Abstract
Die Erfindung betrifft insbesondere Schmelztabletten enthaltend Dapoxetin, bevorzugt Dapoxetinresinat. Ferner betrifft die Erfindung ein Verfahren zur Herstellung von Dapoxetinresinat sowie ein Verfahren zur Herstellung einer Schmelztablette enthaltend Dapoxetinresinat.
Description
Schmelztablette, enthaltend Dapoxetin
Die Erfindung betrifft insbesondere Schmelztabletten enthaltend Dapoxetin, bevorzugt Dapoxetinresinat. Ferner betrifft die Erfindung ein Verfahren zur Herstellung von Dapoxetinresinat sowie ein Verfahren zur Herstellung einer Schmelztablette enthaltend Dapoxetinresinat.
Dapoxetin gehört im weiteren Sinne der Gruppe der Selektiven Serotonin-Reuptake Inhibitoren (SSRI) an, wobei es insbesondere das Transportmolekül von Serotonin hemmt.
Die chemische Bezeichnung von Dapoxetin [INN] ist (S)-(+)-N,N-Dimethyl- l -phenyl-3- ( l -naphthalenyloxy)-propanamin. Die chemische Struktur von Dapoxetin wird in nachstehender Formel ( 1 ) dargestellt:
( 1 } Dapoxetin
Die Synthese von Dapoxetin ist in EP 0 288 188 Bl beschrieben.
EP 1 225 881 B l betrifft die Verwendung von Dapoxetin zur Behandlung oder Handhabung von sexueller Funktionsstörung, insbesondere der Ejaculatio Praecox. Hierfür werden Kapsel- und Tablettenformulierungen mit einem Dapoxetingehalt von etwa 2,5 bis 10 Gew.-% vorgeschlagen. Die Einnahme von Hartgelatinekapseln sowie Tabletten erfolgt in der Regel mit Flüssigkeit, um den Schluckvorgang zu erleichtern.
Diese Form der Einnahme mit Flüssigkeit verlangt allerdings ein bestimmtes Mindestmaß an Organisation, was in Anbetracht des Indikationsbereichs von Dapoxetin und der damit oftmals verbundenen Spontanität vom Patienten als problematisch empfunden werden kann. Ein weiterer Nachteil in Bezug auf die Einnahme von herkömmlichen Kapsel- und Tablettenformulierungen mit Flüssigkeit
ist, dass sich die Einnahme im Liegen äußerst schwierig gestaltet und dass der Patient die Darreichungsform möglicherweise diskret einnehmen will, was ohne Flüssigkeit sehr viel leichter zu bewerkstelligen wäre. Zudem kann sich die Herstellung geeigneter oraler Dapoxetin-Darreichungsformen als problematisch erweisen, da Dapoxetin einen bitteren Geschmack aufweist und eine lokalanästhetische Wirkung zeigt.
Aufgabe der vorliegenden Erfindung war es daher, die geschilderten Nachteile der Formulierungen des Standes der Technik zu überwinden und eine Darreichungsform bereit zu stellen, die eine dem Indikationsbereich angemessene Einnahme ohne Flüssigkeit im Liegen ermöglicht und einen besonders raschen Wirkungseintritt gewährleistet. Ferner war es Aufgabe der Erfindung, orale Darreichungsformen von Dapoxetin (sowie ein Verfahren zu deren Herstellung) bereit zu stellen, die keinen bitteren Geschmack und keine lokalanästhetische Wirkung im Mund erzeugen. Der vorteilhafte Geschmack bzw. die ausbleibende lokalanästhetische Wirkung soll auch mit einer zuckerfreien Formulierung ermöglicht werden.
Schließlich sollen die vorstehend genannten Aufgaben mittels eines technisch einfachen (und somit kostengünstigen) Verfahrens in hoher Raum-Zeit -Ausbeute gelöst werden. Die vorstehenden Aufgaben konnten überraschenderweise durch Bereitstellung von Dapoxetinresinat beziehungsweise von Schmelztabletten enthaltend Dapoxetin gelöst werden. Insbesondere wurde gefunden, dass die Herstellung einer Schmelztablette enthaltend Dapoxetinresinat eine Formulierung bereit stellt, die die vorstehenden Aufgaben unerwartet löst.
Gegenstand der Erfindung ist daher eine Schmelztablette enthaltend Dapoxetin.
Ferner ist Gegenstand der Erfindung Dapoxetinresinat. Des Weiteren ist Gegenstand der Erfindung ein Verfahren zur Herstellung von Dapoxetinresinat, umfassend die Schritte:
(i) Lösen von Dapoxetin (a),
(ii) Dispergieren eines polymeren Adsorptionsmittel (b),
(III) gegebenenfalls Rühren bis ein stabiler pH-Wert erreicht wird,
(iv) Filtrieren der aus (ii) oder (III) erhaltenen Dispersion,
(v) Trocknen des aus (iv) erhaltenen Filterkuchens.
Darüber hinaus ist Gegenstand der Erfindung ein Verfahren zur Herstellung einer Schmelztablette, enthaltend Dapoxetinresinat umfassend die Schritte:
(I) Bereitstellen von Dapoxetinresinat,
(II) Vermischen mit weiteren pharmazeutischen Hilfsstoffen,
(III) gegebenenfalls Granulieren der aus (II) erhaltenen Mischung,
(IV) Verpressen der aus (II) erhaltenen Mischung oder des aus (III) erhaltenen Granulats zu einer Schmelztablette.
Ferner ist Gegenstand der Erfindung Dapoxetinresinat zur Behandlung von sexueller Dysfunktion, wobei Dapoxetinresinat ohne Flüssigkeit eingenommen wird. Schließlich ist Gegenstand der Erfindung die Verwendung eines Kationenaustauscherharzes zur Maskierung einer lokalanästhetischen Wirkung und des bitteren Geschmacks eines Arzneistoffes, bevorzugt eines Arzneistoffes zur Behandlung von sexueller Dysfunktion, im Mund. Die erfindungsgemäßen Gegenstände sind hierbei Alternativlösungen für die vorstehend genannten Aufgaben.
Wie vorstehend erläutert, betrifft die vorliegende Erfindung eine Schmelztablette. Unter dem Begriff "Schmelztablette" wird im Rahmen dieser Erfindung eine Schmelztablette verstanden wie im Europäischen Arzneibuch, 6. Auflage, Grundwerk 2008 definiert. Somit handelt es sich bei einer Schmelztablette um eine orale, in der Mundhöhle zerfallende Tablette oder tablettenförmige orale Darreichungsform. Bei der Schmelztablette handelt es sich um eine nicht überzogene Tablette, das heißt die Tablette ist nicht beschichtet oder unbefilmt. Schmelztabletten werden auch als oral dispersible Tabletten bezeichnet.
Gemäß Ph. Eur. 6. Ausgabe müssen Schmelztabletten innerhalb von 3 Minuten zerfallen. Im Rahmen dieser Erfindung ist es bevorzugt, dass die erfindungsgemäßen Schmelztabletten eine Zerfallszeit von weniger als 150 Sekunden, insbesondere von weniger als 50 Sekunden, im Mund aufweisen.
Die Zerfallszeit wird gemäß Ph. Eur. 6. Auflage Kapitel 2.9.1 Prüfung A bestimmt. Bevorzugt wird als Prüfflüssigkeit Wasser verwendet. Sofern im Rahmen dieser Anmeldung Bezug auf die durchschnittliche Zerfallszeit genommen wird, ist darunter der Mittelwert aus der Zerfallszeitbestimmung von 10 Tabletten zu verstehen.
