustically focus the one or more particles in a flow cell. The fluid is transited within the flow cell with a linear flow velocity ranging between about 0 meters/second and about 10 meters/second in the forward or reverse direction.

Claims

AMENDED CLAIMS received by the International Bureau on 23 September 2008 (23.09.08)CLAIMSWhat is claimed is:
1. A method for acoustically reorienting a fluid in a channel comprising; introducing into a channel a first fluid wherein the first fluid has a first acoustic contrast relative to a second fluid; introducing into the channel the second fluid wherein the second fluid has a second acoustic contrast that is different from the first acoustic contrast; applying acoustic radiation pressure to the channel; acoustically reorienting the second fluid based upon the acoustic contrast of the second fluid.
2. The method of claim 1 further comprising moving the first fluid in a laminar flow stream.
3. The method of claim 2 further comprising moving the second fluid in a second laminar flow stream parallel to a flow stream of the first fluid.
4. The method of claim 1 wherein the second fluid comprises particles therein.
5. The method of claim 4 wherein the first fluid and the second fluid are acoustically reoriented relative to the particles.
6. The method of claim 5 further comprising assaying the particles or fluids to produce an assay,
7. An assay produced by the method of claim 6 wherein the second fluid is a biological fluid.
8. The method of claim 1 wherein the first fluid comprises particles therein.
9. The method of claim 1 wherein the first fluid or the second fluid is acoustically reoriented based upon the first acoustic contrast or the second acoustic contrast.
10. The method of claim 1 further comprising flowing the particles in the second fluid to a particle analyzer in fine with the channel.
11. The method of claim 1 wherein the first fluid or the second fluid comprises reagents.
12. A method for acoustically manipulating a particle using radial acoustic focusing comprising: introducing into a channel a first fluid having a first acoustic contrast with a population of particles suspended therein; introducing into the channel a second fluid having a second acoustic contrast that is greater than or equal to the acoustic contrast of the first fluid; applying a radial acoustic radiation pressure to the channel; and acoustically focusing at least a portion of the population of particles from the first fluid to the second fluid.
13. The method of claim 12 wherein the portion of the population of particles are acoustically focused relative to the first acoustic contrast and the second acoustic contrast.
14. The method of claim 12 further comprising moving the first fluid in a first laminar flow stream and moving the second fluid in a second laminar flow stream.
15. The method of claim 12 wherein the step of acoustically focusing comprises concentrating the population of particles into the second stream.
16. The method of claim 12 further comprising analyzing in a particle analyzer the portion of the population of particles in the second fluid.
17. The method of claim 12 further comprising imaging with an imager the portion of the population of particles in the second fluid.
18. The method of claim 12 further comprising non-dilutive sorting of the portion of the population of particles in the second fluid,
19. The method of claim 12 wherein the first fluid comprises a reagent that reacts with at least some of the population of particles.
20. The method of claim 12 wherein the second fluid is located at a center of the channel.
21. The method of claim 12 wherein the population of particles is selected from the group consisting of cells, beads, or any combination thereof.
22. The method of claim 12 further comprising flowing the particle in the first fluid or the second fluid to a particle analyzer that is in line with the channel.
23. The method of claim 22 wherein the particle analyzer is a flow cytometer.
24. The method of claim 12 wherein the population of particles are cultured cells, the first fluid is cell growth medium in which the cells are grown and the second fluid is new cell growth medium.
25. The method of claim 12 wherein the population of particles are microsphere beads.
26. The method of claim 12 wherein a negative contrast particle is within the population of particles.
27. The method of claim 26 further comprising: capturing a particle of interest from the population of particles with the negative contrast particle; and forcing the particle of interest and the negative contrast particle toward a wall of the channel away from a center of the channel.
28. The method of claim 12 wherein the population of particles is blood cells.
29. The method of claim 28 further comprising assaying the particles or fluids to produce an assay.
30. A method for imaging an acoustically manipulated rare target in an acoustic flow cytorneter comprising: introducing a fluid comprising the rare target to a flow cell; applying acoustic radiation pressure to the flow cell; acoustically focusing the rare target within the flow cell to concentrate the rare target; aligning the concentrated rare target in the flow eel); interrogating the aligned rare target at an interrogation site to obtain an optical signal from the rare target to yield population statistical data: and imaging the rare target to produce a high content image representative of the rare target.
31. The method of claim 30 further comprising correlating the population statistical data with the high content image to produce improved data content.
32. The method of claim 30 further comprising slowing the transit time of the rare target during the interrogating step.
