- prediction of a signal peptide is one of effective means.
- the signal peptide exists as an extended peptide having 15 to 30 residues at the N-terminus of the mature protein.
- a hydrophobic amino acid region is present in the signal peptide sequence, leading to attachment of the nascent polypeptide chain to the endoplasmic reticulum membrane and passage through the membrane, and the signal peptide is cleaved after passage through the membrane.
- the signal peptide since the signal peptide is used for intracellular transport and is cleaved after transport, the signal peptide region of GDH is not involved in the activity of GDH itself. Therefore, it is considered that there is a low possibility that the enzyme activity itself is impaired by deletion within a region known to be a signal peptide. Furthermore, when the fungal FAD-GDH used in the present invention is heterologously expressed in an E. coli host, the signal peptide not only has a transport function observed in the original strain but also has a high hydrophobicity in E. coli. It is suggested that the chain is present as it is added, which may cause a decrease in the expression level, such as reducing the stability of GDH.
- the length of the signal peptide is predicted, a mutant GDH gene in which the N-terminus is deleted at a position in the vicinity thereof is prepared, and these are introduced into Escherichia coli. Comparing the status of enzyme expression is an effective means for determining a suitable length of the N-terminal deletion region.

- the N-terminal region containing the region corresponding to the signal peptide present in the N-terminal region of GDH-derived GDH is deleted in E. coli. It is possible to improve the expression level when expressed. Specifically, for example, in GDH having the amino acid sequence set forth in SEQ ID NO: 1 or SEQ ID NO: 3, the DNA encoding the N-terminal region containing the amino acid sequence of MKITAAIITVATAFASFAS present at the N-terminal is deleted and expressed. By doing so, the signal peptide can be deleted.
- the “amino acid sequence corresponding to MKITAAIITVATAFASFASA present at the N-terminus thereof” has a certain homology (for example, 85% or more) with GDH having the amino acid sequence described in SEQ ID NO: 1 or SEQ ID NO: 3. , Preferably 90%, more preferably 95% or more identity).
- Correspondences of “corresponding positions / sequences” between GDHs having a certain amino acid sequence identity above a certain level are obtained by using a ready-made amino acid homology analysis software such as GENETYX-Mac (manufactured by Software Development).
- the deletion region of the N-terminal peptide of GDH in the present invention is not necessarily limited to the signal peptide region.
- a region including a region adjacent to the signal peptide region and not having an adverse effect on the enzyme activity can also be deleted.
- MKITAIITVATAFASFASA present at the N-terminus thereof may be deleted, or MKITAAIITVATAFASFASAQ in which one residue is further deleted.
- the amino acid sequence may be deleted. More residues may be deleted as long as the enzyme activity is not adversely affected.
- MKITAIITVATAFAFASAQQDTNSS present at the N-terminus thereof may be deleted, or the amino acid sequence of MKITAIITVATAFASFASAQQDTNSSS that is deleted by one more residue may be deleted. May be.
- the N-terminal signal peptide region is substantially deleted, and whether or not some regions are deleted in addition to the N-terminal signal peptide region is an essential requirement of the present invention.
- a plurality of N-terminal region deletion variations are included in the GDH of the present invention.
- N-terminal region for example, an N-terminal region shorter by one to several residues
- this deletion Between the effect and the effect when the entire region to be the N-terminal signal peptide region is deleted, or the effect when the entire region to be the N-terminal signal peptide region is deleted and the longer N-terminal region is deleted Such deletions may also be included in variations of the invention if no significant differences are observed.
- the method for deleting the N-terminal peptide is not limited, and examples thereof include a known method, for example, a method in which the amino acid at the N-terminus is changed to methionine as a start codon by signal peptide cleavage and expressed.
- signal peptide-deficient GDH can be expressed in E. coli by adding methionine, which is a start codon, to the N-terminal amino acid by signal peptide cleavage.
- the amino acid at the N-terminal is changed to methionine, which is the start codon when cleavage is performed in the assumed region.
- Whether or not the expression level of the target enzyme has increased as compared with the state in which the signal peptide sequence is present can be confirmed by comparing the total activity value per 1 ml of the culture solution before and after introducing the mutation into the sequence. It should be noted that the deletion of the signal peptide can be confirmed by the N-terminal amino acid sequence using Edman degradation. PSORT and SignalP are often used as signal peptide prediction programs. Each can be used on the web from the address “psort.nibb.ac.jp/” or “www.cbs.dtu.dk/services/SignalP-2.0/”.

- a technique for producing a gene encoding a useful protein by introducing it into a heterologous host is theoretically known. In reality, however, the degree of the effect is not uniform depending on what gene is introduced into which heterogeneous host, and whether the introduction itself does not work even if it is introduced into the heterologous host. There are many cases where the expression of is not successful.
- the flavin-binding GDH derived from various fungi used in the present invention is not sufficient to produce a flavin-binding GDH when the original microorganism is cultured, but cell culture on a larger scale is not possible. Necessary. Abstract
Description

(反応2) PMS(還元型) + DCIP(酸化型)
→ PMS + DCIP(還元型)
この上清を、緩衝液A(10mM 酢酸緩衝液、2M 硫酸アンモニウム、pH5.0)にて予め平衡化したブチルトヨパール650C(東ソー社製)カラム(26φ×28.5cm)にかけ、緩衝液Aから緩衝液B(10mM 酢酸緩衝液、pH5.0)のリニアグラジエントによって溶出させた。

その結果、得られたアミノ酸配列は、LVENFTPPTPAQIE(配列番号5)およびIRNSTDEWANYY(配列番号6)であった。
なお、配列番号7および8に記載の1文字表記において、混合塩基はそれぞれ、h=a+c+t、r=a+g、y=c+t、d=a+g+tを表す。