Die erfindungsgemäßen Schmelztabletten weisen üblicherweise ein Gesamtgewicht von weniger als 1000 mg auf, mehr bevorzugt von weniger als 750 mg, insbesondere von weniger als 500 mg. Üblicherweise weisen die erfindungsgemäßen Schmelztabletten ein Gewicht von mehr als 50 mg, bevorzugt 100 mg oder mehr, insbesondere mehr als 150 mg, auf.
Die erfindungsgemäßen Schmelztabletten sind daher eindeutig von sogenannten "Kautabletten" zu unterscheiden, da diese üblicherweise ein höheres Gewicht (ca. 1 ,5 bis 3 g) und eine längere Zerfallszeit aufweisen.
Die erfindungsgemäßen Schmelztabletten weisen üblicherweise eine Bruchfestigkeit von 25 bis 80 N, bevorzugt von 35 bis 70 N, mehr bevorzugt von 40 bis 65 N, insbesondere von 45 bis 60 N, auf. Die Bruchfestigkeit wird üblicherweise gemäß Ph. Eur. 6. Auflage Kapitel 2.9.8 bestimmt. Sofern im Rahmen dieser Anmeldung auf die mittlere Bruchfestigkeit Bezug genommen wird, ist darunter der Mittelwert aus der Bruchfestigkeitsbestimmung von 10 Tabletten zu verstehen.
Zudem zeigen die resultierenden Tabletten bevorzugt eine Friabilität von kleiner 5 %, besonders bevorzugt von kleiner 3 %, insbesondere kleiner 1 %, auf. Die Friabilität wird gemäß Ph.Eur. 6.0, Abschnitt 2.9.7 bestimmt.
Das Gewichtsverhältnis von Wirkstoff (bezogen auf Dapoxetin in Form der freien Base) zu Hilfsstoffen beträgt in den erfindungsgemäßen Schmelztabletten üblicherweise 4 : 1 bis 1 : 8, bevorzugt von 2 : 1 bis 1 : 6, insbesondere 1 : 1 bis 1 : 5.
Die erfindungsgemäße Schmelztablette enthält als Komponente (a) Dapoxetin.
Im Rahmen dieser Erfindung umfasst der Begriff "Dapoxetin" (S)-(+)-N,N-Dimethyl- l- phenyl-3-(l -naphthalenyloxy)-propanamin gemäß vorstehender Formel (1). Zudem umfasst der Begriff "Dapoxetin" alle pharmazeutisch verträglichen Salze sowie Hydrate und Solvate davon.
Bei den Salzen kann es sich um Säureadditionssalze handeln. Beispiele für geeignete Salze sind Hydrochloride, Carbonate, Hydrogencarbonate, Acetate, Lactate, Butyrate, Propionate, Sulfate, Methansulfonate, Citrate, Tartrate, Nitrate, Sulfonate, Oxalate und/oder Succinate. Bevorzugt wird Dapoxetin (a) im Rahmen dieser Erfindung in Form des Hydrochlorids verwendet.
Üblicherweise wird das Dapoxetin (a) in der erfindungsgemäßen Schmelztablette in einer Menge (a) von 1 bis 25 Gew.-%, bevorzugt von 2 bis 20 Gew.-%, mehr bevorzugt von 5 bis 15 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, verwendet.
Sofern nichts anderes angegeben beziehen sich alle Gewichtsangaben der Komponente (a) auf Dapoxetin in Form der (wasserfreien) freien Base.
In einer bevorzugten Ausführungsform enthält die erfindungsgemäße Schmelztablette ein polymeres Adsorptionsmittel als Komponente (b). Im Allgemeinen sind darunter ein oder mehrere polymerere Stoffe (d.h. Stoffe mit mehr als zwei sich wiederholenden Monomereinheiten) zu verstehen, die in der Lage sind, die Komponente (a) zu adsorbieren. Bevorzugt handelt es sich bei dem polymeren Adsorptionsmittel um ein hydrophiles Polymer.
Darunter sind Polymere zu verstehen, die hydrophile Gruppen aufweisen. Beispiele für geeignete hydrophile Gruppen sind Hydroxy, Amino, Carbonsäure oder Carboxylat (nachfolgend auch als Carboxyl/Carboxylat bezeichnet), Sulfonsäure oder Sulfonat (nachfolgend auch als Sulfonsäure /Sulfonat bezeichnet). Das Polymer (b) kann linear oder bevorzugt quervernetzt sein. Das Polymer (b) weist in letzterem Fall bevorzugt einen Quervernetzungsgrad von 0,01 bis 10 %, insbesondere von 0, 1 bis 5 %, auf. (Quervernetzungsgrad = Anzahl an Kohlenstoffatomen, die an mehr als an eine Kette anknüpfen / Anzahl an Kohlenstoffatomen in der Polymerkette insgesamt).
In einer bevorzugten Ausführungsform handelt es sich bei dem polymeren Adsorptionsmittel (b) um ein Ionenaustauscherharz. Ein Ionenaustauscherharz ist ein Polymer, mit dem gelöste Ionen gegen andere Ionen gleicher Ladungsart, jedoch gegebenenfalls mit unterschiedlicher Ladung, ersetzt werden können.
Mehr bevorzugt wird als Komponente (b) ein Kationenaustauscherharz verwendet. Unter Kationenaustauscherharz ist ein Polymer zu verstehen, das funktionelle Gruppen mit einem abdissoziierbaren Kation enthält. Beispiele für diese funktionellen Gruppen sind Sulfonsäuregruppen/Sulfonatgruppen oder Carboxylgruppen/Car- boxylatgruppen. Somit wird als Komponente (b) bevorzugt ein Polymer verwendet, das Carboxylgruppen / Carboxylatgruppen und / oder Sulfonylgruppen / Sulfonatgruppen enthält. Sofern Carboxylat oder Sulfonatgruppen vorliegen, können z.B. Ammonium-, Alkali- und Erdalkaliionen als Gegenionen dienen, bevorzugt sind Natrium und Kalium, insbesondere Kalium. Natriumstärkeglykolat ist bevorzugt kein polymeres Adsorptionsmittel (b) im Sinne dieser Erfindung.
Alternativ wird als Komponente (b) ein Anionenaustauscherharz verwendet. Unter Anionenaustauscherharz ist ein Polymer zu verstehen, das funktionelle Gruppen mit einem abdissoziierbaren Anion enthält. Beispiele für diese funktionellen Gruppen sind Ammoniumgruppen .
In einer weiteren bevorzugten Ausführungsform handelt es sich bei dem polymeren Adsorptionsmittel (b) um ein Copoljmier, erhältlich durch Copolymerisation von Styrol und D nylbenzol. Derartige Copolymere sind unter der Bezeichnung Polystyrolsulfonat bekannt.
Ein bevorzugtes Polystyrolsulfonat ist gemäß US Pharmacopeia monographiert und kann durch folgende Strukturformel veranschaulicht werden.
In einer besonders bevorzugten Ausführungsform handelt es sich bei dem polymeren Adsorptionsmittel (b) um ein Copolymer, erhältlich durch Copolymerisation von Methacrylsäure und Divinylbenzol. Ein derartiges Copolymer ist unter der Bezeichnung Polacrilin bekannt.
Insbesondere wird im Rahmen dieser Erfindung Polacrilm in Form des Kaliumsalzes (Polacrilin-Kalium, insbesondere gemäß US Pharmacopeia monographiert) verwendet.
Polacrilin-Kalium kann durch folgende Strukturformel veranschaulicht werden.
wobei x und y natürliche Zahlen sind, beispielsweise von 10' bis 1020, bevorzugt von 106 bis 1018. Das Verhältnis von x zu y beträgt üblicherweise 50 : 1 bis 1 : 1 , bevorzugt 20 : 1 bis 2 : 1 , besonders bevorzugt 10 : 1 bis 3 : 1. Üblicherweise wird das polymere Adsorptionsmittel (b) in der erfindungsgemäßen Schmelztablette in einer Menge (b) von 1 bis 60 Gew.-%, bevorzugt von 2 bis 50 Gew.-%, mehr bevorzugt von 5 bis 40 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, verwendet. Die erfindungsgemäßen Schmelztabletten weisen üblicherweise ein Gewichtsverhältnis Dapoxetin (a) zu polymerem Adsorptionsmittel (b) von 1 : 10 bis 10 : 1 , bevorzugt 1 : 5 bis 5 : 1 , besonders bevorzugt 1 : 3 bis 3 : 1, insbesondere 1 : 2 bis 2 : 1 , auf.
Durch die Kombination von Dapoxetin (a) mit polymerem Adsorptionsmittel (b) kann Dapoxetin in Form eines Resinats entstehen. Somit ist ein weiterer Gegenstand der Erfindung Dapoxetinresinat (auch als Komponente (a-b) bezeichnet). Dapoxetinresinat wird im Anschluss an die Ausführungen zur erfindungsgemäßen Schmelztablette näher erläutert. In einer weiteren bevorzugten Ausführungsform enthält die erfindungsgemäße Schmelztablette ein atemerfrischendes Mittel als Komponente (c).
Als ein atemerfrischendes Mittel werden im Allgemeinen ein oder mehrere Stoffe bezeichnet, die im Patienten bei Einnahme üblicherweise einen frischen Atem erzeugen.
Im Rahmen dieser Anmeldung enthält das "atemerfrischende Mittel" bevorzugt Aromastoffe wie in der Richtlinie des Rates 88/ 388 /EWG„Aromastoffe" vom 22. Juni 1988 definiert.
Atemerfrischende Mittel (c) können wie folgt gewonnen werden: i) durch geeignete physikalische Verfahren (einschließlich Destillation und Extraktion mit Lösungsmitteln) oder durch enzymatische beziehungsweise mikrobiologische Verfahren aus Stoffen pflanzlichen oder tierischen Ursprungs, die als solche verwendet oder mittels herkömmlicher Lebensmittelzubereitungsverfahren (einschließlich Trocknen, Rösten und Fermentierung) für den menschlichen Verzehr verarbeitet werden;
ii) durch chemische Synthese oder durch Isolierung mit chemischen Verfahren, wobei ihre chemische Beschaffenheit mit einer Substanz identisch ist, die in einem Stoff pflanzlichen oder tierischen Ursprungs im Sinne von Ziffer i) natürlich vorkommt; iii) durch chemische Synthese, wobei jedoch ihre chemische Beschaffenheit nicht mit einer Substanz identisch ist, die in einem Stoff pflanzlichen oder tierischen Ursprungs im Sinne von Ziffer i) natürlich vorkommt.
Ein atemerfrischendes Mittel kann hierbei aus einem oder bevorzugt aus mehreren chemischen Verbindungen bestehen. Beispielsweise kann Pfefferminzaroma ein Kollektiv aus mehr als 10 chemischen Verbindungen enthalten.
In einer bevorzugten Ausführungsform enthält die erfindungsgemäße Schmelztablette mehrere verschiedene atemerfrischende Mittel (d.h. mehrere verschiedene Aromarichtungen), besonders bevorzugt enthält die erfindungsgemäße Schmelztablette 2, 3 oder 4 verschiedene atemerfrischende Mittel.
Bevorzugte Kombinationen von atemerfrischenden Mitteln sind z.B. Aroma Pfefferminz mit Aroma Menthol, Aroma Pfefferminz mit Aroma Zitrone, Aroma Pfefferminz mit Aroma Menthol und Aroma Zitrone, Aroma Gartenminze mit Aroma Zitrone, Aroma Gartenminze mit Aroma Menthol, Aroma Gartenminze mit Aroma Zitrone und Aroma Menthol, Aroma Grapefruit mit Aroma Pfefferminze, Aroma Grapefruit mit Aroma Menthol und Aroma Pfefferminze, Aroma Grapefruit mit Aroma Gartenminze, Aroma Grapefruit mit Aroma Gartenminze und Aroma Menthol.
Beispiele für kommerziell erhältliche atemerfrischende Mittel sind Xylit (z.B. Xylisorb®, erhältlich von Roquette), sprühgetrocknetes Sorbitol (z.B. Merisorb®, erhältlich von Syral), L-Menthol (z.B. erhältlich von Caelo). Üblicherweise wird das atemerfrischende Mittel (c) in der erfindungsgemäßen Schmelztablette in einer Menge (c) von 0, 1 bis 7 Gew.-%, bevorzugt von 0,5 bis 5 Gew.-%, mehr bevorzugt von 1 bis 3 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, verwendet. In einer weiteren bevorzugten Ausführungsform enthält die erfindungsgemäße Schmelztablette ein Süßungsmittel als Komponente (d).
Als Süßungsmittel werden im Allgemeinen ein oder mehrere Stoffe bezeichnet, die im Patienten bei Einnahme üblicherweise einen süßen Geschmackseindruck bewirken.
Bevorzugt werden Süßungsmittel verwendet, die eine Süßkraft von 0,2 bis 13.000, bevorzugt von > 1 bis 4.000, insbesondere von 10 bis 3.000 haben, bezogen auf die Süßkraft des Rohrzuckers (= 1 ,0). Beispiele sind Milchzucker (0,27-0,3), Glycerin (0,5-0,8), D-Glucose (0,5-0,6), Maltose (0,6), Galactose (0,6), Invertzucker (0,8-0,9), Rohrzucker ( 1 ,0), Xylit ( 1 ,0), D-Fructose ( 1 ,0- 1 ,5), Natriumcyclamat (30), D-Tryptophan (35), Chloroform (40), Glycyrrhizin (50), Acesulfam ( 130), Aspartam ( 180-200), Dulcin (200), Suosan® (350), Saccharin (Natriumsalz) (400-500), Saccharin (Ammoniumsalz) (600), l -Brom-5-nitroanilin (700), Naringin Dihydrochalcon (1000- 1500), Thaumatin, Monellin (Peptide) (3000), P-4000, n-Propoxy-2-amino-4-nitrobenzol (4000), Alitam (3000) und/oder Neotam ( 13.000). Der Zahlenwert in Klammern gibt die Süßkraft bezogen auf Rohzucker an. Ebenfalls können Thaumatin und /oder Neohesperidin DC verwendet werden. In der erfindungsgemäßen Schmelztablette wird die Komponente (d) üblicherweise in einer Menge von 0,01 bis 10 Gew.-% , mehr bevorzugt von 0, 1 bis 5 Gew.-%, insbesondere von 0,5 bis 2,5 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, verwendet. In einer bevorzugten Ausführungsform enthält die erfindungsgemäße Schmelztablette mehrere verschiedene Süßungsmittel, besonders bevorzugt enthält die erfindungsgemäße Tablette 2, 3 oder 4 verschiedene Süßungsmittel.
In einer mehr bevorzugten Ausführungsform enthält die Komponenten (d)
(d- 1 ) ein Süßungsmittel mit sofortiger Süße, und
(d-2) ein Süßungsmittel mit verzögerter Süße.
Die Komponenten (d- 1 ) und (d-2) können üblicherweise im Gewichtsverhältnis von 5: 1 bis 1 :5, bevorzugt von 3: 1 bis 1 :3, eingesetzt werden.
Beispiele für ein Süßungsmittel mit sofortiger Süße (d- 1 ) sind Saccharin-Natriumsalz, Saccharin-Ammoniumsalz, Sucralose, Neotam, Alitam, Aspartam, Cyclamat, Thaumatin und /oder Acesulfam. Beispiele für Süßungsmittel mit verzögerter Süße (d-2) sind Glycyrrhizin oder Derivate davon, insbesondere Glycyrrhizin in Form des Mono-Ammoniumsalzes, Thaumatin und Neohesperidin DC.
In einer weiteren möglichen Ausführungsform enthält die erfindungsgemäße Schmelztablette einen Schleimstoff als Komponente (e).
Unter einem Schleimstoff werden im Rahmen dieser Erfindung ein oder mehrere Stoffe verstanden, die durch Erhöhung der Viskosität oder durch Einhüllung den Kontakt von Arzneistoffen mit den Zungenpapillen verringern. Dadurch wird üblicherweise eine Verringerung der Intensität der Geschmacksempfindung erzielt.
In einer möglichen Ausführungsform werden als Komponente (e) Stoffe gewählt, die als 2 Gew.-%ige Lösung oder Mischung in destilliertem Wasser zu einer Viskosität von mehr als 2 mPas, bevorzugt von mehr als 4 mPas, insbesondere von mehr als 6 mPas führen, gemessen bei 25 °C, gemäß Ph. Eur. 6. Auflage Kapitel 2.2.10. Die Viskosität der resultierenden Lösung oder Mischung kann bis zu 50.000, bevorzugt bis zu 10.000, insbesondere bis zu 1.000 mPas betragen.
Als Schleimstoffe (e) können bevorzugt natürliche Gummis, Cellulosenderivate, Alginate und /oder nichtionische Hydrokolloide verwendet werden.
Beispiele für Schleimstoffe (e) sind Agar, Alginsäure, Alginat, Chicle, Carrageenan, Dammar, Eibisch Extrakte, Gellan (E 418), Guarkernmehl (E 412), Gummi Arabicum (E 414), Gummi aus Wegerichkernspelze, Gummi aus Fichtensaft, Johannisbrotkernmehl (E 410), Karaya (E 416), Konjakmehl (E 425) aus der Konjakwurzel gewonnen, Tarakernmehl (E 417), Traganth (E 413), Xanthan Gummi (E 415), bevorzugt hergestellt durch bakterielle Fermentation, Guar Gummi und/oder Lecithin.
Als Schleimstoffe auf Basis von Cellulosederivate werden z.B. Carboxymethylcellulose, Hydroxyethylcellulose und /oder Methylcellulose verwendet.
In der erfindungsgemäßen Schmelztablette wird die Komponente (e) üblicherweise in einer Menge von 0 bis 25 Gew.-%, mehr bevorzugt von 0, 1 bis 20 Gew.-%, insbesondere von 0,5 bis 20 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, verwendet.
Darüber hinaus kann die erfindungsgemäße Schmelztablette noch weitere, übliche pharmazeutische Hilfsstoffe enthalten.
Beispiele hierfür sind Füllstoffe (f), Gleitmittel (g), Sprengmittel (h), und Säuremittel (]).
Unter Füllstoffe (f) sind im Allgemeinen Stoffe zu verstehen, die zur Bildung des Tablettenkörpers bei Tabletten mit geringen Wirkstoffmengen dienen. Das heißt, Füllstoffe erzeugen durch "Strecken" der Wirkstoffe eine ausreichende Tablettiermasse. Füllstoffe dienen üblicherweise also dazu, eine geeignete Tablettengröße zu erhalten.
Beispiele für bevorzugte Füllstoffe sind Lactose, Stärke, Stärkederivate, Calciumphosphat, Saccharose, Calciumcarbonat, Calciumsilikat,
Magnesiumcarbonat, Magnesiumoxid, Maltodextrin, Calciumsulfat, Dextrate, Dextrin, Dextrose, hydrogeniertes Pflanzenöl, Kaolin und/oder bevorzugt Zuckeralkohole. Beispiele für geeignete Zuckeralkohole sind Mannitol, Sorbitol, Xylitol, Isomalt, Glucose, Fructose, Maltose und Gemische daraus. Besonders bevorzugt wird Lactose als Füllstoff verwendet.
Die erfindungsgemäßen Schmelztabletten enthalten Füllstoffe üblicherweise in einer Menge von 0 bis 70 Gew.-%, mehr bevorzugt von 1 bis 60 Gew.-%, insbesondere von 2 bis 55 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette.
Unter Gleitmittel (g) sind im Allgemeinen Stoffe zu verstehen, die zur Verbesserung der Pulverfließfähigkeit (= Fließreguliermittel (gi)) und/oder als Schmiermittel (g2) geeignet sind.
Fließreguliermittel (g,) haben die Aufgabe, in einem Tablettiergemisch sowohl die interpartikuläre Reibung (Kohäsion) zwischen den einzelnen Partikeln als auch das Haften dieser an den Wandflächen der Pressform (Adhäsion) zu vermindern. Ein Beispiel für einen Zusatz zur Verbesserung der Pulverfließfähigkeit ist disperses Siliciumdioxid. Bevorzugt wird Siliciumdioxid mit einer spezifischen Oberfläche von 50 bis 400 m2/g, bestimmt nach Gasadsorption gemäß Ph. Eur., 6. Auflage 2.9.26., verwendet. In diesem Zusammenhang wurde unerwartet gefunden, dass durch die Verwendung von Siliciumdioxid mit einer spezifischen Oberfläche von 50 bis 400 m2/g die Menge an Füllstoff vorteilhaft reduziert werden kann.
Ferner können als Komponente (g2) Schmiermittel verwendet werden. Schmiermittel dienen im Allgemeinen zur Verringerung der Gleitreibung. Insbesondere soll die Gleitreibung vermindert werden, die beim Tablettieren einerseits zwischen den sich in der Matrizenbohrung auf und ab bewegenden Stempeln und der Matrizenwand sowie andererseits zwischen Tablettensteg und Matrizenwand besteht. Geeignete Schmiermittel stellen z.B. Stearinsäure, Adipinsäure, Natriumstearylfumarat (z.B. Pruv®), Magnesiumstearat und/oder Calciums tearat dar. In der erfindungsgemäßen Schmelztablette wird die Komponente (g) üblicherweise in einer Menge von 0 bis 10 Gew.-%, mehr bevorzugt von 0, 1 bis 5 Gew.-%, insbesondere von 1 ,0 bis 3 Gew.-%, bezogen auf das Gesamtgewicht der Tablette, verwendet.
Als Sprengmittel (h) werden im Allgemeinen Stoffe bezeichnet, die den Zerfall einer Darreichungsform, insbesondere einer Tablette, nach Einbringen in Wasser beschleunigen. Geeignete Sprengmittel sind z.B. organische Sprengmittel wie
mikrokristalline Cellulose, Stärke, vorverkleisterte Stärke, Natriumcarboxy- methylstärke, Natriumcarboxymethylcellulose, quervernetzte Polyvinylpyrrolidone, Natriumstärkeglycolat, und Crospovidon. Bevorzugt werden quervernetzte Polyvinylpyrrolidone und/oder Natriumstärkeglycolat als Sprengmittel verwendet.
Die erfindungsgemäßen Schmelztabletten enthalten Sprengmittel (h) üblicherweise in einer Menge von 0 bis 35 Gew.-%, mehr bevorzugt von 1 bis 30 Gew.-%, insbesondere von 2 bis 25 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette. Säuremittel (j) dienen zur Erzielung einer sauren Geschmackskomponente. Beispiele für Säuremittel sind Citronensäure, Weinsäure und Salze davon.
Die erfindungsgemäßen Schmelztabletten enthalten Säuremittel (j) üblicherweise in einer Menge von 0 bis 15 Gew.-%, mehr bevorzugt von 1 bis 10 Gew.-%, insbesondere von 2 bis 5 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette.
Die Erfindung umfasst zudem beliebige Kombinationen der vorstehend genannten bevorzugten Ausführungsformen, d.h. bevorzugt ist eine Schmelztablette, enthaltend
(a) 1 bis 25 Gew.-%, bevorzugt 2 bis 20 Gew.-%, mehr bevorzugt 5 bis 15 Gew.-% Dapoxetin,
(b) 1 bis 60 Gew.-%, bevorzugt 2 bis 50 Gew.-%, mehr bevorzugt 5 bis 40 Gew.-% polymeres Adsorptionsmittel,
(c) 0 bis 7 Gew.-%, bevorzugt 0, 1 bis 5 Gew.-%, mehr bevorzugt von 1 bis 3 Gew.-% atemerfrischende Mittel,
(d) 0 bis 10 Gew.-%, mehr bevorzugt von 0,01 bis 5 Gew.-%, insbesondere von 0, 1 bis 2,5 Gew.-% Süßungsmittel,
(e) 0 bis 25 Gew.-%, mehr bevorzugt von 0, 1 bis 20 Gew.-%, insbesondere von 0,5 bis 20 Gew.-% Schleimstoffe,
(f) 0 bis 70 Gew.-%, mehr bevorzugt 1 bis 60 Gew.-%, insbesondere 2 bis 55 Gew.-% Füllstoffe,
(g) 0 bis 10 Gew.-%, mehr bevorzugt 0,1 bis 5 Gew.-%, insbesondere 1 ,0 bis 3 Gew.-% Gleitmittel,
(h) 0 bis 35 Gew.-%, mehr bevorzugt 1 bis 30 Gew.-%, insbesondere 2 bis 25 Gew.-% Sprengmittel, und
(j) 0 bis 15 Gew.-%, mehr bevorzugt 1 bis 10 Gew.-%, insbesondere 2 bis 5 Gew.-% Säuremittel,
bezogen jeweils auf das Gesamtgewicht der Schmelztablette.
Es liegt in der Natur von pharmazeutischen Hilfsstoffen, dass diese teilweise mehrere Funktionen in einer pharmazeutischen Formulierung wahrnehmen. Im Rahmen dieser Erfindung gilt zur unzweideutigen Abgrenzung daher bevorzugt die Fiktion, dass ein
Stoff, der als ein bestimmter Hilfsstoff verwendet wird, nicht zeitgleich auch als weiterer pharmazeutischer Hilfsstoff eingesetzt wird. Beispielsweise wird Polacrilin - sofern als polymeres Adsorptionsmittel (b) eingesetzt - nicht auch zusätzlich als Sprengmittel eingesetzt (obwohl Polacrilin auch eine gewisse Sprengwirkung zeigt).
Die erfindungsgemäße Schmelztablette kann also die Bestandteile (a) bis (h) und (J) enthalten. Wie vorstehend erläutert, kann durch die Kombination von Dapoxetin (a) mit polymerem Adsorptionsmittel (b) Dapoxetin erfindungsgemäß in Form eines Resinats entstehen. Bei dem erfindungsgemäßen Dapoxetinresinat (a-b) handelt es sich üblicherweise um eine Kombination von Dapoxetin und einem polymeren Adsorptionsmittel (b). Hierbei wird auf alle vorstehenden erläuterten bevorzugten Ausführungsformen der Komponente (b) Bezug genommen. Der genaue Mechanismus dieser Kombination von Dapoxetin und dem polymeren Adsorptionsmittel ist bisher nicht bekannt. Es wird jedoch, ohne an eine Theorie gebunden zu sein, angenommen, dass diese Kombination üblicherweise zumindest teilweise in Form eines Salzes vorliegt.
Im Fall von Dapoxetinresinat werden üblicherweise Art und Menge von Dapoxetin (a) sowie von polymerem Adsorptionsmittel (b) so gewählt, dass die resultierende Kombination (d.h. das resultierende Dapoxetinresinat) im Wesentlichen keine lokalanästhetische Wirkung mehr zeigt. Zudem werden bevorzugt Art und Menge von Dapoxetin (a) sowie von polymerem Adsorptionsmittel (b) so gewählt, dass die resultierende Kombination im Wesentlichen keinen bitteren Geschmack mehr aufweist. Somit fallen Kombinationen von z.B. Dapoxetin mit Stärke, mikrokristalliner Cellulose, Lactose und /oder Polyvinylpyrrolidon nicht unter den Ausdruck "Dapoxetinresinat" im Sinne der vorliegenden Erfindung.
Der Wassergehalt von Dapoxetinresinat beträgt üblicherweise 1 bis 12 Gew.-%, bevorzugt 2 bis 10 Gew.-%, besonders bevorzugt 3 bis 7 Gew.-%, bezogen auf das Dapoxetinresinat- Gesamtgewicht. Dabei wird der Wassergehalt mittels coulometrischer Karl-Fischer Messung mit einem Karl-Fischer Titrator AQUA 40.00 (Analytik Jena AG, Deutschland) ermittelt, wobei eine Probe von 20 mg vermessen wird. Gegenstand der Erfindung ist nicht nur das erfindungsgemäße Dapoxetinresinat sondern auch ein Verfahren zu dessen Herstellung.
Gegenstand der Erfindung ist daher ein Verfahren zur Herstellung von Dapoxetinresinat, umfassend die Schritte:
(i) Lösen von Dapoxetin (a),
(ii) Dispergieren eines polymeren Adsorptionsmittel (b),
(iii) gegebenenfalls Rühren bis ein stabiler pH-Wert erreicht wird,
(iv) Filtrieren der aus Schritt (ii) oder Schritt (iii) erhaltenen Dispersion,
(v) Trocknen des aus Schritt (iv) erhaltenen Filterkuchens.
Im Schritt (i) erfolgt das Lösen von Dapoxetin (a) in einem Lösungsmittel. Die im Schritt (i) verwendeten erfindungsgemäßen Lösungsmittel sind im Allgemeinen polare organische Lösungsmittel. In einer bevorzugten Ausführungsform wird ein organisches Lösungsmittel verwendet, mit einer relativen Permittivität (=εΓ ) von größer 15 bis 85, bevorzugt 20 bis 80, bei 20 °C gemessen.
Als Lösungsmittel eignen sich z.B. Wasser, Alkohol (z.B. Methanol, Ethanol, Isopropanol), Dimethylsulfoxid (DMSO), Aceton, Butanol, Ethylacetat, Heptan, Pentanol oder Gemische daraus. Bevorzugt wird Wasser verwendet.
Als "Lösen" wird im Allgemeinen zumindest ein teilweises Lösen, bevorzugt das Lösen eines überwiegenden Teils von Dapoxetin verstanden, üblicherweise größer 80 Gew.-%, bevorzugt größer 90 Gew.-%, besonders bevorzugt größer 95 Gew.-%, Im Schritt (ii) wird das polymere Adsorptionsmittel (b) in der im Schritt (i) erhaltenen Lösung dispergiert, bevorzugt suspendiert. Gegebenenfalls können sich auch Teile des polymeren Adsorptionsmittel (b) lösen. Üblicherweise liegen größer 10 Gew.-%, bevorzugt größer 80 Gew.-%, besonders bevorzugt größer 95 Gew.-% des polymeren Adsorptionsmittel (b) in nicht gelöster (= dispergierter) Form vor.
Bei beziehungsweise nach dem Zusammenbringen von Dapoxetin und Adsorptionsmittel kann sich der pH-Wert der resultierenden Zusammensetzung ändern. Gegebenenfalls wird die in Schritt (ii) erhaltene Dispersion im optionalen Schritt (iii) so lange gerührt, bis ein stabiler pH-Wert erreicht wird. Ein stabiler pH- Wert ist üblicherweise ein pH-Wert, der sich nicht oder nur minimal ändert. Minimal bedeutet dabei eine Änderung von ± 0,2 über einen Zeitraum von einer Minute.
Im Schritt (iv) erfolgt das Filtrieren der aus Schritt (ii) oder Schritt (iii) erhaltenen Dispersion unter Gewinnung eines Filterkuchens.
Im Rahmen dieser Anmeldung wird unter Filtrieren das Abtrennen von festen Partikeln aus Flüssigkeiten mit geeigneten Filtern verstanden.
In Schritt (v) erfolgt das Trocknen des aus Schritt (iv) erhaltenen Filterkuchens.
In dieser Anmeldung wird unter Trocknen die Abtrennung von an Feststoffen anhaftenden Flüssigkeiten verstanden. Bei den anhaftenden Flüssigkeiten handelt es sich bevorzugt um Wasser in Form von Haftwasser, Kapillar-, Hydratations-, Adsorptions-, Hydrat- und Konstitutionswasser. Trocknen erfolgt im Allgemeinen in üblichen Trocknungsvorrichtungen, beispielsweise Schrank- oder Hordentrockner, Vakuumtrockner, Wirbelschichttrockner, Sprühtrockner oder Gefriertrockner. Erfindungsgemäß werden bevorzugt Schrank- oder Hordentrockner verwendet.
Unter Filterkuchen sind dabei im Allgemeinen die auf dem Filter zurückbleibenden Komponenten zu verstehen.
Die Trocknungszeit beträgt üblicherweise 1 bis 20 Stunden, bevorzugt 2 bis 17 Stunden, mehr bevorzugt 5 bis 15 Stunden. Die Trocknungstemperatur beträgt üblicherweise 40 bis 120 °C, bevorzugt 50 bis 100 °C, insbesondere 60 bis 90 °C. Besonders bevorzugt werden die Trocknungsbedingungen so gewählt, dass das resultierende Dapoxetinresinat den vorstehend erläuterten Wassergehalt aufweist.
Grundsätzlich finden alle vorstehend gemachten Erläuterungen zu bevorzugten Ausführungsformen von Dapoxetin (a) und polymerem Adsorptionsmittel sowie alle Kombinationen daraus auch im erfindungsgemäßen Verfahren Anwendung. So wird in einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens in Schritt (i) ein pharmazeutisch verträgliches Salz von Dapoxetin (a), bevorzugt Dapoxetin-HCl in einem Lösungsmittel, bevorzugt in Wasser, gelöst. In Schritt (ii) wird ein polymeres Adsorptionsmittel, bevorzugt ein Kationenaustauscherharz unter Rühren in der im Schritt (i) erhaltenen Lösung dispergiert. Analog können die vorstehend gemachten Mengenangaben auf das erfindungsgemäße Verfahren übertragen werden.
In einer noch mehr bevorzugten Ausführungsform werden im Schritt (i) Dapoxetin-HCl als Komponente (a) und in Schritt (ii) Polacrilin-Kalium als Komponente (b) verwendet. Bevorzugt wird in Schritt (iii) bis zu einem pH-Wert von mindestens 7,0 gerührt. Anschließend wird die Mischung über einen Filter gegeben und der Filterkuchen, z.B. bei einer Temperatur von 60 bis 90 °C getrocknet.
Das durch das erfindungsgemäße Verfahren erhältliche Dapoxetinresinat ist ebenfalls Gegenstand dieser Erfindung. Es wird bevorzugt zur Herstellung der vorstehend beschriebenen Schmelztablette verwendet. Gegenstand der Erfindung ist somit auch ein Verfahren zur Herstellung einer Schmelztablette, enthaltend Dapoxetin, insbesondere Dapoxetinresinat, umfassend die Schritte: (I) Bereitstellen von Dapoxetin, bevorzugt Dapoxetinresinat,
(II) Vermischen mit weiteren pharmazeutischen Hilfsstoffen,
(III) gegebenenfalls Granulieren der aus Schritt (II) erhaltenen Mischung,
(IV) Verpressen der aus (II) erhaltenen Mischung oder des aus (III) erhaltenen Granulats zu einer Schmelztablette.
Grundsätzlich finden für das erfindungsgemäße Verfahren alle vorstehenden Erläuterungen zu bevorzugten Ausführungsformen der erfindungsgemäßen Schmelztablette Anwendung, beispielsweise betreffend Art und Menge der Komponenten (a) und (b) und der möglichen Zugabe von weiteren pharmazeutischen Hilfsstoffen wie atemerfrischendes Mittel, Süßungsmittel, Schleimstoff.
In Schritt (II) des erfindungsgemäßen Verfahrens wird Dapoxetin, bevorzugt Dapoxetinresinat, mit weiteren pharmazeutischen Hilfsstoffen vermischt. Die Vermischung kann in üblichen Mischern erfolgen. Beispielsweise kann die Vermischung in Zwangsmischern oder Freifallmischern erfolgen, z.B. mittels Turbula® T 10B (Bachofen AG, Schweiz).
Bei Vermischung in Zwangsmischern werden üblicherweise Zeiten von 3 bis 15 Minuten benötigt, um eine homogene Mischung zu erzielen. Bei Verwendung von Freifallmischern z.B. mittels Turbula® T 10B (Bachofen AG, Schweiz) oder Containermischern z.B. mittels CM 500 (J. Engelsmann AG, Deutschland) oder Fassmischern z.B. Typ Rhönradmischer (J. Engelsmann AG, Deutschland) werden üblicherweise Zeiten von 10 bis 20 Minuten benötigt, um die homogene Mischung zu erzeugen.
Optional wird die im Schritt (II) erhaltene Mischung des erfindungsgemäßen Verfahrens im Schritt (III) granuliert. Die Granulierung kann mit im Stand der Technik bekannten Verfahren erfolgen. Die Granulation erfolgt bevorzugt als Trockengranulation. Alternativ kann auch eine Feuchtgranulation erfolgen.
In einer bevorzugten Ausführungsform erfolgt eine Schmelzgranulation. Dabei kann das in Schritt (I) bereit gestellte Dapoxetin, bevorzugt Dapoxetinresinat, in eine Schmelze eingebettet werden. Üblicherweise wird die Schmelze aus einem Matrix formenden Polymer (= Matrixformer) gebildet. Bei dem "Matrixformer" handelt es sich im Rahmen dieser Erfindung im Allgemeinen um einen pharmazeutischen Hilfsstoff, welcher beim Erwärmen über den Schmelzpunkt, insbesondere bei einem Schmelzgranulations- oder Schmelzextrusionsprozess, verformbar und in der Lage ist, das erfindungsgemäße Dapoxetinresinat einzubetten, d.h. eine Matrix für partikuläres Dapoxetinresinat zu bilden. Der Matrixformer weist somit bevorzugt ein thermoplastisches Verhalten auf, d.h. es handelt sich um einen thermoplastischen Matrixformer.
Bevorzugt verwendet wird als Matrixformer Polyvinylpyrrolidon, bevorzugt mit einem gewichtsmittleren Molekulargewicht von 10.000 bis 40.000 g/mol, Copolymer aus Vinylpyrrolidon und Vinylacetat, Insbesondere mit einem gewichtsmittleren Molekulargewicht von 40.000 bis 75.000 g/mol, Polyethylenglykol, insbesondere mit einem gewichtsmittleren Molekulargewicht von 2.000 bis 20.000 g/mol, Polyoxy- ethylenalkylether und/oder Polyvlnylalkohol, bevorzugt mit einem gewichtsmittleren Molekulargewicht von 1.000 bis 30.000 g/mol. Als Matrixformer alternativ verwendet werden Co- Blockpolymere aus Polyethylenglykol und Polypropylenglykol, d.h. Polyoxyethylen Polyoxypropylen-Blockpolymerisate. Bevorzugt weisen diese ein gewichtsmittleres Molekulargewicht von 1.000 bis 20.000 g/mol, mehr bevorzugt von 1.500 bis 12.500 g/mol, insbesondere 5.000 bis 10.000 g/mol, auf. Diese Blockpolymerisate sind bevorzugt erhältlich durch Kondensation von Propylenoxld mit Propylenglykol und nachfolgender Kondensation des entstandenen Polymers mit Ethylenoxid. Das heißt, bevorzugt liegt der Ethylenoxldanteil als "Endblock" vor. Bevorzugt weisen die Blockpolymerisate ein Gewichtsverhältnis von Propylenoxld zu Ethylenoxid von 50 : 50 bis 95 : 5, mehr bevorzugt von 70 : 30 bis 90 : 10, auf. Die Blockpolymerisate weisen bevorzugt eine Viskosität bei 25 °C von 200 bis 2000 mPas, mehr bevorzugt von 500 bis 1500 mPas, insbesondere von 800 bis 1200 mPas, auf, gemessen bei 25 °C und gemäß Ph. Eur., 6. Auflage, Kapitel 2.2.10 bestimmt. Das gewichtsmittlere Molekulargewicht wird bevorzugt mittels Gelpermeations- chromatographie bestimmt.
Im Schritt (IV) erfolgt das Verpressen der aus Schritt (II) erhaltenen Mischung oder des aus Schritt (III) erhaltenen Granulats zu einer Schmelztablette, d.h. eine Kompression zu Tabletten. Die Kompression kann mit im Stand der Technik bekannten Tablettiermaschinen erfolgen. Die Kompression erfolgt bevorzugt in Abwesenheit von Lösungsmitteln .
Beispiele für geeignete Tablettiermaschinen sind Exzenterpressen oder Rundlaufpressen. Beispielsweise kann eine Korsch Exzenterpresse EK0 (Korsch AG, Deutschland) verwendet werden. Im Falle von Exzenterpressen und Rundlaufpressen wird üblicherweise eine Presskraft von 3 bis 50 kN, bevorzugt von 7,5 bis 35 kN, angewandt.
In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens wird Dapoxetinresinat mit weiteren pharmazeutischen Hilfsstoffen gemischt, bevorzugt mit der Maßgabe dass kein Gleitmittel (g) vermischt wird. Die Mischung erfolgt bevorzugt für 1 bis 15 Minuten, insbesondere für 3 bis 7 Minuten. Es kann z.B. ein Turbula® T 10B verwendet werden. Das Gemisch wird anschließend bevorzugt über ein Sieb gesiebt, insbesondere mit einer Maschenweite von 500 bis 1000 μιη, z.B. 800 μτη. Bevorzugt wird weitere 1 bis 15 Minuten gemischt. Anschließend wird bevorzugt ein
Gleitmittel (g) über ein Sieb gesiebt, z.B. über ein Sieb der Maschenweite 800 /im, und der Mischung zugegeben. Die nun erhaltene Mischung wird bevorzugt erneut für z.B. 1 bis 5 Minuten gemischt. Die erhaltene Mischung wird anschließend, z.B. unter Verwendung einer Korsch Exzenterpresse EKO, verpresst.
Gegenstand der Erfindung ist ferner Dapoxetinresinat zur Behandlung von sexueller Dysfunktion, wobei eine orale Darreichungsform, enthaltend Dapoxetinresinat, ohne Flüssigkeit eingenommen wird. In dieser Anmeldung versteht man unter „orale Darreichungsformen" alle pharmazeutischen Darreichungsformen die oral eingenommen werden. Beispiele für erfindungsgemäße orale Darreichungsformen sind Tabletten, Kapseln und Granulate. Beispiele für Tabletten sind Schmelztabletten, Lutschtabletten, nicht überzogene Tabletten, überzogene Tabletten, Brausetabletten, magensaftresistente Tabletten, Tabletten mit modifizierter Freisetzung, Tabletten zur Anwendung in der Mundhöhle, Tabletten zur Herstellung einer Suspension. Bevorzugt sind dabei Schmelztabletten.
In dieser Anmeldung wird unter Einnahme der oralen Darreichungsform ohne Flüssigkeit die Einnahme der erfindungsgemäßen oralen Darreichungsform ohne zusätzliche Flüssigkeitsaufnahme wie z.B. die Einnahme der oralen Darreichungsform mit Wasser verstanden.
Unter sexueller Dysfunktion wird im Rahmen dieser Erfindung üblicherweise eine Störung der sexuellen Funktionen oder Reaktionen verstanden, bevorzugt eine Ejaculatio Praecox.
Die Erfinder haben zudem festgestellt, dass sich ein Kationenaustauscherharz vorteilhaft für die Maskierung einer lokalanästhetischen Wirkung eines Arzneistoffes, bevorzugt eines Arzneistoffes zur Behandlung von sexueller Dysfunktion, im Mund eignet.
Im Rahmen dieser Erfindung versteht man unter Maskierung das Abschwächen oder Unkenntlichmachen einer durch ein oder mehrere Substanzen erzeugten Empfindung im Mund des Patienten.
Unter lokalanästhetischer Wirkung im Mund versteht man im Rahmen dieser Erfindung eine betäubende Empfindung im Mund eines Patienten.
Gegenstand der Erfindung ist daher ferner die Verwendung eines Kationenaustauscherharzes zur Maskierung einer lokalanästhetischen Wirkung eines
Arzneistoffes, bevorzugt eines Arzneistoffes zur Behandlung von sexueller Dysfunktion, im Mund.
Die vorliegende Erfindung soll durch nachfolgende Beispiele veranschaulicht werden.
BEISPIELE
Es wurden das erfindungsgemäße Dapoxetinresinat und Schmelztabletten enthaltend Dapoxetinresinat mittels der erfindungsgemäßen Verfahren hergestellt. Die eingesetzten Formulierungen und die physikalischen Eigenschaften der erhaltenen Schmelztabletten ergeben sich aus nachfolgender Tabelle.
Beispiel 1 Es wurde Dapoxetinresinat mittels des erfindungsgemäßen Verfahrens hergestellt.
Dazu wurde 100 ml demineralisiertes Wasser in einem Becherglas mit Rührfisch vorgelegt und 34,02 mg Dapoxetin-HCl dazugegeben. Unter ständigem Rühren wurde anschließend Polacrilin Kalium NF der Mischung zugegeben und der pH-Wert stetig gemessen. Nach 3 h Rühren wurde ein konstanter pH-Wert erreicht. Die Mischung wurde anschließend über ein Filterpapier gegeben und im Trockenschrank bei 80 °C für 14 h getrocknet.
Beispiel 2
Es wurden Schmelztabletten enthaltend Dapoxetinresinat mittels des erfindungsgemäßen Verfahrens hergestellt.
Beispiel 2.1 Direkttablettierung
Dazu wurde das in Beispiel 1 hergestellte Dapoxetinresinat mit Position 2-7 für 5 Minuten im Turbula T 10B gemischt und anschließend zerkleinert. Das Gemisch wurde über ein Sieb der Maschenweite 800 /<m gesiebt und weitere 10 Minuten im Turbula T 10B gemischt. Anschließend wurde Position 8 über ein Sieb der Größe 300 μτη gesiebt und der Mischung zugefügt. Daraufhin wurde für weitere 5 Minuten im Turbula T 10B gemischt. Die erhaltene Mischung wurde unter Verwendung einer Korsch Exzenterpresse EK0 direkt verpresst.
Beispiel 2.2 Tablettierung von Dapoxetinresinat mit vorheriger Schmelzeinbettung
Schmelzeinbettung von Dapoxetinresinat:
30,00 mg PEG 6000 wurde auf dem Wasserbad bei 50 °C aufgeschmolzen. Anschließend wurden 60,00 mg Dapoxetinresinat in die PEG 6000-Schmelze granuliert.

Nach dem Erstarren wurde das entstandene Granulat zerkleinert und über ein Sieb der Maschenweite 800 μτα. gesiebt. Dapoxetingranulat wurde anschließend mit Position 2 bis 7 für 5 Minuten im Turbula T 10B gemischt. Das Gemisch wurde über ein Sieb der Maschenweite 800 μπι gesiebt und weitere 10 Minuten im Turbula T 10B gemischt.
Anschließend wurde Position 8 über ein Sieb der Maschenweite 300 μτη gesiebt und der Mischung zugefügt. Daraufhin wurde erneut für 3 Minuten im Turbula T10B gemischt. Die erhaltene Mischung wurde nun unter Verwendung der Korsch Exzenterpresse EK0 zu Tabletten verpresst.
Beispiel 3: Tabletteneigenschaften
Jeweils 10 der gemäß Beispielen 2.1 und 2.2 hergestellten Schmelztabletten wurden auf folgende Parameter überprüft: Bruchfestigkeit, Zerfallszeit, Geschmack und lokalanästhetische Wirkung. Die in Tabelle 1 aufgezeigten Werte eines jeden Parameters sind als Mittelwert von 10 Tabletten zu verstehen.

Die Prüfung der Bruchfestigkeit wurde dabei gemäß Pharm. Eur. 6.0, Kapitel 2.9.8 ermittelt.
Die Zerfallszeit der Schmelztabletten wurde dabei gemäß Pharm. Eur. 6.0, Kapitel 2.9.1 Prüfung A bei einer Temperatur von 37 °C ermittelt.
Die lokalanästhetische Wirkung wurde mit Hilfe von 10 Versuchspersonen ermittelt. Die Versuchspersonen konnten dabei auf einer Skala von 1 bis 4 ihre Empfindung bezüglich der lokalanästhetischen Wirkung im Mund bewerten. Die einzelnen Stufen 1 bis 4 wurden wie folgt definiert: (1 )„starke lokalanästhetische Wirkung", (2)„mittlere lokalanästhetische Wirkung", (3) „leichte lokalanästhetische Wirkung (4) „keine lokalanästhetische Wirkung". Die Bewertungspunkte der 10 Versuchspersonen wurden addiert und die Summe aller Bewertungspunkte entsprechend klassifiziert: 10 bis 15 Punkte: „deutliche lokalanästhetische Wirkung", 16 bis 30 Punkte: „gemäßigte lokalanästhetische Wirkung", 31 bis 40 Punkte:„keine lokalanästhetische Wirkung".
Da Empfindungen bezüglich des Geschmacks individuell verschieden sein können, wurde zur Geschmacksprüfung der gemäß den Beispielen 2.1 und 2.2 hergestellten Schmelztabletten 10 Versuchspersonen herangezogen und das durchschnittliche Empfinden bezüglich Geschmack ermittelt.
Für die Prüfungen bezüglich Geschmack und lokalanästhetische Wirkung erfolgte die Degustation 1 Stunde nach der letzten Mahlzeit. Die Versuchspersonen waren alle männlich und Nichtraucher. Der Prüfraum war von neutralem Geruch, die Temperatur betrug 20 °C. Die Beurteilung der Reizempfindungen und die Ausbildung der Prüfpersonen erfolgte gemäß DIN 10950.
Claims
1. Schmelztablette, enthaltend Dapoxetin.
2. Schmelztablette gemäß Anspruch 1 , enthaltend
(a) Dapoxetin, und
(b) ein polymeres Adsorptionsmittel, bevorzugt ein Kationenaustauscherharz.
3. Schmelztablette gemäß Anspruch 2, wobei die Komponenten (a) und (b) in Form von Dapoxetinresinat (a-b) vorliegen.
4. Schmelztablette gemäß Ansprüche 1 bis 3, zusätzlich enthaltend ein atemerfrischendes Mittel (c).
5. Schmelztablette gemäß Ansprüche 1 bis 4, zusätzlich enthaltend ein Süßungsmittel (d).
6. Schmelztablette gemäß Ansprüche 1 bis 5, zusätzlich enthaltend einen Schleimstoff (e).
7. Schmelztablette gemäß Ansprüche 2 bis 6, wobei das polymere Adsorptionsmittel (b) Polacrilin-Kalium ist.
8. Schmelztablette gemäß Ansprüche 1 bis 7, wobei das Gewichtsverhältnis Dapoxetin (a) zu polymerem Adsorptionsmittel (b) 1 : 10 bis 10 : 1 ist.
9. Schmelztablette gemäß Ansprüche 1 bis 8, wobei Dapoxetin (a) in einer Menge von 1 bis 25 Gew.-% und das polymere Adsorptionsmittel (b) in einer Menge von 1 bis 60 Gew.-%, bezogen auf das Gesamtgewicht der Schmelztablette, enthalten sind.
10. Verfahren zur Herstellung von Dapoxetinresinat, umfassend die Schritte:
(i) Lösen von Dapoxetin (a),
(ii) Dispergieren eines polymeren Adsorptionsmittel (b),
(iii) gegebenenfalls Rühren bis ein stabiler pH- Wert erreicht wird,
(iv) Filtrieren der aus Schritt (ii) oder Schritt (iii) erhaltenen Dispersion,
(v) Trocknen des aus Schritt (iv) erhaltenen Filterkuchens.
1 1. Verfahren gemäß Anspruch 1 1 , wobei in Schritt (i) Dapoxetin-HCl als Komponente (a) und in Schritt (ii) Polacrilin-Kalium als Komponente (b) verwendet wird, und bevorzugt in Schritt (iii) bis zu einem pH-Wert von mindestens 7,0 gerührt wird.
12. Verfahren zur Herstellung einer Schmelztablette, enthaltend Dapoxetinresinat umfassend die Schritte
(I) Bereitstellen von Dapoxetinresinat,
(II) Vermischen mit weiteren pharmazeutischen Hilfsstoffen,
(III) gegebenenfalls Granulieren der aus Schritt (II) erhaltenen Mischung,
(IV) Verpressen der aus Schritt (II) erhaltenen Mischung oder des aus Schritt (III) erhaltenen Granulats zu einer Schmelztablette.
13. Dapoxetinresinat.
14. Dapoxetinresinat gemäß Anspruch 13, erhältlich durch ein Verfahren gemäß Anspruch 10 oder 1 1.
15. Dapoxetinresinat zur Behandlung von sexueller Dysfunktion, wobei eine orale Darreichungsform enthaltend Dapoxetinresinat ohne Flüssigkeit eingenommen wird.
16. Verwendung eines Kationenaustauscherharzes zur Maskierung einer lokalanästhetlschen Wirkung eines Arzneistoffes, bevorzugt eines Arzneistoffes zur Behandlung von sexueller Dysfunktion, im Mund.
